Droughts and forest fires, induced by the El Niño/Southern Oscillation (ENSO) event, have increased considerably over the last decades affecting millions of hectares of rainforest. We investigated the effects of the 1997–1998 forest fires and drought, associated with an exceptionally severe ENSO event, on fruit species important in the diet of Malayan sun bears (Helarctos malayanus) in lowland dipterocarp forest, East Kalimantan, Indonesian Borneo. Densities of sun bear fruit trees (≥10 cm DBH) were reduced by ~80%, from 167±41 (SD) fruit trees ha?1 in unburned forest to 37±18 fruit trees ha?1 in burned forest. Densities of hemi-epiphytic figs, one of the main fallback resources for sun bears during periods of food scarcity, declined by 95% in burned forest. Species diversity of sun bear food trees decreased by 44% in burned forest. Drought also affected sun bear fruit trees in unburned primary forest, with elevated mortality rates for the duration of 2 years, returning to levels reported as normal in region in the third year after the ENSO event. Mortality in unburned forest near the burn-edge was higher (25±5% of trees ≥10 cm DBH dead) than in the forest interior (14±5% of trees), indicating possible edge effects. Combined effects of fire and drought in burned primary forest resulted in an overall tree mortality of 78±11% (≥10 cm DBH) 33 months after the fire event. Disturbance due to fires has resulted in a serious decline of fruit resources for sun bears and, due to the scale of fire damage, in a serious decline of prime sun bear habitat. Recovery of sun bear populations in these burned-over forests will depend on regeneration of the forest, its future species composition, and efforts to prevent subsequent fire events.  相似文献

Chemotactic motility has previously been shown to be essential for the virulence of Vibrio anguillarum in waterborne infections of fish. To investigate the mechanisms by which chemotaxis may function during infection, mucus was isolated from the intestinal and skin epithelial surfaces of rainbow trout. Chemotaxis assays revealed that V. anguillarum swims towards both types of mucus, with a higher chemotactic response being observed for intestinal mucus. Work was performed to examine the basis, in terms of mucus composition, of this chemotactic response. Intestinal mucus was analyzed by using chromatographic and mass spectrometric techniques, and the compounds identified were tested in a chemotaxis assay to determine the attractants present. A number of mucus-associated components, in particular, amino acids and carbohydrates, acted as chemoattractants for V. anguillarum. Importantly, only upon combination of these attractants into a single mixture were levels of chemotactic activity similar to those of intestinal mucus generated. A comparative analysis of skin mucus revealed its free amino acid and carbohydrate content to be considerably lower than that of the more chemotactically active intestinal mucus. To study whether host specificity exists in relation to vibrio chemotaxis towards mucus, comparisons with a human Vibrio pathogen were made. A cheR mutant of a Vibrio cholerae El Tor strain was constructed, and it was found that V. cholerae and V. anguillarum exhibit a chemotactic response to mucus from several animal sources in addition to that from the human jejunum and fish epithelium, respectively.  相似文献

The biodistribution of 11C-labeled 4-(3-bromoanilino)-6,7-dimethoxyquinazoline, an inhibitor of the epidermal growth factor (EGF) receptor tyrosine kinase, has been evaluated in vivo in rats using positron emission tomography (PET). Time-activity data obtained after i.v. administration in one rat revealed that the radiotracer rapidly cleared from plasma with subsequent uptake in major organs of the body (brain, heart, liver, gastrointestinal tract and bladder). Uptake in proliferating tissue in rats with human neuroblastoma xenografts indicate that [O-11C-methyl]PD153035 shows promise as a new agent for in vivo imaging of tumors with PET.  相似文献

A high throughput protein biomarker discovery tool has been developed based on multiplexed proximity ligation assays in a homogeneous format in the sense of no washing steps. The platform consists of four 24-plex panels profiling 74 putative biomarkers with sub-pm sensitivity each consuming only 1 μl of human plasma sample. The system uses either matched monoclonal antibody pairs or the more readily available single batches of affinity purified polyclonal antibodies to generate the target specific reagents by covalently linking with unique nucleic acid sequences. These paired sequences are united by DNA ligation upon simultaneous target binding forming a PCR amplicon. Multiplex proximity ligation assays thereby converts multiple target analytes into real-time PCR amplicons that are individually quantified using microfluidic high capacity qPCR in nano liter volumes. The assay shows excellent specificity, even in multiplex, by its dual recognition feature, its proximity requirement, and most importantly by using unique sequence specific reporter fragments on both antibody-based probes. To illustrate the potential of this protein detection technology, a pilot biomarker research project was performed using biobanked plasma samples for the detection of colorectal cancer using a multivariate signature.  相似文献

