Clonal populations are often characterized by reduced levels of genotypic diversity, which can translate into lower numbers of functional phenotypes, both of which impede adaptation. Study of partially clonal animals enables examination of the environmental settings under which clonal reproduction is favoured. Here, we gathered genotypic and phenotypic information from 3,651 georeferenced colonies of the fire coral Millepora platyphylla in five habitats with different hydrodynamic regimes in Moorea, French Polynesia. In the upper slope where waves break, most colonies grew as vertical sheets (“sheet tree”) making them more vulnerable to fragmentation. Nearly all fire corals in the other habitats are encrusting or massive. The M. platyphylla population is highly clonal (80% of the colonies are clones), while characterized by the highest genotype diversity ever documented for terrestrial or marine populations (1,064 genotypes). The proportion of clones varies greatly among habitats (≥58%–97%) and clones (328 clonal lineages) are distributed perpendicularly from the reef crest, perfectly aligned with wave energy. There are six clonal lineages with clones dispersed in at least two adjacent habitats that strongly demonstrate phenotypic plasticity. Eighty per cent of the colonies in these lineages are “sheet tree” on the upper slope, while 80%–100% are encrusting or massive on the mid slope and back reef. This is a unique example of phenotypic plasticity among reef‐building coral clones as corals typically have wave‐tolerant growth forms in high‐energy reef areas.  相似文献

Genetic diversity is crucial for species’ maintenance and persistence, yet is often overlooked in conservation studies. Species diversity is more often reported due to practical constraints, but it is unknown if these measures of diversity are correlated. In marine invertebrates, adults are often sessile or sedentary and populations exchange genes via dispersal of gametes and larvae. Species with a larval period are expected to have more connected populations than those without larval dispersal. We assessed the relationship between measures of species and genetic diversity, and between dispersal ability and connectivity. We compiled data on genetic patterns and life history traits in nine species across five phyla. Sampling sites spanned 600 km in the northwest Mediterranean Sea and focused on a 50‐km area near Marseilles, France. Comparative population genetic approaches yielded three main results. (i) Species without larvae showed higher levels of genetic structure than species with free‐living larvae, but the role of larval type (lecithotrophic or planktotrophic) was negligible. (ii) A narrow area around Marseilles, subject to offshore advection, limited genetic connectivity in most species. (iii) We identified sites with significant positive contributions to overall genetic diversity across all species, corresponding with areas near low human population densities. In contrast, high levels of human activity corresponded with a negative contribution to overall genetic diversity. Genetic diversity within species was positively and significantly linearly related to local species diversity. Our study suggests that local contribution to overall genetic diversity should be taken into account for future conservation strategies.  相似文献

The exceptional ability of marine sponges to adapt to often drastic changes of their environments could be due to special structural features in cell membranes, including firstly phospholipids (PL). Thus, PL class composition was investigated in marine sponges (22 species from 19 genera to 15 families) originating from various locations (East Atlantic, North Atlantic, South-West Pacific, Mediterranean Sea, Red Sea, Arabian–Persian Gulf). The quantitative determination of PL class composition was obtained by high-performance thin-layer chromatography (HPTLC) with scanning densitometry of the different spots. Previous reports have shown phosphatidylethanolamine (PE) as the major PL class in marine sponges, followed by phosphatidylcholine (PC), while other papers described PC as a minor class and even lacking. This survey found PE as the major PL class in only two species, while PC was the major class in 13 species including a calcareous one. The great abundance of bacteria in some sponges was evidenced from the relatively high proportions of particular PL classes. Various PL distributions were observed even for the sponge species collected in the same area and belonging to the same genus. Thus, no clear rule on PL composition in marine sponges can be stated to date.  相似文献

