Rooting and the Metabolism of Nicotine in Tobacco Callus Cultures   总被引:2,自引:0,他引:2  
The usefulness of exogenous nicotine as a factor in the induction of morphogenesis in a tobacco tissue culture medium has been demonstrated. Nicotiana rustica callus cell cultures were grown on a modified Murashige and Skoog medium with 2 mg/l indoleacetic acid (IAA) and 0.2 mg/l kinetin (MMS). Root morphogenesis was induced in roller tube callus cell cultures and solid callus cell cultures grown on MMS without kinetin supplemented with 10–100 mg/l nicotine. Optimal nicotine concentration for root induction was 50 mg/l. Other tests using varying combinations of IAA, kinetin and nicotine produced no obvious morphogenesis, although some changes in the amount of callus growth and endogenous protein concentration did correlate with nicotine concentration relative to the presence of IAA and/or kinetin. In liquid MMS medium, 14C-nicotine was primarily incorporated into the protein fraction of cultured cells while primarily incorporated into the cell wall and/or cell membrane fraction of cells cultured on MMS without kinetin in the medium. In MMS without IAA and MMS without both IAA and kinetin, there was incorporation, but to a lesser extent in both the protein and the cell wall and/or cell membrane fractions.  相似文献

Autumn injections of D-D mixture have been annually repeated for three years on a silt soil at Moulton (Holland, Lincolnshire) and a black fen soil at Prickwillow (Ely, Cambridgeshire), with different results. At Moulton there was an increased yield of tubers each time D-D was used, with no significant residual effects after the first year, and no marked long-term effect on the eelworm population. At Prick-willow D-D gave an increased yield in the first season only, with no positive residual effects on yield, and an apparent stimulating effect on the eelworm population. At Moulton in 1948 and 1950 (but not in 1949) the cost of the D-D treatment was heavily outweighed by the value of the resultant increase in crop. However, on the organic soil at Prickwillow D-D treatment was ineffective in 1949 and 1950 and the eelworm population, initially higher than at Moulton, remained at a level inducing failure of the potato crops.  相似文献

ABSTRACT. Developmental processes in multicellular organisms require structural elements, such as adhesion molecules, to stabilize cells at functional positions. In vertebrates, a series of extracellular matrix proteins, e.g. fibronectin and laminin are involved in cell adhesion. These proteins contain Arg-Gly-Asp [RGD] at their binding sites. Here we show that at concentrations above 2 mM the peptide GRG D SPK, comprising the tripeptide RGD (Arg-Gly-Asp), prevents the adhesiveness of cells of the marine amoeba Neopar-amoeba aestuarina. In addition, elevated levels of GRG D SPK cause cells to alter their shapes from those with digitiform subpseudopodia to rounded cells with small lobed pseudopodia. These cells detach from the substratum. These results are specific for the RGD sequence, because incubation in GRG E SPK solution at the same concentrations had no effect on cell attachment or structure. From these data we suggest that the structural adhesion molecules identified in vertebrates shows amino acid homologies with those found in unicellular protozoa.  相似文献

The cytoplasmic distribution of Cl in Nitella translucenswas assessed by means of two contrasting approaches. The firstinvolved the histochemical precipitation of Cl with Ag+followed by X-ray analytical verification of the silver precipitationproducts, and the second, quench-freezing of whole Nitella cellsfollowed by freeze-substitution in the presence of silver underanhydrous conditions. Both methods produced identical evidencefor Cl distribution, showing that a large proportionof the Cl is present in the stationary cortical gel layerwhich includes the chloroplasts. However, the chloroplasts appearedto be low in Cl content while the bulk of the Clappeared to be situated between the chloroplasts and plasmalemma. Experiments were carried out in other to detect the pathwayof the ‘fast component’ of halide ion transfer tothe vacuole. Br was supplied for various time intervalsto low Cl Nitella cells, followed by attempts to differentiatebetween AgCl and AgBr deposits. Solutions of various strengthof NH4OH or ammonium carbonate were used to remove AgCl (butnot AgBr) deposits by formation of a Ag(NH3)2+ complex. AlthoughX-ray analytical verification showed that the method had somepotential usefulness it could not be carried out successfullybecause of loss of structural detail caused by NH+4. The distribution and density of deposits near the plasmalemmasuggested the occurrence of a process in which cytoplasmic loadingis achieved by a sequential rupture and repair of the plasmalemmamembrane. Vesicles and reticulate structures in the streamingcytoplasmic phase generally showed very little deposit, butthese structures, together with the tonoplast, became greatlyenriched with deposits when cells had been given a brief exposers(3 min) to a Cl or Br solution. These rapid changesmay possibly be related to the ‘fast component’of halide ion transfer to the vacuole.  相似文献

