The distribution of neurokinin-2 (NK2) tachykinin receptors was investigated by immunohistochemistry in the guinea-pig oesophagus,
stomach, small and large intestine. Receptor immunoreactivity occurred at the surfaces of smooth muscle cells throughout the
digestive tract. Nerve fibre varicosities in enteric ganglia were also immunoreactive. In myenteric ganglia, these varicosities
were most numerous in the ileum, frequent, but less dense, in the proximal colon and caecum, rare in the distal colon, extremely
infrequent in the rectum and duodenum, and absent from the stomach and oesophagus. Reactive varicosities were rare in the
submucous ganglia. Reactive nerve fibres in the mucosa were only found in the caecum and proximal colon. Strong NK2 receptor
immunoreactivity was also found on the surfaces of enterocytes at the bases of mucosal glands in the proximal colon. Receptors
were not detectable on the surfaces of nerve cells or on non-terminal axons. Reactivity did not occur on nerve fibres innervating
the muscle. Denervation studies showed that the immunoreactive varicosities in the myenteric plexus of the ileum were the
terminals of descending interneurons. Immunoreactivity for nitric oxide synthase was colocalised with NK2 receptor (NK-R)
immunoreactivity in about 70% of the myenteric varicosities in the small intestine. Bombesin immunoreactivity occurred in
about 30% of NK2-R immunoreactive varicosities in the small intestine.

Distant populations of animals may share their non-breeding grounds or migrate to distinct areas, and this may have important consequences for population differentiation and dynamics. Small burrow-nesting seabirds provide a suitable case study, as they are often restricted to safe breeding sites on islands, resulting in a patchy breeding distribution. For example, Thin-billed prions Pachyptila belcheri have two major breeding colonies more than 8,000 km apart, on the Falkland Islands in the south-western Atlantic and in the Kerguelen Archipelago in the Indian Ocean. We used geolocators and stable isotopes to compare at-sea movements and trophic levels of these two populations during their non-breeding season, and applied ecological niche models to compare environmental conditions in the habitat. Over three winters, birds breeding in the Atlantic showed a high consistency in their migration routes. Most individuals migrated more than 3000 km eastwards, while very few remained over the Patagonian Shelf. In contrast, all Indian Ocean birds migrated westwards, resulting in an overlapping nonbreeding area in the eastern Atlantic sector of the Southern Ocean. Geolocators and isotopic signature of feathers indicated that prions from the Falklands moulted at slightly higher latitudes than those from Kerguelen Islands. All birds fed on low trophic level prey, most probably crustaceans. The phenology differed notably between the two populations. Falkland birds returned to the Patagonian Shelf after 2-3 months, while Kerguelen birds remained in the nonbreeding area for seven months, before returning to nesting grounds highly synchronously and at high speed. Habitat models identified sea surface temperature and chlorophyll a concentration as important environmental parameters. In summary, we show that even though the two very distant populations migrate to roughly the same area to moult, they have distinct wintering strategies: They had significantly different realized niches and timing which may contribute to spatial niche partitioning. 相似文献

Hair cells of the mammalian cochlea are specialized for the dynamic coding of sound stimuli. The transduction of sound waves into electrical signals depends upon mechanosensitive hair bundles that project from the cell's apical surface. Each stereocilium within a hair bundle is composed of uniformly polarized and tightly packed actin filaments. Several stereociliary proteins have been shown to be associated with hair bundle development and function and are known to cause deafness in mice and humans when mutated. The growth of the stereociliar actin core is dynamically regulated at the actin filament barbed ends in the stereociliary tip. We show that Eps8, a protein with actin binding, bundling, and barbed-end capping activities in other systems, is a novel component of the hair bundle. Eps8 is localized predominantly at the tip of the stereocilia and is essential for their normal elongation and function. Moreover, we have found that Eps8 knockout mice are profoundly deaf and that IHCs, but not OHCs, fail to mature into fully functional sensory receptors. We propose that Eps8 directly regulates stereocilia growth in hair cells and also plays a crucial role in the physiological maturation of mammalian cochlear IHCs. Together, our results indicate that Eps8 is critical in coordinating the development and functionality of mammalian auditory hair cells. 相似文献

Factors affecting breeding distribution of Storm-petrels Hydrobates pelagicus in Orkney and Shetland

Capsule The main factors are past and present human activities, especially the introduction of rats to islands. Aims To assess factors that influence breeding distribution and abundance of Storm-petrel. Methods We used a database for 142 islands in Shetland and Orkney. Breeding status of Storm-petrel was related to data for each island on introduced and indigenous predators, other human-related features, and aspects of island geography. Results Although 92% of the total land area of the archipelagos comprised islands with rats present, Storm-petrel colonies were almost totally restricted to rat-free islands. They also occurred more frequently on islands with cliffs, far from neighbouring islands with humans, and on islands with a low rate of human visits. Colony size was smaller on the smallest occupied islands. Breeding numbers of Great Skuas Stercorarius skua, Great Black-backed Gulls Larus marinus, and Storm-petrels all correlated, as each increased with island size. Conclusions The presence or absence of rats is the single most important influence on Storm-petrel breeding distribution in Orkney and Shetland. However, geographical and human-related effects, such as the presence of cliffs or the occurrence of human visits, also appear to influence the distribution of Storm-petrels, whereas avian predators appear to have had little effect until now. 相似文献

