 We describe two modifications of the popular reporter green fluorescent protein (GFP) which have short half-lives in our system, the cellular slime mould Dictyostelium discoideum. One of these bears an N-terminal ubiquitin; this GFP was originally planned to be a substrate of the ”N-end-rule” pathway, but deubiquitination does not seem to occur, and a degradation by the UFD (ubiquitin-fusion-degradation pathway seems more probable. The protein half-life is about 3–5 h. The second construct has an N-terminus derived from the L11 ribosomal protein; it is transported to the nucleus and broken down much more rapidly than the ubiquitin fusion (protein half-life about 30 min). Nucleotide sequences from two nuclear loci, alcohol dehydrogenase and internal transcribed spacer-1 of the nuclear ribosomal DNA repeats, and two mitochondrial genes, cytochrome oxidase I and cytochrome oxidase II, were determined from nine species in the Drosophila saltans species group. The partition homogeneity test and partitioned Bremer support were used to measure incongruence between phylogenetic hypotheses generated from individual partitions. Individual loci were generally congruent with each other and consistent with the previously proposed morphological hypothesis, although they differed in level of resolution. Since extreme conflict between partitions did not exist, the data were combined and analyzed simultaneously. The total evidence method gave a more resolved and highly supported phylogeny, as indicated by bootstrap proportions and decay indices, than did any of the individual analyses. The cordata and elliptica subgroups, considered to have diverged early in the history of the D. saltans group, were sister taxa to the remainder of the saltans group. The sturtevanti subgroup, represented by D. milleri and D. sturtevanti, occupies an intermediate position in this phylogeny. The saltans and parasaltans subgroups are sister clades and occupy the most recently derived portion of the phylogeny. As with previous morphological studies, phylogenetic relationships within the saltans subgroup were not satisfactorily resolved by the molecular data.   相似文献

Biogas production is of major importance for the sustainable use of agrarian biomass as renewable energy source. Economic biogas production depends on high biogas yields. The project aimed at optimising anaerobic digestion of energy crops. The following aspects were investigated: suitability of different crop species and varieties, optimum time of harvesting, specific methane yield and methane yield per hectare. The experiments covered 7 maize, 2 winter wheat, 2 triticale varieties, 1 winter rye, and 2 sunflower varieties and 6 variants with permanent grassland. In the course of the vegetation period, biomass yield and biomass composition were measured. Anaerobic digestion was carried out in eudiometer batch digesters. The highest methane yields of 7500-10200 m(N)(3)ha(-1) were achieved from maize varieties with FAO numbers (value for the maturity of the maize) of 300 to 600 harvested at "wax ripeness". Methane yields of cereals ranged from 3200 to 4500 m(N)(3)ha(-1). Cereals should be harvested at "grain in the milk stage" to "grain in the dough stage". With sunflowers, methane yields between 2600 and 4550 m(N)(3)ha(-1) were achieved. There were distinct differences between the investigated sunflower varieties. Alpine grassland can yield 2700-3500 m(N)(3)CH(4)ha(-1). The methane energy value model (MEVM) was developed for the different energy crops. It estimates the specific methane yield from the nutrient composition of the energy crops. Energy crops for biogas production need to be grown in sustainable crop rotations. The paper outlines possibilities for optimising methane yield from versatile crop rotations that integrate the production of food, feed, raw materials and energy. These integrated crop rotations are highly efficient and can provide up to 320 million t COE which is 96% of the total energy demand of the road traffic of the EU-25 (the 25 Member States of the European Union).  相似文献

