Experimental data which establish a correlation between applied compressive stress in a rabbit calvarium and calcium formation, collagen synthesis, and histologic interpretation are presented. Estimates of collagen synthesis and calcium formation are obtained from measuring 45Ca and 3H proline uptakes. Stress distributions are obtained using a finite element analysis and photoelastic techniques. For the cases presented here, maximum 45Ca and 3H proline uptakes, as well as maximal histologic healing indices, are found at stress levels in the 10–30 psi range.  相似文献

Life-history theory predicts that, in long-lived organisms, effort towards reproduction will increase with age, and research from oviparous vertebrates largely supports this prediction. In reptiles, where parental care occurs primarily via provisioning of the egg, older females tend to produce larger eggs, which in turn produce larger hatchlings that have increased survival. We conducted an experimental release study and report that maternal age positively influences offspring survivorship in the painted turtle (Chrysemys picta) and predicts offspring survival at least as well as hatchling body size does. These data suggest that, although increasing hatchling size is a major component of reproductive success in older individuals, other factors also contribute.  相似文献

BACKGROUND: This field-based investigation examined the congruence between skinfolds and bioelectrical impedance in assessing body composition in children. METHODS: Subjects were 162 female and 160 male children 10-15 years of age. Skinfold measures obtained at the triceps and medial calf and a leg-to-leg bioelectrical impedance system were used to determine percent fat using child-specific equations. Pearson product moment correlations were performed on the percent fat values obtained using skinfolds and bioelectric impedance for the entire data set. Separate correlations were also conducted on gender and age/gender subsets. Dependent t tests were used to compare the two techniques. RESULTS: Percent fat did not differ between skinfolds and bioelectrical impedance for the total subject pool. Bioelectrical impedance overestimated percent fat in girls by 2.6% and underestimated percent fat in boys by 1.7% (p < 0.01). Correlations between skinfolds and bioelectrical impedance ranged from r = 0.51 to r = 0.90. CONCLUSIONS: Leg-to-leg bioelectrical impedance may be a viable alternative field assessment technique that is comparable to skinfolds. The small differences in percent fat between the two techniques may have limited practical significance in school-based health-fitness settings.  相似文献

Hormone-sensitive lipase (HSL) is rate limiting for diacylglycerol and cholesteryl ester hydrolysis in adipose tissue and essential for complete hormone-stimulated lipolysis. Gene expression profiling in HSL-/- mice suggests that HSL is important for modulating adipogenesis and adipose metabolism. To test whether HSL is required for the supply of intrinsic ligands for PPARγ for normal adipose differentiation, HSL-/- and wild-type (WT) littermates were fed normal chow (NC) and high-fat (HF) diets supplemented with or without rosiglitazone (200 mg/kg) for 16 weeks. Results show that supplementing rosiglitazone to an NC diet completely normalized the decreased body weight and adipose depots in HSL-/- mice. Additionally, rosiglitazone resulted in similar serum glucose, total cholesterol, FFA, and adiponectin values in WT and HSL-/- mice. Furthermore, rosiglitazone normalized the expression of genes involved in adipocyte differentiation, markers of adipocyte differentiation, and enzymes involved in triacylglycerol synthesis and metabolism, and cholesteryl ester homeostasis, in HSL-/- mice. Supplementing rosiglitazone to an HF diet resulted in improved glucose tolerance in both WT and HSL-/- animals and also partial normalization in HSL-/- mice of abnormal WAT gene expression, serum chemistries, organ and body weight changes. In vitro studies showed that adipocytes from WT animals can provide ligands for activation of PPARγ and that activation is further boosted following lipolytic stimulation, whereas adipocytes from HSL-/- mice displayed attenuated activation of PPARγ, with no change following lipolytic stimulation. These results suggest that one of the mechanisms by which HSL modulates adipose metabolism is by providing intrinsic ligands or pro-ligands for PPARγ.  相似文献

Previous in vitro studies have established that hormone sensitive lipase (HSL) and adipocyte fatty acid-binding protein (AFABP) form a physical complex that presumably positions the FABP to accept a product fatty acid generated during catalysis. To assess AFABP-HSL interaction within a cellular context, we have used lipocytes derived from 293 cells (C8PA cells) and examined physical association using fluorescence resonance energy transfer. Transfection of C8PA cells with cyan fluorescent protein (CFP)-HSL, yellow fluorescent protein (YFP)-adipocyte FABP, or YFP-liver FABP revealed that under basal conditions each protein was cytoplasmic. In the presence of 20 microm forskolin, CFP-HSL translocated to the triacylglycerol droplet, coincident with BODIPY-FA labeled depots. Fluorescence resonance energy transfer analysis demonstrated that CFP-HSL associated with YFP-adipocyte FABP in both basal and forskolin-treated cells. In contrast, little if any fluorescence resonance energy transfer could be detected between CFP-HSL and YFP-liver FABP. These results suggest that a pre-lipolysis complex containing at least AFABP and HSL exists and that the complex translocates to the surface of the lipid droplet.  相似文献