Because of low net production in arctic and subarctic surface water, dissolved organic matter (DOM) discharged from terrestrial settings plays an important role for carbon and nitrogen dynamics in arctic aquatic systems. Sorption, typically controlling the export of DOM from soil, may be influenced by the permafrost regime. To confirm the potential sorptive control on the release of DOM from permafrost soils in central northern Siberia, we examined the sorption of DOM by mineral soils of Gelisols and Inceptisols with varying depth of the active layer. Water‐soluble organic matter in the O horizons of the Gelisols was less (338 and 407 mg C kg?1) and comprised more dissolved organic carbon (DOC) in the hydrophobic fraction (HoDOC) (63% and 70%) than in the O horizons of the Inceptisols (686 and 706 mg C kg?1, 45% and 48% HoDOC). All A and B horizons from Gelisols sorbed DOC strongly, with a preference for HoDOC. Almost all horizons of the Inceptisols showed a weaker sorption of DOC than those of the Gelisols. The C horizons of the Inceptisols, having a weak overall DOC sorption, sorbed C in the hydrophilic fraction (HiDOC) stronger than HoDOC. The reason for the poor overall sorption and also the preferential sorption of HiDOC is likely the high pH (pH>7.0) of the C horizons and the smaller concentrations of iron oxides. For all soils, the sorption of HoDOC related positively to oxalate‐ and dithionite–citrate‐extractable iron. The A horizons released large amounts of DOC with 46–80% of HiDOC. The released DOC was significantly (r=0.78, P<0.05) correlated with the contents of soil organic carbon. From these results, we assume that large concentrations of DOM comprising large shares of HiDOC can pass mineral soils where the active layer is thin (i.e. in Gelisols), and enter streams. Soils with deep active layer (i.e. Inceptisols), may release little DOM because of more frequent infiltration of DOM into their thick mineral horizons despite their smaller contents of reactive, poorly crystalline minerals. The results obtained for the Inceptisols are in agreement with the situation observed for streams connecting to Yenisei at lower latitudes than 65°50′ with continuous to discontinuous permafrost. The smaller sorption of DOM by the Gelisols is in agreement with the larger DOM concentrations in more northern catchments. However, the Gelisols preferentially retained the HoDOC which dominates the DOC in streams towards north. This discrepancy can be explained by additional seepage water from the organic horizons that is discharged into streams without intensive contact with the mineral soil.  相似文献

