Aqueous solutions combining a high concentration of formaldehyde (4%) with low concentrations of glutaraldehyde (0.5--01%) have been used to simultaneously localize amines by the formation of fluorescent products and to fix central nervous tissue for electron microscopy. The fluorescence reaction is produced by the aldehyde mixture at room temperature and the fluorescence is stable when the tissue is maintained in aqueous solution. This means that nerve cell bodies and terminal fields which contain catecholamines can be located accurately in vibratome sections at the light microscope level and, after further processing, can be examined under the electron microscope. With 1% glutaraldehyde in the aldehyde mixture, ultrastructural details are well preserved; there is no significant distortion of any component of the tissue. If vibratome or cryostat sections are dried against glass slides, the intensity of the fluorescence reaction is enhanced and the sections can be permanently mounted. 相似文献

The primary moult of individually colour-ringed, adult yellow-nosed albatrosses at nests on Gough Island was examined in 1983 and related to the status of each bird and its breeding history in the previous year. Adults renew only about half of their primaries each winter and suspend moult while breeding. Birds that bred successfully renewed fewer primaries than did unsuccessful birds or nonbreeders. There were no differences in primary moult between the sexes or in relation to size. Yellow-nosed albatrosses show complex wave moult as an adaptation to slow renewal of flight feathers. The energy, nutrient or time requirements for feather renewal may conflict with breeding annually so that there is a trade-off between the extent of moult desirable to maintain flight efficiency and the benefits of breeding in successive seasons. 相似文献

In an analysis of data collected between 1954 and 1993, the breeding success of swifts Apus apus was found to have been positively related to temperatures in June, especially in recent years. Adult survival until the next breeding season, by contrast, was found to have been more vulnerable to low temperatures in July, at the end of the breeding season. In addition to the effects of changes in June and July temperatures, both breeding success and survival have shown further long-term trends, increasing and then decreasing through the study period. A simulation integrating all these effects suggests that lifetime reproductive success (LRS) in influenced more by changes in lifespan than annual breeding success, and so fluctutations in July temperatures, but not June temperatures, would have important effects. 相似文献

Mercury concentrations in albatrosses and some other large procellariiforms are very much higher than found in other groups of seabirds. Analysis of mercury concentrations in feather samples collected prior to 1950 and after this date showed slight, but significant, increases in three out of ten sub-Antarctic procellariiform species. The lack of widespread and pronounced increases in mercury concentrations in procellariiforms between these periods may indicate that industrial and agricultural emissions of mercury in the southern hemisphere have been relatively minor and the high concentrations are predominantly due to natural processes. Mercury concentrations were relatively consistent between body feathers of individuals, and showed no variation related to adult age (in years). Within species, mercury concentrations tended to be highest in New Zealand populations and lowest in Falkland Island and South Georgia populations. Increasing winter temperatures are expected to cause seasonal activity of Ixodes ricinus ticks to extend further into the winter. We caught birds during winter months (November to February) at a site in the west of Scotland over a period of 24 years (1993–1994 to 2016–2017) to quantify numbers of attached I. ricinus and to relate these to monthly mean temperature. No adult ticks were found on any of the 21,731 bird captures, but 946 larvae and nymphs were found, with ticks present in all winter months, on 16 different species of bird hosts. All ticks identified to species were I. ricinus. I. ricinus are now active throughout the year in this area providing temperature permits. No I. ricinus were present in seven out of eight months when the mean temperature was below 3.5º C. Numbers of I. ricinus attached to birds increased rapidly with mean monthly temperatures above 7º C. Winter temperatures in Scotland have been above the long‐term average in most years in the last two decades, and this is likely to increase risk of tick‐borne disease. 相似文献

We investigated the effects of increased egg production on body condition as well as on measures of reproductive performance in great skuas, Stercorarius skua, over two subsequent years. We experimentally increased egg production from the normal two to six eggs. Six eggs might also be produced under natural circumstances after repeated clutch loss. After the production of the last egg we measured: (i) body mass, (ii) pectoral muscle, and (iii) haematocrit, total red blood cell count and mean corpuscular volume, as indicators of body condition. We took the same measurements of control females who had produced the normal clutch of two eggs. The measurements were repeated one year after the manipulation, and survival, laying dates, clutch sizes and hatching success were recorded for up to three consecutive years. After producing six eggs, females were lighter, had smaller pectoral muscles and lower haematological values than control females. Hatching success of eggs was significantly reduced. Even one year after the experiment there were still differences in body condition. Annual survival was not affected by the manipulations, although there was an indication that survival costs depended on whether chicks were raised after the increased egg production. While pair bonds and egg sizes were not affected in the post‐experimental year, females started breeding significantly later than in the previous year. Two years after the experiment laying dates had advanced again and were not different from those of control females. This pattern of maintaining survival and egg sizes, but delaying breeding in the post‐experimental year was found for two independent groups of females which had both been subjected to increased egg production. These results present evidence that increased egg production can have long‐term effects on female body condition and aspects of reproduction. However, although present, the costs of extra eggs appear to have been relatively small in the great skua in comparison to the two other bird species for which inter‐annual effects have been reported. 相似文献

Antibodies against choline acetyltransferase (ChAT) and the vesicular acetylcholine transporter (VAChT) were used to determine whether neurons that have previously been identified as intrinsic primary afferent neurons in the guinea-pig small intestine have a cholinergic phenotype. Cell bodies of primary afferent neurons in the myenteric plexus were identified by their calbindin immunoreactivity and those in the submucous plexus by immunoreactivity for substance P. High proportions of both were immunoreactive for ChAT, viz. 98% of myenteric calbindin neurons and 99% of submucosal substance P neurons. ChAT immunoreactivity also occurred in all nerve cell bodies immunoreactive for calretinin and substance P in the myenteric plexus, but in only 16% of nerve cells immunoreactive for nitric oxide synthase. VAChT immunoreactivity was in the majority of calbindin-immunoreactive varicosities in the myenteric ganglia, submucous ganglia and mucosa and also in the majority of the varicosities of neurons that were immunoreactive for calretinin and somatostatin and that had been previously established as being cholinergic. We conclude that the intrinsic primary afferent neurons are cholinergic and that they may release transmitter from their sensory endings in the mucosa. 相似文献
