Sparrow , Frederick K. and Yamunga Lingappa . (U. Michigan, Ann Arbor.) Observations on chytridiaceous parasites of phanerogams. VIII. Urophlyctis (Physoderma) pluriannnlatus and U. majus. Amer. Jour. Bot. 47(3): 202—209. Illus. 1960.—Urophlyctis pluriannulatus, an obligate parasite of Sanicula spp., has an endobiotic phase which is strongly polycentric and produces small crateriform galls on the petioles and blades of the host leaves. The agent accomplishing infection is not known but is probably a zygote. The first cell of the parasite established in the host is the so-called “primary turbinate organ.” This becomes multinucleate, is somewhat pyriform and becomes multicellular by 2 methods: (1) by cleavage into peripheral segments; or (2) by division into cells, each with its own cell wall. Replication of the thallus is accomplished by the production of nucleated outgrowths bearing haustoria which elongate, become ribbon-like, somewhat roughened and lumened, and produce distally turbinate organs of a second order. Tertiary, etc. turbinate organs are produced in like manner. Resting spores usually form at the tip of an extremely short outgrowth from the apex of a turbinate organ. These bear a supra-equatorial crown of 7—10 branched haustoria. Rarely, monocentric thalli are formed, in which a single turbinate organ becomes converted into a resting spore. All nuclear division figures were intranuclear. The fungus produced marked enlargement of infected host cells and their nuclei, and caused division of neighboring cells. As development continues, lysis of the surrounding host walls takes place and a large cavity bearing a dense symplast and numerous host nuclei is formed, within which is the thallus of the parasite. At maturity, all traces of symplast and of fungus, except for resting spores, disappear. Urophlyctis majus, a parasite on leaves of Rumex orbiculatus, hitherto known only from its resting spore stage, has a pattern of development strikingly similar to that of U. pluriannulatus. Here, however, turbinate cells only form peripheral segments. Furthermore, the “hyphae” are smooth and without a lumen. Aside from size differences, the mature thallus with resting spores, unbranched (not branched) haustorial tufts, etc. is like that of the Sanicula parasite. The galls produced were compartmentalized, dark red to black, usually surrounded by a reddish zone, and early dropped from the leaf. No undoubted evidence of the epibiotic gametangial phase was found in either species.  相似文献

During the budding of coat protein complex II (COPII) vesicles from transitional endoplasmic reticulum (tER) sites, Sec16 has been proposed to play two distinct roles: negatively regulating COPII turnover and organizing COPII assembly at tER sites. We tested these ideas using the yeast Pichia pastoris. Redistribution of Sec16 to the cytosol accelerates tER dynamics, supporting a negative regulatory role for Sec16. To evaluate a possible COPII organization role, we dissected the functional regions of Sec16. The central conserved domain, which had been implicated in coordinating COPII assembly, is actually dispensable for normal tER structure. An upstream conserved region (UCR) localizes Sec16 to tER sites. The UCR binds COPII components, and removal of COPII from tER sites also removes Sec16, indicating that COPII recruits Sec16 rather than the other way around. We propose that Sec16 does not in fact organize COPII. Instead, regulation of COPII turnover can account for the influence of Sec16 on tER sites.  相似文献

We explored macroinvertebrate size-differential drift in the lower Mississippi River (a 9th order system). Because this river system is highly turbid, we hypothesized that visually-dependent vertebrate predators feeding on drifting organisms would be at a disadvantage. Thus, size-differential drift should not occur. For one 24-hour period in both January and April, six drift nets were used to sample surface drift. Nets were emptied once every four hours. Individual intra-ocular distances of three macroinvertebrate species (Hydropsyche orris: Trichoptera, Hexagenia limbata: Ephemeroptera, Macrobrachium ohione: Crustacea) were measured. Percentages of size classes in the drift were determined. In both months, large individuals of H. orris and H. limbata were prevalent in the nocturnal but scarce in the diurnal drift. In January, large M. ohione drifted regardless of time. In April, large M. ohione predominated the nocturnal drift. Our results could not be attributed solely to vertebrate predator avoidance. Other mechanisms such as diel microhabitat migration and current velocity may have accounted for the results.  相似文献

Cyclic guanosine 3′,5′-monophosphate (cyclic GMP) stimulates nucleic acid synthesis in lymphocytes, and has been implicated as the intracellular effector of the actions of mitogenic agents on these cells. In the present study, we examined the specificity of the mitogenic activity of cyclic GMP and of its 8-bromo (Br) derivatives, and the effects of the T cell mitogens, concanavalin A, phytohemagglutinin, and staphylococcal entertoxin B (SEB) on the cyclic GMP content and guanylate cyclase activity of mouse splenic lymphocytes. Cyclic GMP and guanosine modestly increased the incorporation of [3H]thymidine into DNA by cultured lymphocytes, but were far less effective than their 8-Br-derivatives. However, on a molar basis the mitogenic activity of both 8-Br-guanosine and 8-Br-5′-GMP exceeded that of 8-Br-cyclic GMP, when tested in the presence and absence of serum in the culture media. Combined addition of maximal doses of these nucleotides did not give additive stimulatory effects, suggesting an action on a common subpopulation of cells, and possibly a common mechanism. By contrast, cyclic AMP, 8-Br-cyclic AMP, 8-Br-adenosine, cholera toxin and prostaglandin E1 suppressed both basal [3]thymidine incorporation and stimulation of this parameter by T-cell line mitogens and the guanosine nucleotides. Rapid effects of concanavalin A, phytohemagglutinin, SEB, guanosine, 5′-GMP, 8-Br-guanosine, and 8-Br-5′-GMP on the cyclic GMP content of murine lymphocytes could not be demonstrated. Similarly, concanalin A, phytohemagglutinin and SEB failed to alter guanylate cyclase activity when added directly to cellular homogenates or pre-incubated with intact cels. Conversely, carbamylcholine rapidly increased lymphocyte cyclic GMP but was not mitogenic.These results are consistent with the hypothesis that cyclic GMP and cyclic AMP are antagonistic in their influence on lymphocyte mitogenesis. However, they also demonstrate that related nucleotides are more potent mitogens than cyclic GMP and suggest that activation of murine lymphocytes by concanavalin A, phytohemagglutinin and SEB may not be mediated by rapid increases in cellular cyclic GMP content. Since high concentrations of exogenous cyclic GMP and related nucleotides must be used to influence DNA synthesis, the biologic significance of this effect remains uncertain.  相似文献
