Calcium regulation of porcine aortic myosin   总被引:1,自引:0,他引:1  
Calcium regulation of actin-activated porcine aortic myosin MgATPase was studied. The MgATPase of the purified actomyosin was stimulated about 10-fold by 0.1 mM Ca2+. The 20,000 molecular weight light chain subunit (LC20) of myosin was phosphorylated by an endogenous kinase that required Ca2+. Half-maximal activation of both kinase and ATPase occurred at about 0.9 microM Ca2+. Phosphorylated and unphosphorylated myosins, free of actin, kinase, and phosphatase, were purified by gel filtration. The MgATPase of phosphorylated myosin was activated by rabbit skeletal muscle actin; unphosphorylated myosin was actin activated to a much lesser extent. Actin activation was maximal in the presence of Ca2+. Regulation of the aortic myosin MgATPase seems to involve both direct interaction of calcium with phosphorylated myosin and calcium activation of the myosin kinase. The MgATPase of trypsin-treated actomyosin did not require Ca2+ for full activity. The trypsin-treated actomyosin was devoid of LC20. When purified unphosphorylated aortic myosin was treated with trypsin, the LC20, was cleaved and the MgATPase, which was not appreciably actin activated before exposure to protease, was increased and was activated by skeletal muscle actin. After incubation of this light chain-depleted myosin with light chain from rabbit skeletal muscle myosin, the actin activation but not the increased activity, was abolished. Unphosphorylated LC20 seems to inhibit actin activation in this smooth muscle.  相似文献

Highly purified porcine brain actin has been prepared by a procedure involving anion-exchange chromatography, polymerization-depolymerization, and gel filtration. Electrophoresis of purified brain actin on polyacrylamide gels in the presence of sodium dodecyl sulfate shows a single protein band corresponding to more than 95% of the applied protein and migrating with the relative mobility of skeletal muscle actin. The amino acid composition of brain actin is similar but not identical to that of rabbit skeletal muscle actin. Brain actin activates the low ionic strength Mg2+-ATPase of skeletal muscle myosin to the same extent that skeletal muscle actin potentiates the muscle ATPase. Although similar to its skeletal muscle counterpart, brain actin is distinctly different. Isoelectric focusing experiments indicate that brain actin consists of at least two species, each of which is more basic than the α-species of skeletal muscle actin. The polymerization of brain actin was followed by viscometry and sedimentation techniques as a function of protein concentration, temperature, and ionic conditions. The critical actin concentrations of both brain and skeletal muscle actins polymerized at low ionic strength in the presence of 2.0 mm MgCl2 are similar and show little dependence upon temperature. When polymerized in the presence of 0. 1 m KCl, brain actin has a critical actin concentration that is higher and more dependent upon temperature than the corresponding critical concentration of skeletal muscle actin.  相似文献

Ionizing radiation and many cancer drugs have the potential to produce germ-cell mutations that might lead to genetic disease in the next generation. In a population-based study, we identified, from records in the Danish Cancer Registry, 4,676 children treated for cancer. Their 6,441 siblings provided a comparison cohort. The results of a search of the Central Population Register identified 2,630 live-born offspring of the survivors and 5,504 live-born offspring of their siblings. The occurrence of abnormal karyotypes diagnosed in these offspring and also in any pregnancies terminated following prenatal diagnosis of a chromosome abnormality was determined from the Danish Cytogenetic Registry. After exclusion of hereditary cases and inclusion of the prenatal cases, after correction for expected viability, the adjusted proportion of live-born children in survivor families with abnormal karyotypes (5.5/2,631.5 [0.21%]) was the same as that among the comparison sibling families (11.8/5,505.8 [0.21%]). There were no significant differences in the occurrence of Down syndrome (relative risk [RR]=1.07; 95% CI 0.16-5.47) or Turner syndrome (RR=1.32; 95% CI 0.17-7.96) among the children of cancer survivors, compared with the children of their siblings. These reassuring results are of importance to the survivors, to their families, and to genetic counselors.  相似文献

The physiology of the sulfur disproportionator Desulfocapsa sulfoexigens was investigated in batch cultures and in a pH-regulated continuously flushed fermentor system. It was shown that a sulphide scavanger in the form of ferric iron was not obligatory and that the control of pH allowed production of more biomass than was possible in carbonate buffered but unregulated batch cultures. Small amounts of sulphite were produced during disproportionation of elemental sulfur and thiosulphate. In addition, it was shown that in the presence of hydrogen, a respiratory type of process is favored before the disproportionation of sulphite, thiosulphate and elemental sulfur. Sulphate reduction was not observed. D. sulfoexigens assimilated inorganic carbon even in the presence of organic carbon sources. Inorganic carbon assimilation was probably catalyzed by the reverse CO-dehydrogenase pathway, which was supported by the constitutive expression of the gene encoding CO-dehydrogenase in cultures grown in the presence of acetate and by the high carbon fractionation values that are indicative of this pathway.  相似文献

Argentavis magnificens , the largest known flying bird, had a wingspan of over 6 m with a mass of 80 kg. Its enormous size suggests that it was not a powerful flapper. The wing shape is inferred as more like that of large extant birds that soar relatively slowly on thermals over land than of large pelagic birds that soar over water. Its high wing loading would have allowed it to fly in moderate to strong winds that must have been prohibitive for the largest known contemporary thermal soarers. The high wing loading would have been ill-suited to flight under poor thermal conditions, but it would have been useful in slope-soaring on uprising air current against hillsides. We propose that Argentavis had a large home range that included a nesting area in the mountains of western and northwestern Argentina, and a feeding area in the Pampas.  相似文献

Three species are often accepted in Taeniatherum Nevski. Analysis of morphological characters show that indistinct limits occur between them. All taxa are found to be diploid (2n=14), with the same karyotype, but artificial hybridization shows genetical barrier to exist. For these reasons subspecific level is chosen for the taxa, viz. T. caput-medusae (L.) Nevski ssp. caput-medusae, T. caput-medusae ssp. crinitum (Schreb.) Melderis and T. caput-medusae ssp. asperum (Simk.) Melderis. Large population declines were reported for the thick-billed murre (Uria lomvia) in Greenland for the period 1930s–1980s, but no national status has been published for the past 20 years. Meanwhile, the murres have gained more protection and several human-induced mortality factors have been markedly reduced. Here, we give an updated status based on the past 30 years of murre count data. The total Greenland population in 2011 was estimated to 468,300 birds (95 % CI 430,700–505,900) or around 342,000 breeding pairs, distributed within 19 colonies. This represents an overall reduction of 13 % since the mid-/late 1980s. In the same period, five colonies went extinct. Large and apparently stable colonies in Qaanaaq (Northwest Greenland) account for more than half the population (68 %), but most other colonies declined heavily, with up to 6 % p.a. in the most critical areas. So far, nothing indicates that food is a limiting factor in Greenland during the breeding season, although rather few colonies have been studied in details. In contrast, illegal hunting and disturbances during the breeding season are still a problem in Greenland, despite more restrictive hunting regulations, and may explain much of the continued population decline. In addition, recent studies from Svalbard indicate that a large-scale deterioration of the marine environment in the North Atlantic, due to oceanographic changes, may impact recruitment to some of the Greenland colonies. Murre colonies in southern Upernavik, Disko Bay, South Greenland and East Greenland are in urgent need of additional conservation initiatives to avoid further declines and local extinctions.  相似文献
