The control of phytochrome A expression at the protein and mRNA levels was investigated in wild-type and phyB-1 mutant sorghum ( Sorghum bicolor [L.] Moench). PHYA mRNA abundance follows a diurnal rhythm in both genotypes, with maximal accumulation near the latter part of the light period. PHYA mRNA is more abundant in the phyB-1 mutant. The level of PHYA message correlates with both R : FR and photon flux density in wild-type, but only with photon flux density in the phyB-1 mutant. The differences in mRNA abundance are reflected in the level of phyA protein, which is elevated in the phyB-1 mutant and accumulates under low photon flux density. During de-etiolation, PHYA message accumulation is initially repressed solely by a very low fluence response (VLFR) presumably mediated by phyA. The phyB-mediated low fluence response maintains the repression of accumulation past the time controlled by the VLFR. With repetitive photoperiods, the transition from the etiolated growth form to autotrophic competency is accompanied by a transition from light-induced reduction of PHYA mRNA abundance to enhanced accumulation during the light period. The loss of phyB function allows partial de-repression of PHYA message accumulation under repetitive photoperiods, resulting in plants deficient in phyB but enriched in phyA. The modification of PHYA mRNA and protein levels in the phyB-1 mutant documented in this study may help clarify the molecular basis of the phyB-1 phenotype. The tailoring of phyA abundance in wild-type to the time of day and shade signals suggests a plastic role for this pigment in controlling development in light-grown plants.  相似文献

The complete coding sequence for P-450 PBc2 was inserted into a T7-phage promoter system, and a capped cRNA was generated using T7 RNA polymerase. The P-450 PBc2 cRNA was translated in a rabbit reticulocyte lysate. The in vitro translation product was indirectly immunoprecipitated by the monoclonal antibodies 2F5 and 3C3 that recognize P-450 K. SDS-polyacrylamide gel electrophoresis revealed that the translated protein product exhibits the same relative electrophoretic mobility as P-450 K. The N-terminal amino acid sequence was determined to be MDLVVVLGL-LS-LLLLSL- for P-450 K immunopurified from rabbit kidney using the monoclonal antibody 2F5. This sequence agrees with the predicted amino acid sequence derived from the P-450 PBc2 cDNA. These results indicate that P-450 K or a protein closely related to P-450 K is encoded by the plasmid pP-450 PBc2.  相似文献

Over the last two and half decades, strong evidence showed that the terrestrial ecosystems are acting as a net sink for atmospheric carbon. However the spatial and temporal patterns of variation in the sink are not well known. In this study, we examined latitudinal patterns of interannual variability (IAV) in net ecosystem exchange (NEE) of CO2 based on 163 site-years of eddy covariance data, from 39 northern-hemisphere research sites located at latitudes ranging from ∼29°N to ∼64°N. We computed the standard deviation of annual NEE integrals at individual sites to represent absolute interannual variability (AIAV), and the corresponding coefficient of variation as a measure of relative interannual variability (RIAV). Our results showed decreased trends of annual NEE with increasing latitude for both deciduous broadleaf forests and evergreen needleleaf forests. Gross primary production (GPP) explained a significant proportion of the spatial variation of NEE across evergreen needleleaf forests, whereas, across deciduous broadleaf forests, it is ecosystem respiration (Re). In addition, AIAV in GPP and Re increased significantly with latitude in deciduous broadleaf forests, but AIAV in GPP decreased significantly with latitude in evergreen needleleaf forests. Furthermore, RIAV in NEE, GPP, and Re appeared to increase significantly with latitude in deciduous broadleaf forests, but not in evergreen needleleaf forests. Correlation analyses showed air temperature was the primary environmental factor that determined RIAV of NEE in deciduous broadleaf forest across the North American sites, and none of the chosen climatic factors could explain RIAV of NEE in evergreen needleleaf forests. Mean annual NEE significantly increased with latitude in grasslands. Precipitation was dominant environmental factor for the spatial variation of magnitude and IAV in GPP and Re in grasslands.  相似文献

Both carbon dioxide and ethylene can affect the rate of root elongation. Carbon dioxide can also promote ethylene biosynthesis by enhancing the activity of 1-aminocylopropane-1-carboxylic acid (ACC) oxidase. Since the amount of CO2 in the soil air, and in the atmosphere surrounding roots held in enclosed containers, is known to vary widely, we investigated the effects of varying CO2 concentrations on ethylene production by excised and intact sunflower roots (Helianthus annuus L. cv. Dahlgren 131). Seedlings were germinated in an aeroponic system in which the roots hung freely in a chamber and were misted with nutrient solution. This allowed for treatment, manipulation and harvest of undamaged and minimally disturbed roots. While exposure of excised roots to 0.5% CO2 could produce a small increase in ethylene production (compared to roots in ambient CO2), CO2 concentrations of 2% and above always inhibited ethylene evolution. This inhibition of ethylene production by CO2 was attributed to a reduction in the availability of ACC: however, elevated CO2 had no effect on ACC oxidase activity. ACC levels in excised roots were depressed by CO2 at a concentration of 2% (as compared to ambient CO2), but n-malonyl-ACC (MACC) levels were not affected. Treating intact roots with 2% CO2 inhibited elongation by over 50%. Maximum inhibition of elongation occurred 1 h after the CO2 treatment began, but elongation rates returned to untreated values by 6 h. Supplying these same intact roots with 2% CO2 did not alter ethylene evolution. Thus, in excised sunflower roots 2% CO2 treatment reduces ethylene evolution by lowering the availability of ACC. Intact seedlings respond differently in that 2% CO2 does not affect ethylene production in roots. These intact roots also temporarily exhibit a significantly reduced rate of elongation in response to 2% CO2.  相似文献