Abstract.— The painted turtle, Chrysemys picta , is currently recognized as a continentally distributed polytypic species, ranging across North America from southern Canada to extreme northern Mexico. We analyzed variation in the rapidly evolving mitochondrial control region (CR) in 241 turtles from 117 localities across this range to examine whether the painted turtle represents a continentally distributed species based on molecular analysis. We found strong support for the novel hypothesis that C. p. dorsalis is the sister group to all remaining Chrysemys , with the remaining Chrysemys falling into a single, extremely wide-ranging and genetically undifferentiated species. Given our goal of an evolu-tionarily accurate taxonomy, we propose that two evolutionary lineages be recognized as species within Chrysemys : C. dorsalis (Agassiz 1857) in the southern Mississippi drainage region, and C. picta (Schneider 1783) from the rest of the range of the genus. Neither molecular nor recent morphological analyses argue for the hybrid origin of C. p. marginata as previously proposed. Within C. picta , we find evidence of at least two independent range expansions into previously glaciated regions of North America, one into New England and the other into the upper Midwest. We further find evidence of a massive extinction/recolonization event across the Great Plains/Rocky Mountain region encompassing over half the continental United States. The timing and extent of this colonization is consistent with a recently proposed regional aridification as the Laurentide ice sheets receded approximately 14,000 years ago, and we tentatively propose this paleoclimatological event as a major factor shaping genetic variation in Chrysemys .  相似文献

High-dose intravenous immunoglobulin (IVIG) prevents immune damage by scavenging complement fragments C3b and C4b. We tested the hypothesis that exogenous immunoglobulin molecules also bind anaphylatoxins C3a and C5a, thereby neutralizing their pro-inflammatory effects. Single-cell calcium measurements in HMC-1 human mast cells showed that a rise in intracellular calcium caused by C3a and C5a was inhibited in a concentration-dependent manner by IVIG, F(ab)2-IVIG and irrelevant human monoclonal antibody. C3a- and C5a-induced thromboxane (TXB2) generation and histamine release from HMC-1 cells and whole-blood basophils were also suppressed by exogenous immunoglobulins. In a mouse model of asthma, immunoglobulin treatment reduced cellular migration to the lung. Lethal C5a-mediated circulatory collapse in pigs was prevented by pretreatment with F(ab)2-IVIG. Molecular modeling, surface plasmon resonance (SPR) and western blot analyses suggested a physical association between anaphylatoxins and the constant region of F(ab)2. This binding could interfere with the role of C3a and C5a in inflammation.  相似文献

To develop and validate a modified OMNI rating of perceived exertion (RPE) scale for use during bench stepping exercise (OMNI-BS). Thirty women (age: 19.8 ± 1.8 years) undertook 2 experimental trials, separated by 7 days. Concurrent validity was established by examining the relation between physiological criterion variables, oxygen consumption (VO2), and heart rate (HR), with the concurrent variable, RPE from OMNI-BS, during 2 trials in which the intensity increased linearly (test 1) and intermittently (test 2). The first test consisted of 3-minute stages. Subjects stepped up and down on the bench at 120 b·min(-1). The test was terminated owing to subject fatigue. Exercise intensity increased as bench height increased every 3 minutes. The second test consisted of three 3-minute exercise bouts that reproduced exercise stage 1 (low intensity), stage 3 (moderate intensity), and stage 5 (high intensity) performed in the first test. The order of these 3 exercise bouts was counterbalanced. Intraclass correlation analysis from experimental trials indicated a strong positive association between RPE and VO2 (r = 0.96 and r = 0.95) and HR (r = 0.95 and r = 0.95). Concurrent validity for the OMNI-BS RPE scale was established for women performing bench stepping exercise.  相似文献

Extreme environmental events (EEEs) are likely to exert deleterious effects on populations. From 1996 to 2012 we studied the nesting dynamics of a riverine population of painted turtles (Chrysemys picta) that experienced seven years with significantly definable spring floods. We used capture–mark–recapture methods to estimate the relationships between more than 5 m and more than 6 m flood events and population parameters. Contrary to expectations, flooding was not associated with annual differences in survival, recruitment or annual population growth rates of the adult female segment of the population. These findings suggest that female C. picta exhibit resiliency to key EEE, which are expected to increase in frequency under climate change.  相似文献