Intravenous administration of endotoxin to mares causes synthesis and release of PGF(2alpha); this process affects corpus luteum function, either causing a transient drop in progesterone levels or complete luteolysis with shortening of the estrous cycle. In contrast to the African great apes, orangutans (Pongo spp.) are semisolitary: Individuals are often on their own, but form aggregations more often than expected by chance. These temporary aggregations provide social benefits such as mating opportunities. When fruit availability is high, costs of aggregating should be lower, because competition is less pronounced. Therefore, average party size is expected to be higher when fruit availability is high. This hypothesis would also explain why orangutans in highly fruit-productive habitats on Sumatra are more gregarious than in the usually less productive habitats of Borneo. Here, we describe the aggregation behavior of orangutans in less productive Sumatran habitats (Sikundur and Batang Toru), and compare results with those of previously surveyed field sites. Orangutans in Sikundur were more likely to form parties when fruit availability was higher, but the size of daily parties was not significantly affected by fruit availability. With regard to between-site comparisons, average party sizes of females and alone time of parous females in Sikundur and Batang Toru were substantially lower than those for two previously surveyed Sumatran sites, and both fall in the range of values for Bornean sites. Our results indicate that the assessment of orangutans on Sumatra as being more social than those on Borneo needs revision. Instead, between-site differences in sociality seem to reflect differences in average fruit availability.  相似文献

The pH dependence of the steady-state kinetic parameters of cytochrome oxidase has been determined in the pH range 5.4-8.4 with the enzyme in detergent solution at high ionic strength. The catalytic constant increases continuously with decreasing pH, whereas the specificity constant for reduced cytochrome c is essentially unchanged. The effect of pH on the aerobic transient kinetics has also been investigated in two types of experiments in a stopped-flow apparatus. In one series, a 20-fold molar excess of reduced cytochrome c was the only reducing substrate, whereas in the other an excess of ascorbate was used together with a mediator and varying concentrations of cytochrome c. In both sets the time course of the reduction levels of cytochrome c, cytochrome a, and CuA was monitored at specific wavelengths. In the first type of experiment, the reoxidation of cytochrome a was slower than cytochrome c oxidation. In the second type, four kinetic phases were observed, including a long steady state. The time courses, including these features, were simulated on the basis of a mechanistic model for cytochrome oxidase as a proton pump. In this model the enzyme exists in two conformations, E1 and E2. The intramolecular electron transfer from the primary electron acceptors to the dioxygen-reducing site is rapid in E2 only. The transition from E1 to E2 is triggered by the reduction of cytochrome a and CuA. Bacteria associated with seaweeds can both harm and benefit their hosts. Many seaweed species are known to produce compounds that inhibit growth of bacterial isolates, but the ecological role of seaweed metabolites for the associated bacterial community structure is not well understood. In this study the response of a colonizing bacterial community to the secondary metabolite (1,1,3,3-tetrabromo-2-heptanone) from the red alga Bonnemaisonia hamifera was investigated by using field panels coated with the metabolite at a range of concentrations covering those measured at the algal surface. The seaweed metabolite has previously been shown to have antibacterial effects. The metabolite significantly affected the natural fouling community by (i) altering the composition, (ii) altering the diversity by increasing the evenness and (iii) decreasing the density, as measured by terminal restriction fragment length polymorphism in conjunction with clone libraries of the 16S rRNA genes and by bacterial enumeration. No single major bacterial taxon (phylum, class) was particularly affected by the metabolite. Instead changes in community composition were observed at a more detailed phylogenetic level. This indicates a broad specificity of the seaweed metabolite against bacterial colonization, which is supported by the observation that the bacterial density was significantly affected at a lower concentration (0.02 μg cm?2) than the composition (1-2.5 μg cm?2) and the evenness (5 μg cm?2) of the bacterial communities. Altogether, the results emphasize the role of secondary metabolites for control of the density and structure of seaweed-associated bacterial communities.  相似文献