Background
Bioelectrical impedance analysis (BIA) is a potentially valuable method for assessing lean mass and body fat levels in children from different ethnic groups. We examined the need for ethnic- and gender-specific equations for estimating fat free mass (FFM) from BIA in children from different ethnic groups and examined their effects on the assessment of ethnic differences in body fat.Methods
Cross-sectional study of children aged 8–10 years in London Primary schools including 325 South Asians, 250 black African-Caribbeans and 289 white Europeans with measurements of height, weight and arm-leg impedance (Z; Bodystat 1500). Total body water was estimated from deuterium dilution and converted to FFM. Multilevel models were used to derive three types of equation {A: FFM = linear combination(height+weight+Z); B: FFM = linear combination(height2/Z); C: FFM = linear combination(height2/Z+weight)}.Results
Ethnicity and gender were important predictors of FFM and improved model fit in all equations. The models of best fit were ethnicity and gender specific versions of equation A, followed by equation C; these provided accurate assessments of ethnic differences in FFM and FM. In contrast, the use of generic equations led to underestimation of both the negative South Asian-white European FFM difference and the positive black African-Caribbean-white European FFM difference (by 0.53 kg and by 0.73 kg respectively for equation A). The use of generic equations underestimated the positive South Asian-white European difference in fat mass (FM) and overestimated the positive black African-Caribbean-white European difference in FM (by 4.7% and 10.1% respectively for equation A). Consistent results were observed when the equations were applied to a large external data set.Conclusions
Ethnic- and gender-specific equations for predicting FFM from BIA provide better estimates of ethnic differences in FFM and FM in children, while generic equations can misrepresent these ethnic differences. 相似文献98.

Periods of ischemia followed by restoration of blood flow cause ischemia/reperfusion (I/R) injury. In the intestine, I/R damage
to the mucosa and neurons is prominent. Functionally, abnormalities occur in motility, most conspicuously a slowing of transit,
possibly as a consequence of damage to neurons and/or muscle. Here, we describe degenerative and regenerative changes that
have not been previously reported in intestinal muscle. The mouse small intestine was made ischemic for 1 h, followed by re-perfusion
for 1 h to 7 days. The tissues were examined histologically, after hematoxylin/eosin and Masson’s trichrome staining, and
by myeloperoxidase histochemistry to detect inflammatory reactions to I/R. Histological analysis revealed changes in the mucosa,
muscle, and neurons. The mucosa was severely but transiently damaged. The mucosal surface was sloughed off at 1–3 h, but re-epithelialization
occurred by 12 h, and the epithelium appeared healthy by 1–2 days. Longitudinal muscle degeneration was followed by regeneration,
but little effect on the circular muscle was noted. The first signs of muscle change were apparent at 3–12 h, and by 1 and
2 days, extensive degeneration within the muscle was observed, which included clear cytoplasm, pyknotic nuclei, and apoptotic
bodies. The muscle recovered quickly and appeared normal at 7 days. Histological evidence of neuronal damage was apparent
at 1–7 days. Neutrophils were not present in the muscle layers and were infrequent in the mucosa. However, they were often
seen in the longitudinal muscle at 1–3 days and were also present in the circular muscle. Neutrophil numbers increased in
the mucosa in both I/R and sham-operated animals and remained elevated from 1 h to 7 days. We conclude that I/R causes severe
longitudinal muscle damage, which might contribute to the long-term motility deficits observed after I/R injury to the intestine. 相似文献

The effect of age and year on the survival of breeding adult Great Skuas Catharacta skua in Shetland

The survival rates of breeding adult Great Skuas Catharacta skua were examined at Foula, the largest colony in the world, where numbers have been declining since the late 1970s. Resightings of colour-ringed breeding adults over a 12-year period were analysed using Cormack-Jolly Seber models to estimate survival rates. Annual survival rates averaged 0.89 but varied among years between 0.82 and 0.93, with annual variations being temporally associated with variations in sandeel abundance during the breeding season. Most birds appeared to die outside the breeding season and so it is possible that nutritional stress and reproductive costs of breeding in years of poor food supply affect survivorship on migration or in the wintering range. Survival rates of adult Great Skuas were affected by their age according to a quadratic equation, with survival increasing significantly with age from 0.73 in 5-year-old-birds to between 0.85 and 0.96 in birds from 7 to 22 years old, with a sharp decline to between 0.75 and 0.87 in birds over 22 years old. Year effects were evident when controlling for age, indicating that annual variations in survival rates are not explained by changes in age-composition of the marked population among years. 相似文献

PKC is involved in mediating the tonic component of gastrointestinal smooth muscle contraction in response to stimulation by agonists for G protein-coupled receptors. Here, we present pharmacological and immunohistochemical evidence indicating that a member of the novel PKC isoforms, PKC-delta, is involved in maintaining muscarinic receptor-coupled tonic contractions of the guinea pig ileum. The tonic component of carbachol-evoked contractions was enhanced by an activator of conventional and novel PKCs, phorbol 12,13-dibutyrate (PDBu; 200 nM or 1 microM), and by an activator of novel PKCs, ingenol 3,20-dibenzoate (IDB; 100 or 500 nM). Enhancement was unaffected by concentrations of bisindolylmaleimide I (BIM-I; 22 nM) that block conventional PKCs or by a PKC-epsilon-specific inhibitor peptide but was attenuated by higher doses of BIM-I (2.2 microM). Relevant proteins were localized at a cellular and subcellular level using confocal analysis. Immunohistochemical staining of the ileum showed that PKC-delta was exclusively expressed in smooth muscles distributed throughout the layers of the gut wall. PKC-epsilon immunoreactivity was prominent in enteric neurons but was largely absent from smooth muscle of the muscularis externa. Treatment with PDBu, IDB, or carbachol resulted in a time- and concentration-dependent translocation of PKC-delta from the cytoplasm to filamentous structures within smooth muscle cells. These were parallel to, but distinct from, actin filaments. The translocation of PKC-delta in response to carbachol was significantly reduced by scopolamine or calphostin C. The present study indicates that the tonic carbachol-induced contraction of the guinea pig ileum is mediated through a novel PKC, probably PKC-delta. 相似文献
