Transgenic Trifolium subterraneum expressing a phytase gene (phyA) from Aspergillus niger were generated. Five independently transformed lines showed an average 77‐fold increase in exuded phytase activity in comparison with null segregant and wild‐type controls. Unlike other phosphatases, exuded phytase activity was unaffected by P supply, verifying the constitutive expression of phyA. Transgenic T. subterraneum grown in agar with P supplied as phytate, took up 1.3‐ to 3.6‐fold more P than controls and had equivalent P uptake to plants supplied with orthophosphate. This unique phenotype was compromised when the plants were grown in soil. None of the five lines showed increased shoot biomass or total P uptake in an unfertilized, low‐P soil taken from under permanent pasture. With addition of P, one of the five transgenic lines had consistently greater P nutrition compared with control plants. Despite variable growth and P nutrition responses, P uptake per root length was on average greater for transgenic lines. Exudation of phytase by transgenic T. subterraneum allowed utilization of P from phytate in non‐sorbing, sterile laboratory media, but was less effective when plants were grown in soil. Release of extracellular phytase is therefore not the only requirement for the acquisition of P from endogenous soil phytate by plants.  相似文献

SYNOPSIS. Sporulated oocysts of Eimeria nieschulzi Dieben, a rat coccidium, were exposed for 1 hr to Cobalt-60 γ-radiation (15, 30. or 60 k-rads), to heat (35, 40, or 45 C). or to both concurrently (15, 30, or 60 k-rads at 35 C) to compare the excystation capabilities of treated vs nontreated parasites. Intact, treated oocysts appeared structurally unaltered when viewed with the light microscope. Excystation of sporozoites occurred in all treated groups when their sporocysts were exposed to a trypsin-sodium taurocholate (TST) fluid, but after 150 min in TST the excystation rate was significantly lower than in non-treated sporocysts. Sporozoites which excysted from treated sporocysts were abnormal both in the excystation process and in their form and movement once outside the sporocyst.  相似文献

1. Nursery areas are commonly recognized as important habitats for the management and conservation of fish stocks. Here we report the use of nursery areas by an exploited offshore cichlid in Lake Malawi, Rhamphochromis longiceps .
2. Like all haplochromine cichlids that have been studied, the species is a maternal mouthbrooder that broods eggs for several weeks following spawning. We found evidence that during this brooding period females migrate from open water to release juveniles in three shallow peripheral waterbodies (Chia Lagoon, Unaka Lagoon, Lake Malombe). However, it was unclear whether there is geographical population structuring within the species, which could indicate stock differences in their use of these nursery habitats, or if the lake contains a genetically panmictic population that employs nursery habitats opportunistically.
3. To investigate spatial and temporal population structuring within the lake we genotyped populations at seven microsatellite DNA loci. Overall, we found no significant spatial structuring among juveniles from the peripheral lagoons or among breeding males within the main lake body. Moreover, we found no evidence of temporal structuring among males on the breeding grounds within Lake Malawi or females entering Chia lagoon. Together, these results suggest that Lake Malawi contains a genetically homogeneous population of R. longiceps .
4. At present we know little of the distribution of juvenile R. longiceps elsewhere in the Lake Malawi basin, but their absence from surveyed rocky and sandy littoral habitats makes it possible that the species is dependent upon a small number of nearshore nursery areas, including these lagoons. As such, conservation of lagoon habitats and monitoring of exploited fish stocks within them may be important for effective preservation of biodiversity within the catchment.  相似文献

A field study was conducted to determine how atmospheric and edaphic conditions influenced the water relations of avocado trees (Persea americana Mill. cv. Bacon). With high and low levels of incident photosynthetically active radiation (PAR, 400–700 nm wave length), and either wet or dry soil, leaf conductance decreased as the absolute humidity difference from leaf to air increased. For any water stress treatment, conductance was higher at high PAR than at low PAR. Both conductance and transpiration were higher in well-watered trees than in stressed trees, and in prestressed trees levels were intermediate to unstressed and stressed trees. A model for water flux through the soil-plant-atmosphere continuum was used to examine the relationship of leaf xylem pressure potential to transpiration in well-watered trees and in trees stressed by dry soil. There was a close linkage between leaf xylem pressure potential and transpiration in unstressed and previously stressed trees with high or low PAR, i.e. similar potentials occurred with equivalent transpiration regardless of previous treatment or time of day. In stressed trees, xylem pressure potential was lower than in unstressed trees both during the day and night, and at a given transpiration rate the potential was lower after 1400 h than before that time. The model indicated that in stressed trees xylem pressure potential was uncoupled from transpiration, presumably because of altered resistance in the soil-root portion of the transport system.  相似文献

