We examined how UV radiation and phosphorus (P) affect the taxonomic composition, abundance, and biomass of phytoplankton in an oligotrophic boreal lake. We exposed phytoplankton to three different solar radiation regimes (PAR + UV‐A radiation [UVAR]+ UV‐B radiation [UVBR], PAR + UVAR, and PAR only) and to five levels of P. The biomass of small chrysophytes was reduced by 350% after exposure to PAR + UVAR + UVBR compared with PAR only. No other taxa were found to be negatively affected by exposure to UVBR. Several taxa (e.g. Chry‐ sochromulina laurentiana Kling) were sensitive to UVAR, whereas others (e.g. Tabellaria flocculosa (Roth) Kutzing) were not affected by UV radiation exposure. Principal components analysis ordination separated phytoplankton that were negatively affected by UV radiation and/or positively affected by P treatments (e.g. small chrysophytes, Cryptomonas rostratiformis, T. flocculosa) from those that generally were unaffected by either treatment (e.g. desmids, some Cyanobacteria). Richness, Shannon‐Weaver diversity, and evenness were significantly higher in phytoplankton communities shielded from UVAR and UVBR. The relationship between diversity and richness was positive in all phytoplankton samples except in those exposed to UVBR. Thus, UVBR‐exposed phytoplankton communities were dominated by a few species even though the number of taxa remained relatively unchanged. Consequently, alterations in the UV environments of lakes resulting from climate warming (e.g. drought) and land‐use change (e.g. increased P export) will likely promote shifts in the community composition of lake phytoplankton.  相似文献

A sample of 117 fossil cercopithecids has been collected from the Middle Pleistocene site of Asbole, Afar Region, Ethiopia. A minimum of five species is present. There are two species of Cercopithecini, here recognized as cf. Chlorocebus aff. aethiops, and cf. Chlorocebus cf. patas. There are also two species of Papionini: Papio hamadryas ssp. indet. and Theropithecus oswaldi leakeyi. Finally, there is a single species of colobine present, Colobus sp. indet. The assemblage is chronologically constrained and is derived from sediments dated to approximately 600 ka. Within this sample Colobus sp. is by far the most common species present, outnumbering the other four species combined. The cercopithecid assemblage is most consistent with a woodland habitat, corroborating an earlier interpretation based on the non-primate fauna. Taxonomic, biogeographic, and evolutionary implications of the assemblage are also discussed.  相似文献

Theoreticians predict that animal 'personality' traits may be maladaptive if fixed throughout different contexts, so the present study aimed to test whether these traits are fixed or plastic. Rainbow trout (Onchorhyncus mykiss) were given emboldening or negative experiences in the forms of watching bold or shy individuals responding to novelty or winning or losing fights to examine whether prior experience affected boldness. Bold individuals that lost fights or watched shy demonstrators became more shy by increasing their latency to approach a novel object, whereas shy observers that watched bold demonstrators remained cautious and did not modify their responses to novelty. Shy winners became bolder and decreased their latency to approach a novel object, but shy losers also displayed this shift. In comparison, control groups showed no change in behaviour. Bold fishes given negative experiences reduced their boldness which may be an adaptive response; however, shy fishes may base their strategic decisions upon self-assessment of their relative competitive ability and increase their boldness in situations where getting to resources more quickly ensures they outcompete better competitors.  相似文献

Background  

Aberrant activation of the Hedgehog (Hh) signaling pathway in different organisms has shown the importance of this family of morphogens during development. Genetic screens in zebrafish have assigned specific roles for Hh in proliferation, differentiation and patterning, but mainly as a result of a loss of its activity. We attempted to fully activate the Hh pathway by removing both receptors for the Hh proteins, called Patched1 and 2, which are functioning as negative regulators in this pathway.  相似文献

Poly(ADP-ribose) polymerase (PARP) senses DNA breaks and facilitates DNA repair via the polyADP-ribosylation of various DNA binding and repair proteins. We explored the mechanism of potentiation of temozolomide cytotoxicity by the PARP inhibitor ABT-888. We showed that cells treated with temozolomide need to be exposed to ABT-888 for at least 17 to 24 hours to achieve maximal cytotoxicity. The extent of cytotoxicity correlates with the level of double-stranded DNA breaks as indicated by gammaH2AX levels. In synchronized cells, damaging DNA with temozolomide in the presence of ABT-888 during the S phase generated high levels of double-stranded breaks, presumably because the single-stranded DNA breaks resulting from the cleavage of the methylated nucleotides were converted into double-stranded breaks through DNA replication. As a result, treatment of temozolomide and ABT-888 during the S phase leads to higher levels of cytotoxicity. ABT-888 inhibits poly(ADP-ribose) formation in vivo and enhances tumor growth inhibition by temozolomide in multiple models. ABT-888 is well tolerated in animal models. ABT-888 is currently in clinical trials in combination with temozolomide.  相似文献

The application of LC-MS for untargeted urinary metabolite profiling in metabonomic research has gained much interest in recent years. However, the effects of varying sample pre-treatments and LC conditions on generic metabolite profiling have not been studied. We aimed to evaluate the effects of varying experimental conditions on data acquisition in untargeted urinary metabolite profiling using UPLC/QToF MS. In-house QC sample clustering was used to monitor the performance of the analytical platform. In terms of sample pre-treatment, results showed that untreated filtered urine yielded the highest number of features but dilution with methanol provided a more homogenous urinary metabolic profile with less variation in number of features and feature intensities. An increased cycle time with a lower flow rate (400mul/min vs 600mul/min) also resulted in a higher number of features with less variability. The step elution gradient yielded the highest number of features and the best chromatographic resolution among three different elution gradients tested. The maximum retention time and mass shift were only 0.03min and 0.0015Da respectively over 600 injections. The analytical platform also showed excellent robustness as evident by tight QC sample clustering. To conclude, we have investigated LC conditions by studying variability and repeatability of LC-MS data for untargeted urinary metabolite profiling.  相似文献

The advent of molecular markers has created opportunities for a better understanding of quantitative inheritance and for developing novel strategies for genetic improvement of agricultural species, using information on quantitative trait loci (QTL). A QTL analysis relies on accurate genetic marker maps. At present, most statistical methods used for map construction ignore the fact that molecular data may be read with error. Often, however, there is ambiguity about some marker genotypes. A Bayesian MCMC approach for inferences about a genetic marker map when random miscoding of genotypes occurs is presented, and simulated and real data sets are analyzed. The results suggest that unless there is strong reason to believe that genotypes are ascertained without error, the proposed approach provides more reliable inference on the genetic map.  相似文献