The plasma half lives of antipyrine, paracetamol, and lignocaine given by mouth were measured in 23 patients with stable chronic liver diseases of varying severity. Fifteen patients received all three drugs and 19 at least two. The half life of paracetamol was abnormally prolonged in nine out of 17 patients (mean 2-9 hours, normal 2-0 hours), of antipyrine in 10 out of 19 patients (mean 30-4 hours, normal 12-0 hours), and of lignocaine in 19 out of 21 patients (mean 6-6 hours, normal 1-4 hours). Prolongation of the half lives of all three drugs was significantly correlated with an increase of the vitamin-K1-corrected prothrombin time ratio and a reduction in serum albumin concentration. There was no correlation with serum bilirubin concentration or serum alanine aminotransferase activity. This suggests that impaired drug elimination was related to depressed hepatic protein synthesis. Considerable prolongation of the half life of one drug was invariably associated with delayed elimination of the others. The half life of lignocaine, however, was always the most prolonged and was a highly sensitive indicator of hepatic dysfunction. The pharmacokinetic characteristics of a drug as well as the severity of liver disease should be taken into account when considering drug dosage in patients with chronic liver disease.  相似文献

The wetlands of the Alligator Rivers Region of northern Australia have been recognized as having high national and international conservation value. The diversity and productivity of these habitats is, however, under current and future threat from invasive feral animals (Asian water buffalo and pigs) and naturalized alien plants (mimosa, salvinia and para grass) and also from climate change and sea level rise. Some habitats have already been severely degraded and require rehabilitation. In response to this situation feral animal management has centered on eradicating the buffalo population and introducing measures to control pigs. Weed management has focused on control of mimosa and salvinia with an increasing emphasis on integrated control measures. The vulnerability of the freshwater wetlands to climate change and sea level rise is considerable, although the exact nature of environmental change has not been determined.Rehabilitation of the degraded wetlands has centered on controlling the cause of the problem (e.g., the invasive species) and allowing subsequent natural succession to occur. It is recommended that further monitoring and assessment of successional change is undertaken to ascertain if this is sufficient. An integrated monitoring program for detecting the extent and rate of ecological change because of climate change and sea level rise is also proposed. Specific management and research tasks for each of the major broad causes of wetland degradation within the region are made. It is then strongly recommended that all rehabilitation and monitoring activities, including related research, are conducted within a holistic management framework that takes into account the different land jurisdictions within the region and also within the broader landscape context. The utilization of existing management and research structures and processes is stressed as one means of achieving an integrated approach.  相似文献

Regional, national and local wetland classifications have been developed and successfully applied. These have invariably been orientated towards conservation and management goals, and the information used to assess wetland loss or to assign management priorities. Existing national and regional classification systems have not only been useful, but they provide an essential base for developing an international system. At the international level, differences among existing systems in the definition of a wetland and how wetland types are defined assume great importance and need to be resolved. Classification is an essential prerequisite for wetland inventory. A number of international inventories have been undertaken, although these have not generally utilized the available high technology and data storage systems available through remote sensing and geographic information systems. More extensive international inventories will require standardization of techniques for data collection, storage and dissemination. A minimum data set needs to be defined with standards for data accuracy. An international committee under the auspices of an international agency (e.g. IWRB, Ramsar Bureau, IUCN) needs to be established to develop an international classification system and guidelines for carrying out a complete inventory of the world's wetlands.  相似文献

Reconstitution of wild-type apoaspartate aminotransferase from Escherichia coli with [4'-3H]pyridoxamine 5'-phosphate results in stereospecific release of the pro-S C-4' 3H to the solvent. The reaction follows first-order kinetics (t1/2 = 15 min at pH 7.5 and 25 degrees C), its rate constant being similar to that found previously with mitochondrial aspartate aminotransferase from chicken (Tobler, H.P., Christen, P., and Gehring, H. (1986) J. Biol. Chem. 261, 7105-7108). Substituting the active site residue Lys258 by alanine via site-directed mutagenesis yields a catalytically inactive enzyme (Malcolm, B. A., and Kirsch, J. F. (1985) Biochem. Biophys. Res. Commun. 132, 915-921). This mutant enzyme fails to release any measurable 3H from bound [4'-3H]pyridoxamine 5'-phosphate. The data are consistent with earlier proposals that Lys258 is indispensable for the ketimine/aldimine tautomerization, and corroborate the previous conclusion that 3H exchange from enzyme-bound pyridoxamine 5'-phosphate mechanistically corresponds to the deprotonation at C-4' of the ketimine intermediate during the transamination reaction.  相似文献