Flow injection electrospray-mass spectrometry (FIE-MS) is finding utility as a first-pass metabolite fingerprinting tool in many research fields. We provide a protocol that has proved reliable in large-scale research projects involving diverse sample matrices originating from plants, microbes and mammalian biofluids. Using Brachypodium leaf material as an example matrix all steps are summarized from sample extraction to data quality assessment. Alternative procedures for dealing with other common matrices such as bloods and urine are included. With little sample pretreatment, no chromatography and instrument cycle times of <5 min it is feasible to analyze >1,000 samples per week. Analysis of a typical batch of 240 samples (including first-pass data analysis) can be accomplished within 48 h.  相似文献

Objective: Mitochondrial activity is altered in skeletal muscle of obese, insulin‐resistant or type 2 diabetic patients. We hypothesized that this situation was associated with profound adaptations in resting muscle energetics. For that purpose, we used in vivo 31P‐nuclear magnetic resonance (31P‐NMR) in male sedentary Wistar rats fed with obesogenic diets known to induce alterations in muscle mitochondrial activity. Methods and Procedures: Two experimental diets (high sucrose and high fat) were provided for 6 weeks at two levels of energy (standard, N and high, H) and compared to control diet. The rates of the adenosine triphosphate (ATP) exchange between phosphocreatine (PCr) and γ ‐ATP (ka) and β ‐adenosine diphosphate ( β ‐ADP) to β ‐ATP (kb) were evaluated using 31P‐NMR in resting gastrocnemius muscle. Muscle contents in phosphorylated compounds as well as creatine, were assessed using 31P‐NMR and biochemical assays, respectively. Results: ATP content increased by 6.7–8.5% in standard‐energy high‐sucrose (NSU), high‐energy high‐fat (HF) and high‐energy high‐sucrose (HSU) groups compared to control (P < 0.05), whereas PCr content decreased by 4.2–6.4% (P < 0.01). Consequently, PCr to ATP ratio decreased in NSU, HF, and HSU groups, compared to control (P < 0.01). Furthermore in high‐energy groups (HF and HSU) compared to control, creatine contents were decreased by 14–19% (P < 0.001), whereas ka and kb fluxes were increased by 89–133% (P < 0.001) and 243–277% (P < 0.01), respectively. Discussion: Our in vivo data showed adaptations of resting skeletal muscle energetics in response to high‐energy diets. Increased activity of enzymes catalyzing ATP production may reflect a compensatory mechanism to face impaired mitochondrial ATP synthesis in order to preserve intracellular energy homeostasis.  相似文献

Experiments on Cd desorption were conducted with a range of water-to-soil ratios to assess the desorption characteristics of Cd in soils and the availability of Cd for absorption by plant roots and leaching to groundwater, Soil samples were collected from sites contaminated by a former Pb and Zn smelter, by sewage irrigation, or with artificial additions of Cd and sewage sludge. Glasshouse pot experiments were conducted in which the yield and Cd uptake of crop plants were determined. Cadmium leached from soil columns was also studied using soil lysimeters. The soil solution Cd concentration decreased with increasing solution-to-soil ratio and followed a negative power function. Two constants obtained from logarithmic linear regression were identified. The intercept (C1) was Cd concentration in the soil solution where the solution/soil ratio was equal to 1 and this constant was the intensity factor of the initial element supply in the soil. The slope (a) showed a decreasing trend for Cd concentration in the soil solution which was related to the soil buffering capacity. A corrected concentration (C1/a) is proposed for expressing soil desorption ability. This combined index was significantly correlated with Cd uptake by plants and also with Cd leached from soil columns.  相似文献

Between 1952 and 1963, the Abbé Glory conducted excavation in the Lascaux Cave. He discovered many artefacts useful for the Prehistoric painter. Today this material is kept in the “Institut de Paléontologie Humaine” in Paris. Some of this material, as well as the painting matter of the “Blazons” of the Black Cow Panel of the Nef, were examined and analysed by electron microscopy: SEM and TEM. Yellow, ocher and red pigments are iron oxides and black pigments are manganese oxides. The results indicate that the pigments were by no means heated, nor associated with any extender. Nevertheless the variety of the raw matter indicates a real strategy to exploit nature in order to match the desired hue and the texture of the painted panel.  相似文献
