- The formation and reversion of spheroplasts of the diaminopimelic acid-auxotrophic mutant Escherichia coli K 12, 335, dap −, R+TEM in a medium lacking diaminopimelic acid have been investigated by microphotography: During their development from rod form cells to spheroplasts cells on slide-surface-agar preparations underwent two successive cell divisions in the course of which the cells retained their rod form. The cells formed by these divisions partitioned into a varying number of spheroplasts of different size. The reversion of spheroplasts to rod form cells, started by the addition of diaminopimelic acid showed two characteristic steps: Each spheroplast partitioned again into several spheroplast-like cell bodies; most of them reverted directly to rod form cells.
- The release of the R-factor mediated periplasmic TEM-β-lactamase, E. C. 3.4.2.6., into the growth medium during the development of spheroplasts attained more than 50% of the entire TEM-β-lactamase activity.

Most organisms need to acquire various resources to survive and reproduce. Individuals should adjust their behavior to make optimal use of the landscape and limit the costs of trade-offs emerging from the use of these resources. Here we study how African elephants Loxodonta africana travel to foraging places between regular visits to waterholes. Elephant herds were tracked using GPS collars during two consecutive dry seasons in Hwange National Park, Zimbabwe. We segmented each individual movement track at each visit to water to define foraging trips, and then used trip-level statistics to build an understanding of movement strategies. Travel speed within these individually-consistent movement bouts was also analyzed to understand if speed was better linked to distance to water or progression in the trip over time. We found that elephants went further from water when drinking less often, which could result from a trade-off between drinking and foraging in less depleted, far from water, places. Speed increased towards the beginning and the end of the trips, and was also greater than observed during the wet season, suggesting that elephants were trying to save time. Numerous short trips traveled at greater speed, particularly when commuting to a different waterhole, was tentatively explained by the inability to drink at specific waterholes due to intra-specific interference. Unexpectedly elephants did not always minimize travel time by drinking at the closest waterhole, but the extra distance traveled remained never more than a few kilometers. Our results show how individuals may adjust movement behavior to deal with resource trade-offs at the landscape scale. We also highlight how behavioral context, here progression in the trip, may be more important than spatial context, here distance to water, in explaining animal movement patterns. 相似文献

Two new proteins, named perlucin and perlustrin, with M(r) 17,000 and 13,000, respectively, were isolated from the shell of the mollusc Halotis laevigata (abalone) by ion-exchange chromatography and reversed-phase HPLC after demineralization of the shell in 10% acetic acid. The sequence of the first 32 amino acids of perlucin indicated that this protein belonged to a heterogeneous group of proteins consisting of a single C-type lectin domain. Perlucin increased the precipitation of CaCO(3) from a saturated solution, indicating that it may promote the nucleation and/or the growth of CaCO(3) crystals. With pancreatic stone protein (lithostathine) and the eggshell protein ovocleidin 17, this is the third C-type lectin domain protein isolated from CaCO(3) biominerals. This indicates that this type of protein performs an important but at present unrecognized function in biomineralization. Perlustrin was a minor component of the protein mixture and the sequence of the first 33 amino acids indicated a certain similarity to part of the much larger nacre protein lustrin A. 相似文献

Biochemical purifications from mammalian cells and Xenopus oocytes revealed that vertebrate Mi-2 proteins reside in multisubunit NuRD (Nucleosome Remodeling and Deacetylase) complexes. Since all NuRD subunits are highly conserved in the genomes of C. elegans and Drosophila, it was suggested that NuRD complexes also exist in invertebrates. Recently, a novel dMec complex, composed of dMi-2 and dMEP-1 was identified in Drosophila. The genome of C. elegans encodes two highly homologous Mi-2 orthologues, LET-418 and CHD-3. Here we demonstrate that these proteins define at least three different protein complexes, two distinct NuRD complexes and one MEC complex. The two canonical NuRD complexes share the same core subunits HDA-1/HDAC, LIN-53/RbAp and LIN-40/MTA, but differ in their Mi-2 orthologues LET-418 or CHD-3. LET-418 but not CHD-3, interacts with the Krüppel-like protein MEP-1 in a distinct complex, the MEC complex. Based on microarrays analyses, we propose that MEC constitutes an important LET-418 containing regulatory complex during C. elegans embryonic and early larval development. It is required for the repression of germline potential in somatic cells and acts when blastomeres are still dividing and differentiating. The two NuRD complexes may not be important for the early development, but may act later during postembryonic development. Altogether, our data suggest a considerable complexity in the composition, the developmental function and the tissue-specificity of the different C. elegans Mi-2 complexes. 相似文献

Alternariol (AOH) was reported recently to act as a topoisomerase poison. To underline the relevance of topoisomerase targeting for the genotoxic properties of AOH, we addressed the question whether human tyrosyl-DNA phosphodiesterase 1 (TDP1), an enzyme vital to the repair of covalent DNA-topoisomerase adducts, affects AOH-mediated genotoxicity. The relevance of TDP1 activity on AOH-induced genotoxicity was investigated by the comet assay in human cells overexpressing GFP chimera of TDP1 or the inactive mutant TDP1H263A as well as in cells subjected to siRNA-mediated knock-down of endogenous TDP1. Cells overexpressing TDP1 exhibited significantly less DNA damage after treatment with AOH in comparison to cells expressing the inactive mutant TDP1H263A. In accordance with these results, levels of AOH inducing DNA strand breaks were increased in TDP1-suppressed cells in comparison to cells transfected with control siRNA. The specific topoisomerase poisons camptothecin and etoposide caused comparable effects, underlining that TDP1 plays an important role in the repair of topoisomerase-mediated DNA damage. In summary, the repair enzyme TDP1 was identified as a factor for the modulation of AOH-mediated DNA damage in human cells. 相似文献

