The hydration behavior of three naturally occurring sphingomyelins (SM) has been studied. Adsorption isotherms for these SM, singly and in combination with other lipids, were obtained and the isotherms analyzed by the application of BET theory. The results are compared with those found previously by us for the phosphatidylcholines (PC). We find that, depending on the SM studied, both "weak" and "strong" water adsorption can be observed and thus, the presence of choline in the phospholipid head group, does not guarantee "strong" water adsorption. When the head group is removed from SM, the resulting ceramides are found to be very weak, water adsorbers. Cholesterol, when present in a mixture with SM, has a rather dramatic effect on the hydration of the SM. A mixed system of PC and SM exhibits water adsorption characteristics very similar to those exhibited by PC itself. We speculate that these hydration results may well play a role in the cell signaling action of SM and, moreover, may be closely associated with the hypothesized formation of lipid "rafts." 相似文献

Quantitation of the expression of cell surface antigens has hitherto been limited to analysis by either cytotoxicity tests or radioimmune assays (5, 15). We report here the use of a new methodology to analyze and quantitate the expression of mouse histocompabililty antigens (H-2 locus) in hybrid clones and parental cell types. The binding of fluorescein-tagged antibody is measured on a cell-to-cell basis in large viable cell populations using flow microfluorimetric techniques. These techniques have been used to measure hapten and immunoglobulin binding to lymphocyte populations (8, 9, 14). However, this is the first report in which these techniques have been used to examine the expression of the H-2 locus. The advantage of this approach is twofold: first, a large and statistically significant sample population may be analyzed one cell at a time, thus revealing the fine detail of heterogeneity in the expression of the cell surface markers within a population. Second, as has been demonstrated for analysis of specific components of the immune system, this method does permit fluorescence-activated sorting of cell types according to their different surface populations (8, 9, 14). 相似文献

Fad35R from Mycobacterium tuberculosis binds to the promoter site of Fad35 operon and its DNA binding activities are reduced in the presence of tetracycline and palmitoyl-CoA. We resolved the crystal structure of Fad35R using single-wavelength anomalous diffraction method (SAD). Fad35R comprises canonical DNA binding domain (DBD) and ligand binding domain (LBD), but displays several distinct structural features. Two recognition helices of two monomers in the homodimer are separated by ~ 48 Å and two core triangle-shaped ligand binding cavities are well exposed to solvent. Structural comparison with DesT and QacR structures suggests that ligand binding-induced movement of α7, which adopts a straight conformation in the Fad35R, may be crucial to switch the conformational states between repressive and derepressive forms. Two DBDs are packed asymmetrically, creating an alternative dimer interface which coincides with the possible tetramer interface that connects the two canonical dimers. Quaternary state of alternative dimer mimics a closed-state structure in which two recognition helices are distanced at ~ 35 Å and ligand binding pockets are inaccessible. Results of biophysical studies indicate that Fad35R has the propensity to oligomerize in solution in the presence of tetracycline. We present the first structure of a FadR homologue from mycobacterium and the structure reveals DNA and ligand binding features of Fad35R and also provides a view on alternative quaternary states that mimic open and closed forms of the regulator. 相似文献

Chemokines are an important group of soluble molecules with specialized functions in inflammation. The roles of many specialized chemokines and their receptors remain poorly understood in the human intervertebral disc. We investigated CXCL16 and its receptor, CXCR6, to determine their immunolocalization in disc tissue and their presence following exposure of cultured human annulus fibrosus cells to proinflammatory cytokines. CXCL16 is a marker for inflammation; it also can induce hypoxia-inducible factor 1α (HIF-1α), which is a phenotypic marker of heathy nucleus pulposus tissue. We found CXCL16 and CXCR6 immunostaining in many cells of the annulus portion of the disc. Molecular studies showed that annulus fibrosus cells exposed to IL-1ß, but not TNF-α, exhibited significant up-regulation of CXCL16 expression vs. control cells. There was no significant difference in the percentage of annulus cells that exhibited immunolocalization of CXCL16 in grade I/II, grade III or grade IV/V specimens. The presence of CXCL16 and its receptor, CXCR6, in the annulus in vivo suggests the need for future research concerning the role of this chemokine in proinflammatory functions, HIF-1α expression and disc vascularization. 相似文献

Water dispersions of egg phosphatidyl choline, dioleoyl phosphatidyl choline and lyso egg phosphatidyl choline have been studied by means of 220 MHz proton magnetic resonance techniques. The N+(CH3)3 proton signal, for phosphatidyl choline vesicles, consists of two components. The two components are thought to arise from N+(CH3)3 groups interior and exterior to the phosphatidyl choline vesicle. Anions were found to increase the separation of the two components. The effectiveness of the anions follows their order in the lyotropic series. For a given anion, the increase in component separation depends on the nature of the phospholipid. Iodine was found to modify the anion effect. The results are related to the results of other workers on water transport across lipid bilayer membranes and on phospholipid-halide-binding. 相似文献
