Tyrosinase, the key gene in melanin pigment synthesis, is tissue-specifically expressed in melanocytic cells. Expression of this gene is regulated by various hormones, carcinogens, and environmental factors. The molecular basis underlying tyrosinase gene regulation is still not clear. In this report, we present the effects of tumor suppressor p53 protein on tyrosinase gene expression and melanin synthesis in human melanoma. After stable transfection of wild type p53 expression plasmid into a highly pigmented melanoma cell line, overexpression of wt p53 suppressed the pigmentation of the melanoma cells. The loss of pigmentation was associated with the loss of endogenous tyrosinase expression at the activity and mRNA levels. In order to determine whether the p53 repression of tyrosinase mRNA involved modulation of tyrosinase promoter activity, transient transfection approaches involving p53 expression plasmid and construct containing chloramphenicol acetyl transferase (CAT) reporter gene linked to 270 bp tissue-specific tyrosinase promoter have been used. p53 specifically repressed CAT gene expression from the tyrosinase promoter and not from the Rous sarcoma virus promoter. These data suggest that in human melanoma p53 down-regulates the tissue-specific expression of tyrosinase gene and subsequent melanin synthesis.  相似文献

In many bird species males provision their mates prior to egg-laying. Courtship feeding has been suggested to function in several ways: to advance laying date by improving female condition, to induce a female to copulate or to allow a female to assess her mate. The role of courtship feeding in Ospreys Pandion haliaetus was investigated in British Columbia, Canada. Courtship feeding rate affected the probability of a pair initiating a clutch. Pairs that laid eggs had higher rates of courtship feeding than pairs that did not lay eggs in both 1991 and 1992. Male courtship feeding rate also correlated negatively with the duration of the courtship period. Experimentally increasing the amount of food available to females prior to egg-laying resulted in a nonsignificant reduction in the duration of the courtship period. This study found no evidence to support the suggestion that female Ospreys trade copulations for food during the courtship period; only 63 of 385 copulations observed were associated with feeds, and courtship feeding rate did not correlate with the copulation rate of a pair. Male provisioning rates, however, were predictable; courtship feeding rate correlated with both male delivery rate to the nest when chicks were 1–2 weeks old and mean brood growth rate. Female Ospreys therefore may be able to predict the quality of subsequent paternal care using courtship feeding rate. As predicted if optimal hatching asynchrony is dependent on food availability, mean brood growth rate, an indirect measure of male parental care, was negatively correlated with hatching asynchrony. This suggests that female Ospreys may manipulate hatching asynchrony in response to male courtship feeding rate, thereby maximizing the productivity of their brood at predicted food levels.  相似文献

The effect of oxamyl on the growth of peas attacked by pea cyst nematode   总被引:1,自引:0,他引:1  
Oxamyl treatment at drilling greatly increased the growth of the plants, the yield and quality of the peas harvested, in soil infested by small or large numbers of Heterodera goettingiana. Treated plants were continuing to set pods when non-treated plants were senescing. Rhizobium nodulation, sap nitrate and tissue nitrogen content were increased in treated plants. Oxamyl applied to growing plants reduced later nematode invasion and enabled root nodulation to proceed with an increase in tissue nitrogen but no increase in yield. Oxamyl applied at drilling in April was inactive against invasion by H. goettingiana in June, which enabled the nematode to multiply equally in treated and non-treated plots. The most economic treatments were at the smallest rates, giving the greatest increase in yield/kg of oxamyl applied.  相似文献

Leaves of 97 taxa representing all the genera at present recognized in the family Oleaceae were surveyed for flavonoids. Four flavonol glycosides were found to be common, the 3-glucmides and 3-rutinosides of quercetin and kaempferol, as were four flavone glycosides, namely the 7-glurosides arid 7-rutinosides of luteolin and apigenin. Among rarer constituents detected were luteolin 4'-glucoside, eriodictyol 7-glucoside, chrysoeriol 7-glucoside, an apigenin-di-C-glycoside and several higher glycosides of quercetin. The species and genera surveyed fell into two groups: those with flavonol glycosides alone; and those with both flavonol and flavone glycosides. The most striking correlation was with chromosome number (and subfamily division) since almost all taxa with a basic number of 11, 13 and 14 had only flavonol glycosides, whereas most taxa with x = 23 had both types of flavonoid. Evolutionary advancement in the family appears to involve the gradual replacement of flavonol by flavone glycosides. Indeed, a few tam, notably Nestegis apelala, Picconia excelsa and Tesserandra fluminense , lacked flavonol glycosides in the leaves completely. At the lower levels of classification, the distribution of flavonoids is of less interest. However, the patterns in Linociera and Chionanthus , two taxa recently made congeneric, are sufficiently different to suggest that this decision might have to be reconsidered when more is known of their chemistry. Otherwise leaf patterns generally fit in with the existing generic classification in the family.  相似文献

Due to inherent difficulties in the construction of tube solarimetersby ‘conventional’ techniques, an alternative methodis presented. Construction is based on the use of printed circuitboards, and further simplified mechanical procedures were employed.Response was linear over the range up to 600 W m–2 totalsolar radiation, at a mean calibration constant of 26 mV kW–1m–2. ABSTRACT

Attraction of flying mole crickets to individual males calling in an outdoor arena was influenced significantly by the relative intensity of the males calling in the arena. Louder males attracted more individuals than males whose calling songs were less intense. Making simple assumptions about the acoustic output from calling males and about the flight pattern and response of flying females, the differential attraction can be explained by a mathematical model. Computer simulation of the model was used to examine the importance of a male's intensity relative to others and the effect of distance between males on the attraction of females. The model and its relation to active female choice and passive attraction are discussed. The model makes predictions about differences in spacing behaviour of males that maximize attraction relative to other males.  相似文献