In the Lepidoptera, sex-linked genes have been found to be of importance for species differences in, for example, host-plant preference, and have been implicated in ecological speciation. Variation within species is typically not sex-linked. However, in the comma butterfly Polygonia c-album (Nymphalidae) an X-linked gene has been found to play a major role in determining differences in host-plant use between two well separated populations. For this reason, we studied the role of sex-linked genes for host-plant preference within a single Swedish population of this species. Three generations of females with known pedigrees were studied in the laboratory, and they were given a choice between Urtica dioica and Salix caprea in flight cages. We found strong variation among females and significant genetic variance for host-plant preference, but no evidence for major importance of sex linkage of host-plant preference on this local scale. To what extent the observed genetic variation was due to additive genes and/or effects of major genes was not clear from the maximum likelihood analysis. In a follow-up study we sampled females over a larger area. We found strong variation among females, but not among localities, suggesting an open population structure with strong gene flow. From the combined stock, a selection experiment was performed over 2 years and six generations. The selection lines diverged after the first generation of selection and remained separate, but did not diverge further, suggesting a low degree of narrow-sense heritability and that the genetic differences may be effects of major genes. The effect of the inoculation date on the spread of Beet mosaic virus (BtMV) in sugar beet field plots was studied. Two plants in the centre of each plot were inoculated with BtMV using Myzus persicae. The spread of the infection around these sources was monitored by inspecting the plants on two diagonal transects through the centre of the plot. Early inoculations resulted in a greater spread than late inoculations, but any inoculation before the onset of the aphid migration resulted in a similar‐sized spread. The spread was concentrated in patches around the inoculated plants, and its rate was explained by vector pressure, as shown by regression analysis and a mechanistic simulation model. This vector pressure was quantified using data obtained by catching aphids in a green water trap in the crop, catching aphids in a 12 m high suction trap at a distant location, and infection of bait plants from adjacent virus source plants. The daily total aphid catches obtained by a suction trap provided the best statistical explanation for the spread of this virus. The parameter r, describing the relationship between vector pressure and the rate of disease progress, was remarkably robust. This parameter varied less than 10% between treatments (infection date) within a single experiment, and less than a factor two between four experiments performed at different sites in two years. The robustness of this parameter suggests that the spread of a potyvirus may be predicted on the basis of the initial infection date and vector abundance.  相似文献

SUMMARY. 1. Resting eggs (ephippia) of Daphnia galeata, D. hyalina, D. cucullata and D. galeata X D. hyalina hybrids were collected from the upper 8 cm of sediments at various water depths from two glacial lakes in the Northern Federal Republic of Germany.
2. The horizontal distribution of 'intact' ephippia (i.e. those containing two apparently healthy eggs) was extremely patchy, with mean densities in the upper 8 cm ranging from 1325 to 113,907 m−2 (coefficient of variation, C=65.5%; Schöhsee, Holstein) and 25,343–159,143 m−2 (C=58.5%; Kel-lersee. Holstein). Ephippial densities were greatest in sediments from deep waters. The highest densities of intact ephippia were in the upper 4 cm of sediment, with progressively fewer with increasing depth.
3. A proportion of eggs obtained from sediments during autumn (September-November 1985) and spring (March-May 1986) were exposed to continuous light (approximately 18 W m−2) at three temperatures (6, 12, 20°C), and the incidence of hatches recorded.
4. Hatching success was consistently low (maximum 14.4%), with most hatching occurring at 12°C. There was no significant difference between the hatching success of ephippia collected during autumn and spring. Decapsulation of ephippial eggs inhibited hatching. Hatching was highly synchronous within each treatment, and the pattern did not differ significantly with sediment depth, though hatching success was lower in deep sediments. A huge reservoir of ephippia is present in lake sediments, of which a proportion may remain viable for many years. Periodic recruitment of hatchlings to the pelagic population may provide an important mechanism for the maintenance of genetic diversity in Daphnia populations, as well as affecting rates of evolutionary change.  相似文献

The first small nematodes to be described, free-living in the seventeenth century and plant-parasitic in the eighteenth, are identifiable only from their peculiar habitats; taxonomy came relatively late in nematology because adequate optical equipment was a prerequisite. In the study of the free-living and the plant-parasitic species, the development of two readily-presented aspects of taxonomy, figure drawing and mensuration (each showing oscillation between paucity and excess of detail), is traced in the work of five founders, all of whom were living in 1904: Bastian, Bütschli, de Man, Cobb and Goodey. Possible future developments in the identification of species and strains, the investigation of host-parasite relationships, and the training of nematologists are briefly discussed.  相似文献
