In shake-flask culture asymmetrin was produced by PenicIlliumthomiiaud Byssochlamys niveaduring autolysis. A bioassay wasdevised to estimate relative concentrations of this compound.Culture filtrates of P. thomiicontaining asymmetrin caused adecrease in wet weight, dry weight, and total nitrogen of Phaseolusvulgarisup to 18 days after treatment. At certain intervalsafter treatment with culture filtrate of B. nivea,respirationwas inhibited, and changes were observed in isotope ratio valuesof plants supplied with glucose-14 C. Plants treated previouslywith culture filtrates of B. nivearesponded but little to gibberelhcacid. Amplified Fragment Length Polymorphism (AFLP) was used to clarify the glacial history of the rare, disjunctly distributed, alpine cushion plant Androsace wulfeniana , which is endemic to the Eastern Alps (Austria and Italy). Disjunct populations in the Dolomites are genetically very distinct from those in the main distributional area. It is hypothesized that they are descendants of long-term isolated glacial survivors and are not a result of recent long-­distance dispersal. Within the main distributional area of the species in the central Eastern Alps, two groups of ­populations can be distinguished, which are congruent with hotspots of rare relictual vascular plant taxa. In the ­taxonomically closely related A. brevis growing in the Southern Alps (Italy, Switzerland), no genetic-geographical structure was found. Genetic variation is extremely low in disjunct populations of A. wulfeniana in the Dolomites and in A. brevis . Multiple molecular analyses provide a congruent and well‐supported phylogeny of the charadriiform family‐level taxa, which conflicts with previous hypotheses based on osteological data. In order to revise the latter and to identify new characters of phylogenetic significance, skeletons of most charadriiform family‐level taxa were examined and 49 characters analysed. Tree topology was sensitive to outgroup choice, but the result of the analysis rooted with Columbidae (doves and pigeons) recovered a monophyletic Scolopaci, Charadrii, and nonturnicid Lari. With regard to the inclusion of Alcidae and Glareolidae in the Lari, the results of the present study are also in better concordance with the new molecular phylogenies than previous analyses of morphological data. Furthermore, for the first time an apomorphy of a clade including Thinocoridae, Pedionomidae, Rostratulidae, and Jacanidae was identified. Inclusion of Turnicidae in the Lari could not be supported, but there is no strong morphological evidence for an alternative placement. Pluvianus shares derived osteological features with the Burhinidae, and its position in the molecular analyses likewise cannot be corroborated with morphological data. Based on the topology of the molecular consensus tree, the ancestral state of selected characters is reconstructed. Several years ago, it was observed that sterile microbial membrane preparations stimulated recovery of certain radiation-injured bacteria. Later it was noted that these same preparations reduce dissolved oxygen to water in a variety of environments, including bacteriological media. This reduction of oxygen is an enzymatic process and is influenced by parameters such as temperature, pH, and the availability of specific oxidizable substrates. Oxygenreducing membrane preparations can be made from several different bacterial species. When added to liquid or solid bacteriological media, membrane preparations rapidly produce and maintain anaerobic conditions favorable for the growth of a wide variety of oxygen-sensitive microorganisms. When used with a specifically designed disposable dish, membrane preparations allow the development of colonies of many anaerobic microorganisms on the surface of agar without the use of anaerobic hoods or other devices. In addition to providing conditions suitable for the growth of anaerobes, membrane preparations stimulate recovery of heat and cold injured bacteria of several different genera including facultative organisms. These results are reminiscent of the early observations regarding the recovery of radiation-injured bacteria. In addition to their usefulness in microbiology, oxygen-reducing membrane preparations have the potential for protecting a wide variety of oxygen-sensitive organic compounds.  相似文献

Activation of opioid or opioid-receptor-like (ORL1 a.k.a. NOP or orphanin FQ) receptors mediates analgesia through inhibition of N-type calcium channels in dorsal root ganglion (DRG) neurons (). Unlike the three types of classical μ, δ, and κ opioid receptors, ORL1 mediates an agonist-independent inhibition of N-type calcium channels. This is mediated via the formation of a physical protein complex between the receptor and the channel, which in turn allows the channel to effectively sense a low level of constitutive receptor activity (). Further inhibition of N-type channel activity by activation of other G protein-coupled receptors is thus precluded. ORL1 receptors, however, also undergo agonist-induced internalization into lysosomes, and channels thereby become cointernalized in a complex with ORL1. This then results in removal of N-type channels from the plasma membrane and reduced calcium entry (). Similar signaling complexes between N-type channels and GABAB receptors have been reported (). Moreover, both L-type and P/Q-type channels appear to be able to associate with certain types of G protein-coupled receptors (). Hence, interactions between receptors and voltage-gated calcium channels may be a widely applicable means to optimize receptor channel coupling.  相似文献