The RecX protein is a potent inhibitor of RecA protein activities. RecX functions by specifically blocking the extension of RecA filaments. In vitro, this leads to a net disassembly of RecA protein from circular single-stranded DNA. Based on multiple observations, we propose that RecX has a RecA filament capping activity. This activity has predictable effects on the formation and disassembly of RecA filaments. In vivo, the RecX protein may limit the length of RecA filaments formed during recombinational DNA repair and other activities. RecX protein interacts directly with RecA protein, but appears to interact in a functionally significant manner only with RecA filaments bound to DNA. 相似文献

Question: Can recent satellite imagery of coarse spatial resolution support forest cover assessment and mapping at the regional level? Location: Continental southeast Asia. Methods: Forest cover mapping was based on digital classification of SPOT4‐VEGETATION satellite images of 1 km spatial resolution from the dry seasons 1998/1999 and 1999/2000. Following a geographical stratification, the spectral clusters were visually assigned to land cover classes. The forest classes were validated by an independent set of maps, derived from interpretation of satellite imagery of high spatial resolution (Landsat TM, 30 m). Forest area estimates from the regional forest cover map were compared to the forest figures of the FAO database. Results: The regional forest cover map displays 12 forest and land cover classes. The mapping of the region's deciduous and fragmented forest cover remained challenging. A high correlation was found between forest area estimates obtained from this map and from the Landsat TM derived maps. The regional and sub‐regional forest area estimates were close to those reported by FAO. Conclusion: SPOT4‐VEGETATION satellite imagery can be used for mapping consistently and uniformly the extent and distribution of the broad forest cover types at the regional scale. The new map can be considered as an update and improvement on existing regional forest cover maps. 相似文献

EB1 proteins bind to microtubule ends where they act in concert with other components, including the adenomatous polyposis coli (APC) tumor suppressor, to regulate the microtubule filament system. We find that EB1 is a stable dimer with a parallel coiled coil and show that dimerization is essential for the formation of its C-terminal domain (EB1-C). The crystal structure of EB1-C reveals a highly conserved surface patch with a deep hydrophobic cavity at its center. EB1-C binds two copies of an APC-derived C-terminal peptide (C-APCp1) with equal 5 microM affinity. The conserved APC Ile2805-Pro2806 sequence motif serves as an anchor for the interaction of C-APCp1 with the hydrophobic cavity of EB1-C. Phosphorylation of the conserved Cdc2 site Ser2789-Lys2792 in C-APCp1 reduces binding four-fold, indicating that the interaction APC-EB1 is post-translationally regulated in cells. Our findings provide a basis for understanding the dynamic crosstalk of EB1 proteins with their molecular targets in eukaryotic organisms. 相似文献

Increasing the oleic to linoleic acid ratio (O/L) in peanut has positiveeffects on peanut quality and its nutritional value. 12-Fattyacid desaturases (12-Fad) have been targeted as logicalcandidates controlling the high oleate trait. A previous study using genomicDNA identified an insertion and a polymorphism resulting in an amino acid changeassociated with the high oleate trait in Spanish-type peanut cultivars. Theobjectives of this research were to use RT-PCR to confirm that the SingleNucleotide Polymorphims (SNPs) identified by analysis of genomic DNA wereexpressed, and to determine if expression patterns for 12-Fadwere the same in both seeds and leaves. A polymorphic region of the12-Fad containing a series of nucleotide changes wasamplified, cloned, and sequenced from mRNA of 155 clones of two parental linesand their independent derived backcross lines (IDBLs). The latter differed intheir oleic to linoleic ratio. Data indicated that the Ainsertion and the amino acid change were expressed in both leaf and seed tissue of thehigh and low-intermediate O/L genotypes. It is postulated that several copiesof the 12-Fad are present in the genome. It is reasonable toconclude that total activity, and ultimately the O/L ratio, is dependent on thenumber of functional copies. The results provide the basis for an assay toscreen for the high O/L ratio at the molecular level. We also report thepresence of another isozyme of 12-Fad with high homology tosoybean isozyme 2 that was expressed in seeds.

The grass snake (Natrix natrix) is Europe's most widely distributed and, in many regions, most common snake species, with many morphologically defined subspecies. Yet, the taxonomy of grass snakes is relatively little studied and recent work has shown major conflicts between morphologically defined subspecies and phylogeographical differentiation. Using external morphology, osteological characters, and information from 13 microsatellite loci and two mitochondrial markers, we examine differentiation of the subspecies N. n. astreptophora from the North African Maghreb region, the Iberian Peninsula and neighbouring France. According to previous studies, N. n. astreptophora corresponds to a deeply divergent mitochondrial clade and constitutes the sister taxon of all remaining grass snakes. In the French Pyrenees region, there is a contact zone of N. n. astreptophora with another subspecies, N. n. helvetica. Our analyses of microsatellites and mitochondrial DNA reveal that the distribution ranges of the two taxa abut there, but both hybridize only exceptionally. Even though many morphological characters are highly variable and homoplastic in grass snakes, N. n. astreptophora differs consistently from all other grass snakes by its reddish iris coloration and in having significantly fewer ventral scales and another skull morphology. Considering further the virtual absence of gene flow between N. n. astreptophora and N. n. helvetica, and acknowledging the morphological distinctiveness of N. n. astreptophora and its sister group relationship to all remaining subspecies of grass snakes, we conclude that Natrix astreptophora (Seoane, 1884) should be recognized as a distinct species. Further research is needed to explore whether N. astreptophora is polytypic because a single sample of N. astreptophora from Tunisia turned out to be genetically highly distinct from its European conspecifics. 相似文献
