Single node explants of ‘Baccara’ rose were fed through the basal cut surface with 14C-sucrose in agar. Accumulation of 14C-metabolites by buds is a steady process, while a quick uptake occurs in the stem piece during the initial stage of incubation suggesting a passive uptake at this stage. The uppermost 5-leaflet node buds had an initially higher 14C-uptake than the lowest bud. This difference disappeared after 2–3 days when growth started in both buds. Light enhanced uptake of 14C-sucrose by both stem pieces and buds. Accumulation of 14C-metabolites by buds was in direct relationship to buds deviation from the direction of gravity. This phenomenon was not evident in the stem part of the explant.  相似文献

In the past decade, molecular probe‐based methods have proved successful in improving both the efficiency and accuracy of the identification of microorganisms, especially those that are devoid of distinct morphological features. However until recently, these methods had the major drawback of being limited to the identification of only one or just a few species at a time. With the use of DNA microarrays, it is possible to identify large numbers of taxa on a single‐glass slide, the so‐called phylochip. There are numerous microarray protocols in the literature. These protocols share the same principles, but vary in details, e.g. labelling approach or detergent concentration in the washing buffer. In this study, we show that even small variations in hybridization protocols can have a strong impact on the outcome of the microarray hybridization. An optimized protocol for species identification on phylochips is presented. The optimized protocol is the result of a joined effort of three laboratories to develop phylochips for microbial species identification.  相似文献

Wildfires have major effects on forest dynamics, succession and the carbon cycle in the boreal biome. They are a significant source of carbon emissions, and current observed changes in wildfire regimes due to changes in climate could affect the balance of the boreal carbon pool. A better understanding of postwildfire vegetation dynamics in boreal forests will help predict the future role of boreal forests as a carbon sink or source. Time series of Normalized Difference Vegetation Index (NDVI) and Normalized Difference Shortwave Infrared Index (NDSWIR) derived from Moderate Resolution Imaging Spectroradiometer (MODIS) aboard the Terra satellite were used to investigate whether characteristic temporal patterns exist for stands of different ages in the Siberian boreal forests and whether their postwildfire dynamics are influenced by variables such as prewildfire vegetation cover. Two types of forests, evergreen needle‐leaf (ENF) and deciduous needle‐leaf (DNF), were studied by analysing a sample of 78 burned forest areas. In order to study a longer time frame, a chronosequence of burned areas of different ages was built by coupling information on location and age provided by a forest burned area database (from 1992 to 2003) to MODIS NDVI and NDSWIR time series acquired from 2001 to 2005. For each of the burned areas, an adjacent unburned control plot representing the same forest type was selected, with the aim of separating the interannual variations caused by climate from changes in NDVI and NDSWIR behaviour due to a wildfire. The results suggest that it takes more than 13 years for the temporal NDVI and NDSWIR signal to recover fully after wildfire. NDSWIR, which is associated to canopy moisture, needs a longer recovery period than NDVI, which is associated to vegetation greenness. The results also suggest that variability observed in postwildfire NDVI and NDSWIR can be explained partially by the dominant forest type: while 13 years after a fire NDVI and NDSWIR are similar for ENF and DNF, the initial impact appears to be greater on the NDVI and NDSWIR of ENF, suggesting a faster recovery by ENF.  相似文献

Experiments were designed to provide information about the physiological basis of flood-tolerance in Senecio species. The oxygen concentration in roots of S. jacobaea L., S. viscosus L. and S. vulgaris L. became almost zero after transplantation to a solution of low oxygen concentration, and it was concluded that the flood-sensitivity of these Senecio species could be due to insufficient oxygen transport from the shoots to the roots. The oxygen concentration in the roots of the flood-tolerant S. congestus (R.Br.) DC., growing in a solution of low oxygen tension, was almost sufficient to maintain oxygen utilization at the rate observed in roots of plants, grown in an air-saturated solution. Oxygen utilization by roots of the flood-tolerant S. aquaticus Hill, growing in a solution of low oxygen tension, was inhibited 50%. However, the oxygen concentration in the roots of this species remained high enough to maintain cytochrome oxidase activity and oxidative phosphorylation at the rate observed in roots from an air-saturated environment. The activity of a second (“alternative”) oxidase must have been drastically reduced. Alternative NADH-oxidizing enzymes, like nitrate reductase which was induced by anaerobiosis in roots of S. aquaticus, might replace the regulatory function of the alternative oxidase. — Thus, in S. aquaticus root porosity and root length contributed to the maintenance of an oxygen concentration which was sufficient for uninhibited cytochrome oxidase activity and oxidative phosphorylation rate in roots growing in a solution of low oxygen tension.  相似文献

SYNOPSIS. The sensitivity of Paramecium tetraurelia (=P. aurelia syngen 4) cells to pulse treatments with various doses of Actinomycin D (AMD) was estimated by comparing the generation times of treated and untreated sister cells. It was found that the delay of division in treated cells depended on the concentration of AMD, on their “age” at the time of the pulse treatment, and on their individual sensitivity. Sensitivity of Paramecium to AMD changes during the cell cycle in a predictable way. About 3 1/2 hr before the normally expected cell fission (total generation time ~ 5 1/2 hr) there is a decrease of sensitivity. Thereafter, the cell enters a new stage with a progressive increase of sensitivity. This 2nd phase ends at the “transition point” (~ 2 hr before cell division), when sensitivity drops abruptly. The division process itself may be altered and slowed down by high concentrations of AMD, even if the drug is applied after the transition point, but this process can never be completely annulled. The impairment of the division mechanism may lead to morphologic anomalies in the offspring. Resorption of oral anlagen in P. tetraurelia probably never occurs during the cell cycle after AMD treatment. The reason for individual variability of the cells, mechanisms controlling development, and the question of an obligate sequence of gene action in each cell cycle are discussed.  相似文献

Although numerous reports have documented the effect of bacterially-inducedineffectiveness on root nodule structure, function, and plantgene expression, few studies have detailed the effect of theplant genome on similar parameters. In this report effective(N2-fixing) broadbean {Vicia faba L.) and plant-controlled ineffective(non-N2-fixing) broadbean recessive for the sym-1 gene werecompared for nodule structure, developmental expression of noduleenzyme activities, enzyme proteins, and mRNAs involved in Nassimilation, leghemoglobin (Lb) synthesis, and acetylene reductionactivity (ARA). During development of effective wild-type nodules,glutamine synthetase (GS), aspartate aminotransferase (AAT),phosphoenolpyruvate carboxylase (PEPC) and NADH-glutamate synthase(GOGAT) activities and enzyme proteins increased coincidentwith nodule ARA. The increases in GS, AAT, and PEPC were associatedwith increased synthesis of mRNAs for these proteins. Synthesisof Lb polypeptides and mRNAs during development of effectivenodules was similar to that of GS, AAT, and PEPC. By contrast,ineffective sym-1 nodules displayed little or no ARA and hadneither the increases in enzyme activities nor enzyme proteinsand mRNAs as seen for effective nodules. The effect of the sym-1gene appeared to occur late in nodule development at eitherthe stage of bacterial release from infection threads or differentiationof bacteria into bacteroids. High in vitro enzyme activities,enzyme polypeptides, and mRNA levels in parental effective noduleswere dependent upon a signal associated with effective bacteroidsthat was lacking in sym-1 nodules. Nodule organogenesis didnot appear to be a signal for the induction of GS, PEPC, AAT,and Lb expression in sym-1 nodules. Key words: Vicia faba, mutation, sym-1 gene, nodules  相似文献