Morphogenesis of herpesviral virions is initiated in the nucleus but completed in the cytoplasm. Mature virions contain more than 25 tegument proteins many of which perform both nuclear and cytoplasmic functions suggesting they shuttle between these compartments. While nuclear import of herpesviral proteins was shown to be crucial for viral propagation, active nuclear export and its functional impact are still poorly understood. To systematically analyze nuclear export of tegument proteins present in virions of Herpes simplex virus type 1 (HSV1) and Epstein‐Barr virus (EBV), the Nuclear EXport Trapped by RAPamycin (NEX‐TRAP) was applied. Nine of the 22 investigated HSV1 tegument proteins including pUL4, pUL7, pUL11, pUL13, pUL21, pUL37d11, pUL47, pUL48 and pUS2 as well as 2 out of 6 EBV orthologs harbor nuclear export activity. A functional leucine‐rich nuclear export sequence (NES) recognized by the export factor CRM1/Xpo1 was identified in six of them. The comparison between experimental and bioinformatic data indicates that experimental validation of predicted NESs is required. Mutational analysis of the pUL48/VP16 NES revealed its importance for herpesviral propagation. Together our data suggest that nuclear export is an important feature of the herpesviral life cycle required to co‐ordinate nuclear and cytoplasmic processes.   相似文献

Mammalian cortical expansion is tightly controlled by fine‐tuning of proliferation and differentiation of neural progenitors in a region‐specific manner. How extrinsic cues interface with cell‐intrinsic programs to balance proliferative versus neurogenic decisions remains an unsolved question. We examined the function of Semaphorin receptors Plexin‐B1 and ‐B2 in corticogenesis by generating double mutants, whereby Plexin‐B2 was conditionally ablated in the developing brain in a Plexin‐B1 null mutant background. Absence of both Plexin‐Bs resulted in cortical thinning, particularly in the caudomedial cortex. Plexin‐B1/B2 double, but not single, mutants exhibited a reduced neural progenitor pool, attributable to decreased proliferation and an altered division mode favoring cell cycle exit. This resulted in deficient production of neurons throughout the neurogenic period, proportionally affecting all cortical laminae. Consistent with the in vivo data, cultured neural progenitors lacking both Plexin‐B1 and ‐B2 displayed decreased proliferative capacity and increased spontaneous differentiation. Our study therefore defines a novel function of Plexin‐B1 and ‐B2 in transmitting extrinsic signals to maintain proliferative and undifferentiated states of neural progenitors. As single mutants displayed no apparent cortical defects, we conclude that Plexin‐B1 and ‐B2 play redundant or compensatory roles during forebrain development to ensure proper neuronal production and neocortical expansion. © 2015 Wiley Periodicals, Inc. Some pesticides may interfere with the female hormonal function, which may lead to negative effects on the reproductive system through disruption of the hormonal balance necessary for proper functioning. Previous studies primarily focused on interference with the estrogen and/or androgen receptor, but the hormonal function may be disrupted in many more ways through pesticide exposure. The aim of this review is to give an overview of the various ways in which pesticides may disrupt the hormonal function of the female reproductive system and in particular the ovarian cycle. Disruption can occur in all stages of hormonal regulation: 1. hormone synthesis; 2. hormone release and storage; 3. hormone transport and clearance; 4. hormone receptor recognition and binding; 5. hormone postreceptor activation; 6. the thyroid function; and 7. the central nervous system. These mechanisms are described for effects of pesticide exposure in vitro and on experimental animals in vivo. For the latter, potential effects of endocrine disrupting pesticides on the female reproductive system, i.e. modulation of hormone concentrations, ovarian cycle irregularities, and impaired fertility, are also reviewed. In epidemiological studies, exposure to pesticides has been associated with menstrual cycle disturbances, reduced fertility, prolonged time-to-pregnancy, spontaneous abortion, stillbirths, and developmental defects, which may or may not be due to disruption of the female hormonal function. Because pesticides comprise a large number of distinct substances with dissimilar structures and diverse toxicity, it is most likely that several of the above-mentioned mechanisms are involved in the pathophysiological pathways explaining the role of pesticide exposure in ovarian cycle disturbances, ultimately leading to fertility problems and other reproductive effects. In future research, information on the ways in which pesticides may disrupt the hormonal function as described in this review, can be used to generate specific hypotheses for studies on the effects of pesticides on the ovarian cycle, both in toxicological and epidemiological settings.  相似文献

