Beitrag zur Kenntnis der chemischen Zusammensetzung derPhragmiti-Magnocaricetea und derAgropyro-Rumicion-Arten

Zwanzig Pflanzen der KlassePhragmiti-Magnocaricetea (Kennarten der einzelnen pflanzen-soziologischen Einheiten) und des VerbandesAgropyro-Rumicion crispi wurden den chemischen Analysen unterzogen. Die Pflanzenproben wurden auf den Wiesen der Trockengebiete S-Mährens und der SW-Slowakei gesammelt, u. zw. aus den pflanzensoziologisch, definierbaren Einheiten (Assoziationen der VerbändePhragmition, Caricion gracilis, Caricion rostratae undAgropyro-Rumicion). Die Resultate wurden mit der chemischen Zusammensetzung der in anderen Gebieten vorkommenden Pflanzen verglichen. Es zeigten sich Zusammenhänge zwischen der genetisch bedingten chemischen Zusammensetzung bestimmter Artengruppen und den Eigenschaften des Substrats. In diesem Sinne gibt es allgemeine Unterschiede zwischen den VerbändenCaricion gracilis undCaricion rostratae. Die Arten desAgropyro-Rumicion zeigen im Durchschnitt engere Bindung an die Gesellschaften desCnidion-Verbandes als an dasCaricion gracilis.     

Cytochemical and immunofluorescence investigations on insulin-like producing cells in the intestine of Mytilus edulis L. (Bivalvia)

Summary Insulin-like immunoreactivity can be localized to cells of the intestine in the-area of the hepatopancreas of Mytilus edulis L. No cross-reactivity can be obtained with anti-glucagon, anti-gastrin, anti-pentagastrin or anticaerulin. The cells containing the substance immunoreactive to mammalian anti-insulin, can be restained with paraldehyde-fuchsin.I thank Prof. A.G.E. The occurrence of argyrophilic and argentaffin cells in the gut of Ciona intestinalis L.

Summary Argyrophilic and argentaffin cells occur in the stomach and intestinal epithelium of the sea-squirt, Ciona intestinalis L.. These cells are characterized by their basal swelling which contains the nucleus surrounded by small secretory granules and by a filamentous cell-apex which reaches the gut lumen. The cells are scattered unevenly within the epithelium. Their number decreases rapidly towards the lower part of the intestine. The localization, size of granules and their shape are features which differentiate these cells from other secretory cells in the gut epithelium such as mucous cells. These cells are thought to possess an endocrine function.The excellent technical assistance of Mrs. R. Sprang is gratefully acknowledged     

Reconciling the biogeography of an invader through recent and historic genetic patterns: the case of topmouth gudgeon <Emphasis Type="Italic">Pseudorasbora parva</Emphasis>

The genetic variability and population structure of introduced species in their native range are potentially important determinants of their invasion success, yet data on native populations are often poorly represented in relevant studies. Consequently, to determine the contribution of genetic structuring in the native range of topmouth gudgeon Pseudorasbora parva to their high invasion success in Europe, we used a dataset comprising of 19 native and 11 non-native populations. A total of 666 samples were analysed at 9 polymorphic microsatellite loci and sequenced for 597 bp of mitochondrial DNA. The analysis revealed three distinct lineages in the native range, of which two haplogroups were prevalent in China (100%), with a general split around the Qinling Mountains. Dating of both haplogroups closely matched past geological events. More recently, its distribution has been influenced by fish movements in aquaculture, resulting in gene flow between previously separated populations in Northern and Southern China. Their phylogeography in Europe indicate as few as two introductions events and two dispersal routes. Microsatellite data revealed native populations had higher genetic diversity than those in the invasive range, a contrast to previous studies on P. parva. This study confirms the importance of extensive sampling in both the native and non-native range of invasive species in evaluating the influence of genetic variability on invasion success.     

Design and validation of a homogeneous time-resolved fluorescence cell-based assay targeting the ligand-gated ion channel 5-HT3A

Ligand-gated ion channels (LGICs) are considered as attractive protein targets in the search for new therapeutic agents. Nowadays, this strategy involves the capability to screen large chemical libraries. We present a new Tag-lite ligand binding assay targeting LGICs on living cells. This technology combines the use of suicide enzyme tags fused to channels of interest with homogeneous time-resolved fluorescence (HTRF) as the detection readout. Using the 5-HT3 receptor as system model, we showed that the pharmacology of the HALO-5HT3 receptor was identical to that of the native receptor. After validation of the assay by using 5-HT3 agonists and antagonists of reference, a pilot screen enabled us to identify azelastine, a well-known histamine H1 antagonist, as a potent 5-HT3 antagonist. This interesting result was confirmed with electrophysiological experiments. The method described here is easy to implement and could be applicable for other LGICs, opening new ways for the screening of chemical libraries.
