The cell surface ofIsotricha intestinalis was composed of predominantly longitudinal cytoplasmic ridges between the rows of cilia. On the dorsilateral surface these ridges were modified into sheet-like cytoplasmic processes extending up to 9 m from the cell surface, and up to 35 m long, forming a ridge projecting from the cell surface. The ridge was visible in living cells examined under phase contrast microscopy, and became more prominent after fixation with glutaraldehyde. This region of the cell surface was used for the attachment of the cell to surfaces. The cytoplasmic processes coalesced and bifurcated at points distal from the cell surface; underlying the plasma membrane of these processes was an extensive sheet of microtubules. Fixation of attached cells normally resulted in a separation of the cells from the substratum.  相似文献

Dizene dicarboxylic acid bis-(N,N-dimethylamide), commonly called diamide, is known to oxidize stoichiometrically intracellular pools of reduced glutathione and inhibit the accumulation of sugars and amino acids by rat kidney slices. Incubation of rat renal cortical slices in diamide also leads to a significant decrease in the level of endogenous protein kinase activity. The inhibition of sugar and amino acid transport and protein kinase activity by diamide is partially reversible by the addition of exogenous glutathione or other thiols. A comparison of protein kinase activity with amino acid and sugar transport at various concentrations of diamide indicates that there is a high degree of correlation between these two processes.  相似文献

The primer-independent phosphorylase isozyme, a2, of Cyanidiumcaldarium was used for immunization of rabbits. The immune serumwas tested against pure a2 isozymes from blue-green, red, andgreen algae. Double immunodiffusion in agar indicated that therewas structural similarity in the isozyme from Cyanidium caldarium,the blue-green algae, Oscillaloria princeps, Pleclonema nostocorumand the red alga, Rhodymenia pertusa. Complete fusion of theprecipitin lines was obtained with these algae. However, onlypartial fusion was observed with the a2 isozymes from Chlorophyceaesuch as Chlorella pyrenoidosa and Spirogyra setiformis. Spurformation on the precipitin lines occurred when the isozymesfrom these algae were tested against the immuneserum. The results were interpreted as indicative of the possible transitionstatus of Cyanidium caldarium between prokaryotic blue-greenalgae and eukaryotic red algae. Kinetic studies of the primer-independent, aa phosphorylaseisozyme of two thermophilic algae, Cyanidium caldarium and Oscillatoriaprinceps, and two mesophilic algae, Spirogyra setiformis andChlorella pyrenoidosa, indicate that the isozyme of thermophilicspecies is more resistant to denaturation by heat, and has lessaffinity for the glucose-1-phosphate substrate than the sameisozyme of mesophilic algae. This points out the possibilitythat this enzyme's structure is more rigid in thermophiles andmay lack the flexible conformation found in the mesophilic isozyme.The evolutionary significance of thermophily as a primitivecharacter rather than an adaptive trait is discussed in thelight of studies with a low temperature variant strain of thethermophilic alga, Oscillatoria princeps, LTV. An alkaline endoribonuclease was purified 1800-fold from the cytosolic, latent ribonuclease fraction of porcine thyroids by gentle procedures specifically designed to exclude both heating and acidification steps. Polyacrylamide gel electrophoresis revealed a broad peak of enzyme activity that was coincident with the stained protein band. As estimated by gel filtration chromatography the major form of the enzyme (59%) had a molecular weight of 51,000; the remainder of the activity was distributed among six minor forms. Carboxymethyl-cellulose chromatography showed that the enzyme had at least three interconvertible forms. The latent alkaline ribonuclease had a pH optimum of 8.1 in both Tris and 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid buffers and was stimulated by a number of monovalent chloride and potassium salts at ionic strengths between 10 and 70 mm; above 100 mm the salts were all inhibitory with the exception of ammonium chloride. At 1 mm both MgCl2 and CaCl2 were stimulatory, whereas CuCl2 ZnCl2 and EDTA were inhibitors. Both native and denatured DNA were slightly stimulatory. The porcine thyroid latent alkaline ribonuclease was specific for pyrimidine homopolymers and yielded a mixture of cyclic mononucleotides and oligonucleotides when incubated with poly(C). It did not hydrolyze 2′(3′)-cyclic CMP, purine homopolymers, native or denatured DNA or poly(A) · poly(U). Its activity toward rRNA was greater than toward tRNA and it cleaved the former to a mixture of mononucleotides and oligonucleotides. The properties of the intracellular, cytosolic, latent, alkaline ribonuclease distinguish it from pancreatic ribonuclease A and other nonsecretory ribonucleases.  相似文献

Synopsis Production rates of seven species of fishes were determined for one year in a 0.95 ha pond. Total annual production of all fishes in the pond was 209 g m–2y–1 (after the subtraction of 16.2 g m–2y–1 of negative production). Over 90% of the production occurred in the months of June through September, with young-of-the-year fishes contributing 84% of the total. When Y of Y fishes shifted to benthic diets in the spring, the small biomass of benthos available could not sustain the large biomas of fish. Severe competition for the available food then resulted in decreased production rates for the entire fish taxocene.  相似文献

Conventionally, local seed sources are normally believed to perform better than introduced materials. However, studies show possibility of contrary results with many species such as Acacia senegal. The species is an economically and ecologically important tree of arid and semi‐arid lands of sub‐Saharan Africa. It produces gum arabic, used in land reclamation and agroforestry production. The species is however underutilized in Kenya due to lack of information on growth performance of different seed sources. Glasshouse provenance trial using seeds and soils from seven provenances in Kenya were used to evaluate interactions between seed sources and soils on growth and biomass. Seedling growth was assessed for 12 weeks in a randomized complete block design. Seed length, width, thickness and weight were measured. Seedlings height, root collar diameter, root dry weight, shoot dry weight and biomass were assessed and data subjected to univariate and multivariate analyses. No significant interaction between seed provenance and soils were evident; however, some provenances performed better across all the soils. Significant heritability and relationship between growth and environmental factors are reported. Overall, Ntumburi and Ngarendare provenances showed superior growth and plasticity. These provenances can be used tentatively as seed sources; however, field trials are recommended.  相似文献

In bacteria, initiation of translation is kinetically controlled by factors IF1, IF2, and IF3, which work in conjunction with the 30S subunit to ensure accurate selection of the initiator tRNA (fMet-tRNA(fMet)) and the start codon. Here, we show that mutations G1338A and A790G of 16S rRNA decrease initiation fidelity in vivo and do so in distinct ways. Mutation G1338A increases the affinity of tRNA(fMet) for the 30S subunit, suggesting that G1338 normally forms a suboptimal Type II interaction with fMet-tRNA(fMet). By stabilizing fMet-tRNA(fMet) in the preinitiation complex, G1338A may partially compensate for mismatches in the codon-anti-codon helix and thereby increase spurious initiation. Unlike G1338A, A790G decreases the affinity of IF3 for the 30S subunit. This may indirectly stabilize fMet-tRNA(fMet) in the preinitiation complex and/or promote premature docking of the 50S subunit, resulting in increased levels of spurious initiation.  相似文献
