Abstract. 1. Euedaphic collembola alter their soil distribution in response to above‐ground aphid herbivory of Poa annua L. Graminae, a host grass. 2. Two mechanisms potentially underpin this effect. Carbon‐rich aphid honeydew falling onto the soil surface may affect mycophagous collembola; alternatively aphid‐induced changes in root biomass may be necessary to produce changes in collembola abundance. 3. When compared to a plant‐only control, aphid herbivory increased the number of collembola in the top 5 cm of soil, reduced both foliar and root biomass, and increased shoot/root ratio. Honeydew addition had no effect on collembola numbers or any recorded host‐plant parameter. 4. Honeydew deposition is not responsible for the increased numbers of collembola found in the upper soil after aphid herbivory; aphid‐induced reductions in root biomass may be the most important factor explaining knock‐on effects of aphid herbivory on soil fauna.  相似文献

1. Shallow lakes are often cited as classic examples of systems that exhibit trophic cascades but, whilst they provide good model systems with which to test general ecological theory and to assess long‐term community change, their food web linkages have rarely been resolved, so changes associated with the structure and dynamics of the ecological network as a whole are still poorly understood. 2. We sought to redress this, and to demonstrate the potential benefits of integrating palaeolimnological and contemporary data, by constructing highly resolved food webs and stable isotope derived measures of trophic interactions and niche space, for the extant communities of two shallow U.K. lakes from different positions along a gradient of eutrophication. The contemporary surface sediment cladoceran and submerged macrophyte assemblages in the less enriched site, Selbrigg Pond, matched the palaeolimnological assemblages of the more enriched site, Felbrigg Hall Lake, in its more pristine state during the 1920s. Thus, Selbrigg was a temporal analogue for Felbrigg, from which the consequences of long‐term eutrophication on food web structure could be inferred. These data represent the first steps towards reconstructing not only past assemblages (i.e. nodes within a food web), but also past interactions (i.e. links within a food web): a significant departure from much of the previous research in palaeolimnology. 3. The more eutrophic food web had far fewer nodes and links, and thus a less reticulate network, than was the case for the more pristine system. In isotopic terms, there was vertical compression in δ15N range (NR) and subsequent increased overlap in isotopic niche space, indicating increased trophic redundancy within Felbrigg. This structural change, which was associated with a greater channelling of energy through a smaller number of nodes as alternative feeding pathways disappear, could lead to reduced dynamic stability, pushing the network towards further simplification. These changes reflected a general shift from a benthic‐dominated towards a more pelagic system, as the plant‐associated subweb eroded. 4. Although these data are among the first of their kind, the palaeo‐analogue approach used here demonstrates the huge potential for applying food web theory to understand how and why these ecological networks change during eutrophication. Furthermore, because of the rich biological record preserved in their sediments, shallow lakes represent potentially important models for examining long‐term intergenerational dynamics, thereby providing a means by which models and data can be integrated on meaningful timescales – a goal that has long proved elusive in food web ecology.  相似文献

Regardless of their tissue of origin, multipotent mesenchymal stromal cells (MSCs) are commonly expanded in vitro for several population doublings to achieve a sufficient number of cells for therapy. Prolonged MSC expansion has been shown to result in phenotypical, morphological and gene expression changes in MSCs, which ultimately lead to the state of senescence. The presence of senescent cells in therapeutic MSC batches is undesirable because it reduces their viability, differentiation potential and trophic capabilities. Additionally, senescent cells acquire senescence-activated secretory phenotype, which may not only induce apoptosis in the neighboring host cells following MSC transplantation, but also trigger local inflammatory reactions. This review outlines the current and promising new methodologies for the identification of senescent cells in MSC cultures, with a particular emphasis on non-destructive and label-free methodologies. Technologies allowing identification of individual senescent cells, based on new surface markers, offer potential advantage for targeted senescent cell removal using new-generation senolytic agents, and subsequent production of therapeutic MSC batches fully devoid of senescent cells. Methods or a combination of methods that are non-destructive and label-free, for example, involving cell size and spectroscopic measurements, could be the best way forward because they do not modify the cells of interest, thus maximizing the final output of therapeutic-grade MSC cultures. The further incorporation of machine learning methods has also recently shown promise in facilitating, automating and enhancing the analysis of these measured data.  相似文献

The total activity (capacity) of phosphoenolpyruvate carboxylasein crude extracts of leaves of Bryophyllum fedtschenkoi, measuredon a fresh weight basis, varies considerably with leaf positionand with photoperiodic treatment. No diurnal variation in capacitywas detected under long – day or short – day conditions.The enzyme in freshly prepared crude extracts is about 50 timesmore sensitive to inhibition by malate at pH 7 than that inaged extracts, or in the fully purified state. Desensitizationin extracts proceeds rapidly unless protective measures aretaken, and appears to be irreversible. A pH-dependence studyshows that the effect of desensitization on the kinetic parametersof malate inhibition is identical to that of increasing thepH by 2.0 units over a wide range, but the maximum velocityat pH 7.8 is virtually unaffected. The significance of the resultsfor both the experimental determination of cyclic changes inenzyme capacity, and the theories concerning the mechanism ofa circadian rhythm of phosphoenolpyruvate carboxylase activityin vivo are discussed.  相似文献

The effect of pH on host plant ‘preference’ for strains of R. trifolii was studied using the fluorescent ELISA technique. Four white clover cultivars were compared growing at pH 5, 6 and 7 inoculated with 1:1 mixtures of two strains of R. trifolii, CP3B and R4. The growth of these two bacterial strains was also studied at the same pH levels in pure culture. At pH 5, in pure culture, CP3B grew very well but R4 failed to reach the log phase. CP3B also produced the majority of nodules at this pH (86%). At pH 7, in pure culture, R4 grew better than CP3B and also produced 66% of the nodules in the nodulation experiment. However, there was good evidence of host cultivar ‘preference’ with cv. Milkanova having no nodules inhabited by CP3B at pH 7 but cv. S100 having 32%. The results are discussed from the point of view of the establishment of white clover in acid soils and the usefulness of the fluorescent ELISA technique for Rhizobium strain identification is also emphasised.  相似文献