Changes in the protein content of and the rate at which labelled protein appears in the accessory reproductive glands (ARG), fat body, and haemolymph were studied in normal and allatectomized (CA?) males of the migratory grasshopper, Melanoplus sanguinipes. In addition, the effects of treatment of CA? insects with synthetic juvenile hormone (SJH), copulation, and removal of the ARG were examined.In normal males the protein content of the ARG increases linearly during the first 14 days after emergence. Incorporation of label by the ARG is maximal at day 7 and then decreases until, at day 14, it is the same as at day 1. The protein content of the fat body and haemolymph increases up to day 10 then declines, whereas changes in the uptake of label by the fat body and haemolymph parallel those of the ARG.Removal of the corpora allata (CA) prevents the normal increase in protein content of the ARG, but the protein content of the fat body and haemolymph increases steadily throughout the 14 days. Incorporation of label into the ARG, fat body, and haemolymph remained low throughout the experiment. Treatment of CA? insects with SJH, or copulation, stimulates the uptake of label by the ARG, fat body, and haemolymph and also results in an increase in their protein content.Removal of the ARG leads to an increase in the protein content of the fat body and haemolymph. Uptake of label by the fat body remains low after the operation. Although the rate at which labelled protein appears in the haemolymph is high initially, it declines steadily to day 14.We conclude that the CA regulate ARG development. It is suggested that the fat body, under CA control, synthesizes proteins which are incorporated into secretions of the ARG. Further, it is proposed that the primary effect of copulation is activation of the CA.  相似文献

Ecologists have long been concerned that contemporary fire regimes of central Australia have poor consequences for some plant species, vegetation communities and the native animals they support. Fire frequency, size and intensity (the ‘fire regime’) have all been implicated in the decline of native biota and in vegetation changes that potentially constitute ecological drift. However, not all perceived declines and changes are quantified or proven. The fire regimes themselves defy quantification and are arguably unknowable. We examine the relationships between fire, vegetation and the physical landscape and consider the adequacy of available knowledge for guiding fire management. Devising targeted ‘fire management regimes’, which take into account vegetation type and management objectives such as pastoral production, conservation and cultural observance, and which actively use fire to achieve those objectives, is a more realistic goal than controlling unquantifiable fire regimes in spatially diverse landscapes.  相似文献

Effectiveness of natural stimuli (recorded and replayed male vibratory courtship signals) in eliciting a female's vibratory response was compared with that of synthetic stimuli in a plant-dwelling spider (Cupiennius salei). Specifically, the role of interseries (pauses between consecutive male series) was evaluated. Effectiveness was assessed by the number of responding females and the number of responses per female per stimulus. In case of both synthetic and natural stimuli, effectiveness increases with increasing duration of interseries in both measures. Interseries are thus represented in the female's innate releasing mechanism. However, synthetic stimuli are less effective (about 55%) than natural stimuli (set 100%). With decreasing interseries, the number of female responses per unit time sharply increases in case of playback of natural stimuli, but only slightly in case of synthetic stimuli. Since males courting on plants decrease interseries as soon as a female responds, thus facilitating their orientation, the data obtained with natural stimuli much better fit natural male courtship behaviour. This finding brings into question the validity of the synthetic stimulus. In the bioacoustics literature there is neither consensus on the effectiveness of a synthetic stimulus necessary to qualify it for behavioural analysis nor on how to measure effectiveness. We propose three measures : the percentage of responding animals, the action pattern of the response and the number of responses. It may be crucial, particularly when addressing evolutionary questions, to use either natural stimuli or synthetic stimuli (nearly) as effective as natural stimuli to avoid biased sampling of experimental animals and results difficult to interpret in biological terms.  相似文献
