SUMMARY. The difference between the results of viable and total counting procedures for bacteria are exemplified by vertical profiles from a deep and a shallow lake and from seasonal changes in the epilimnion and hypolimnion of a shallow eutrophic lake. The viable count was, on average, 0.25% of the total count, the greatest difference being noted in the anoxic hypolimnion, probably due to the inadequacy of the viable counting procedure for the isolation of bacteria from such samples. There was a general trend for the more nutrient-rich waters to support larger bacterial populations but such observations did not provide any further information on the factors responsible for the population changes observed. Seasonal fluctuations in the counts are studied and the qualitative and quantitative changes resulting from artificial enclosure of water are discussed. Not all the temporal changes could be explained and short-term changes resulting from nutrient additions to the experimental enclosures were not always reproducible. Horizontal variability was examined, found to be significant and could play an important role where water movement and turbulence is considerable. Results from six sites sampled between 1969 and 1974, representing total and viable bacterial population estimates and a total of eighteen independent or regressor variables were then subjected to principal components analysis. Results taken from the whole water column showed the overwhelming effect of the process of stratification on the bacterial population accounting for 30%-60% of its variability. Secondary components representing algal productivity could account for 10% to 20% of the variability. Many of the chosen regressor variables were acting as measures of the same phenomenon without providing significant information on what affected the bacterial population. To overcome this problem results from the hypolimnion and epilimnion were analysed separately. The analysis demonstrated the importance of seasonal changes in nutrient concentrations in the epilimnion and the development of anoxic conditions in the hypolimnion. Algal biomass, phosphate concentration and the interaction of pH and ammonia appeared to be important. It was concluded that most of the variability in the bacterial population estimates could have been explained by five of the regressor variables and that the factors most likely to provide more information would include some measure of predation and lake retention time. 相似文献

In a study of female House Sparrows Passer domesticus , it was established that almost all of the lean dry material lost from the flight muscles during the period of egg formation and laying was protein. The two large fractions of muscle protein, the sarcoplasmic and myofibrillar fractions, were measured biochemically. Protein in the myofibrillar fraction, which formed the greater part of the total protein, decreased during the study period. After graft inoculation with rubus yellow net virus (RYNV), 12 of 34 Rubus species and cultivars developed noticeable symptoms. R. macraei developed the most conspicuous symptoms and is recommended as an improved indicator plant. In attempts to determine the cause of raspberry veinbanding mosaic, a disease in which RYNV is involved, several European and North American red raspberry cvs were graft-inoculated with RYNV and three other aphid-borne viruses, black raspberry necrosis (BRNV), raspberry leaf mottle (RLMV) and raspberry leaf spot, singly and in all combinations. In periods of up to 4 yr, classical veinbanding mosaic symptoms developed in sensitive cvs only when they contained both RYNV and RLMV. These symptoms were intensified in plants co-infected with additional viruses. Veinbanding mosaic disease did not develop in any of 11 cvs infected with RYNV + BRNV, the combination of viruses previously assumed to be responsible for this disease in Britain and North America. 相似文献

In terms of control of slug pests, the differing modes of action of the two molluscicides metaldehyde and methiocarb appear to provide some advantages for the former during warm, dry weather and for the latter during cool, wet weather. With a view to exploiting these different modes of action, the feeding behaviour of Deroceras reticulatum (Müller) presented with molluscicidal baits containing various combinations of metaldehyde with methiocarb was investigated. Voluntary feeding trials conducted in the laboratory with both candidate and standard treatments (containing metaldehyde or methiocarb alone) are described. By combining the two active ingredients in certain ratios the total concentration of active ingredients may be reduced without adversely affecting the efficacy of a bait formulation in terms of slug mortality. 相似文献

The flexuous filamentous particles of wineberry latent virus (WLV) were found to measure 620. 12 nm and not 510. 12 nm as previously reported. Analysis of dsRNA from infected plants detected a major species of c. 5.7. 106 mol. wt and minor species of lower mol. wt. Purified virus particles formed a major and a minor buoyant density component in solutions of caesium salts with densities of 1.26 and 1.25 g cm-3 in Cs2SO4 and 1.30 and 1.29 g cm-3 in CsCl. The particles contained a single nucleic acid species, presumably single stranded RNA, and a single polypeptide of estimated mol. wt 2.78. 106 and 31 000 respectively. In indirect ELISA, purified particles of WLV and particles in plant sap failed to react specifically with antiserum to nine carlaviruses, 12 potexviruses, three capilloviruses or apple chlorotic leafspot closterovirus, nor was WLV found to react with several of these antisera in immunosorbent electron microscopy or immunoblots. Calcicolous plants are generally more Fe-efficient than calcifugous plants, because they respond to Fe stress by releasing H-ions and “reductants” from their roots that causes Fe to become available. The objective of our study was to determine if differential response to Fe stress in calcicolous and calcifugous varieties affects nitrate reductase activity. T3238FER (Fe-efficient) and T3238fer (Fe-inefficient) tomato (Lycopersicon esculentum Mill.) cultivars were grown in nutrient solutions supplied with N as NH4+-N plus NO3?-N, and as NO3?-N only. The chemical reactions induced by Fe stress concomitantly increased nitrate reductase activity in roots and tops of calcicolous, but not in calcifugous tomato. This nitrate reductase activity decreased, however, when Fe was made available to the plants. When Fe stress was eliminated by adding Fe, nitrate reductase activity was comparable in the two cultivars. 相似文献