CROSS has reported1, 2 chromosome complements in rodents ranging up to 2n=84 (in the North American geomyid Geomys breviceps) and 2n=86 (in the heteromyid Dipodomys merriami). Matthey has contested3 this claim; he maintained that in Geomys bursarius 2n =70 or 72 and he doubted whether a higher number had been confirmed. Numbers in the 2n=70 to 80 range have since been reported4 in several genera of Canidae and Ursidae. In general, rodent chromosomes fall into the lower group numbers but, among murines, Lophuromys aquillus is reported to have a chromosome complement of 70 (ref. 5) and Cricetomys gambianus one of 78 (ref, 6). In the gerbilline, Meriones shawii, the number can vary up to 78 (ref. 7).  相似文献

Squid rhodopsin (λmax 493 mµ)—like vertebrate rhodopsins—contains a retinene chromophore linked to a protein, opsin. Light transforms rhodopsin to lumi- and metarhodopsin. However, whereas vertebrate metarhodopsin at physiological temperatures decomposes into retinene and opsin, squid metarhodopsin is stable. Light also converts squid metarhodopsin to rhodopsin. Rhodopsin is therefore regenerated from metarhodopsin in the light. Irradiation of rhodopsin or metarhodopsin produces a steady state by promoting the reactions, See PDF for Equation Squid rhodopsin contains neo-b (11-cis) retinene; metarhodopsin all-trans retinene. The interconversion of rhodopsin and metarhodopsin involves only the stereoisomerization of their chromophores. Squid metarhodopsin is a pH indicator, red (λmax 500 mµ) near neutrality, yellow (λmax 380 mµ) in alkaline solution. The two forms—acid and alkaline metarhodopsin—are interconverted according to the equation, Alkaline metarhodopsin + H+ acid metarhodopsin, with pK 7.7. In both forms, retinene is attached to opsin at the same site as in rhodopsin. However, metarhodopsin decomposes more readily than rhodopsin into retinene and opsin. The opsins apparently fit the shape of the neo-b chromophore. When light isomerizes the chromophore to the all-trans configuration, squid opsin accepts the all-trans chromophore, while vertebrate opsins do not and hence release all-trans retinene. Light triggers vision by affecting directly the shape of the retinene chromophore. This changes its relationship with opsin, so initiating a train of chemical reactions.  相似文献

Abstract The sugarbeet root maggot Tetanops myopaeformis (Röder) overwinters as a freeze‐tolerant third‐instar larva. Although most larvae are considered to overwinter for only 1 year, some may exhibit prolonged diapause in the field. In the laboratory, they can live for over 5 years using a combination of diapause and post‐diapause quiescence. In the present study, the cold survival strategies of these larvae during storage is investigated by measuring their supercooling points in combination with survival data. Supercooling points (SCPs) change significantly during storage, highlighted by a marked increase in the range of SCPs recorded, although the ability to tolerate freezing is not affected. Additionally, a freezing event ‘re‐focuses’ the SCPs of aged larvae to levels similar to those seen at diapause initiation. This change in SCPs is dependant not only on the initial freezing event, but also on the parameters of the incubation period between freezing events. Finally, the temperatures of larval overwintering microhabitats are monitored during the 2007–2008 boreal winter. The results indicate that, although overwintering larva are physiologically freeze‐tolerant, they may essentially be freeze avoidant during overwintering via microhabitat selection.  相似文献

EXCESS fat deposition after castration is thought to be a response to hyperinsulinism induced by an increased level of adrenal glucocorticoids1. Two mutant alleles, lethal yellow (Ay) and viable yellow (Avy), at the agouti locus in the mouse induce excess fat deposition; the Ay allele has also been found to induce insulin resistance2. Katsh3 concluded that in adrenalectomized KL strain mice a considerable portion of the high insulin tolerance depended on normal adrenal function. In the inbred YS/Wf and VY/Wf strains, both yellow pheno-types modulate serum insulin concentration4. Castration of inbred YS strain males causes a large decrease in serum insulin4, suggesting a possible relationship1 to the concentration of adrenal glucocorticoids. Growth of allogeneic tumour cells has different effects on the serum insulin concentrations of YS and VY strain mice4.  相似文献
