Strains of type 6 (S 43) and type 14 group A streptococci were grown with M-protein production in the presence of chemically defined synthetic media slightly modified from that previously employed for the growth of a nonproducer of M protein (type 4). The M protein, which is associated with virulence in group A streptococcus, was previously produced in growing cultures only with complex media. The bacterial growth with the biosynthesis of M protein in synthetic medium was obtained by successive adaptation in steady-state culture with decreasing amounts of Todd-Hewitt broth. The synthesis continued for at least 480 generations at pH 7.3 and with a generation time of 84 min. Glucose was the limiting nutrilite and the concentration of reducing agents in the medium was critical. The M protein was identified by gel diffusion against type-specific antisera from the Communicable Disease Center and from R. Lancefield. The yield of M protein obtained from organisms grown in the continuous-culture device was comparable to that from standard broth stationary cultures. 相似文献

Differences in the induction response and the initial two reactions of quinoline degradation between short-term (2 days)- and long-term (60 to 80 days)-starved cells of a subsurface Pseudomonas cepacia strain were examined by using continuous-flow columns. The ability of bacteria that are indigenous to oligotrophic environments to respond to a contaminant was assessed by using long-term starvation to induce a cell physiology that simulates the in situ physiology of the bacteria. With quinoline concentrations of 39 and 155 μM, long-term-starved cells converted quinoline to degradation products more efficiently than did short-term-starved cells. Quinoline concentrations of 155 μM and, to a greater extent, 775 μM had an inhibitory effect on induction in long-term-starved cells. However, only the length of the induction process was affected with these quinoline concentrations; degradation of quinoline at the steady state for long-term-starved cells was equal to or better than that for short-term-starved cells. The induction time for short-term-starved cells did not increase progressively with increasing quinoline concentration. Experiments with starved cells are important for the development of accurate predictive models of contaminant transport in the subsurface because starvation, which induces a cell physiology that simulates the in situ physiology of many bacteria, may affect the induction process. 相似文献

Procedures were developed for the differential enumeration of an added strain of Lactobacillus plantarum and indigenous lactic acid bacteria (LAB) during the fermentation of brined cucumbers. The added strain was an N,N-nitrosoguanidine-generated mutant that lacked the ability to produce CO2 from malic acid (MDC-). The MDC- phenotype is desirable because CO2 production from malic acid decarboxylation has been shown to contribute to bloater formation in fermented cucumbers. A basal medium containing malic acid and adjusted to pH 4.0 permitted growth of indigenous LAB (predominantly MDC+), but not growth of the added MDC- culture. Transformation of the MDC- culture by electroporation with cloning vector pGK12 conferred chloramphenicol resistance, which permitted selective enumeration of this culture. The reversion frequency of the MDC- mutation was determined by a fluctuation test to be less than 10-10. The level of retention of plasmid pGK12 was greater than 90% after 10 generations in cucumber juice medium at 32°C. With the procedures developed, we were able to establish the ratio of MDC- to MDC+ LAB that results in malic acid retention in fermentations of filter-sterilized cucumber juice and unsterilized whole cucumbers under specified conditions. 相似文献

Isozyme variation was investigated in populations ofPhragmites australis (Poaceae) which have recently invaded and taken over marsh habitat of the Mississippi River delta. Infrared aerial photographs in the Garden Island Bay subdelta of the extensiveP. australis populations reveal distinct, clone-like circular patches within a predominant background. Preliminary evidence indicates that the infrared color differences represent distinct morphological types. However, there are no obvious environmental factors that could account for the peculiar patterns.P. australis collections were taken from five separate and distinct patches and adjacent background. Only two electrophoretic phenotypes were found: one from patches and one from the background. In comparing the two, 20% of the 40 loci scored are fixed for alternate alleles. These results indicate a clear correspondence of infrared reflectance with electrophoretic phenotype. In addition, the genetic uniformity as evidenced by the discovery of only two electrophoretic phenotypes supports the contention that the recent spread ofP. australis throughout the Mississippi River delta has been primarily, if not exclusively, a result of vegetative propagation. 相似文献

Two hypotheses about the ralationship of diabroticina beetles and plants in the family Cucurbitaceae are tested: (1) evolution of sensory receptors for cucurbitacins by diabroticina beetles was in part due to the predator protection offered by ingestion of these compounds, and (2) commercial varieties of cucurbitacin-producing cucumber offer Diabrotica undecimpunctata howardi Barber chemical protection from some potential sources of natural controlSpotted cucumber beetles fed either Marketmore 70 cucumber which contains cucurbitacin-C or Marketmore 72 which totally lacks cucurbitacin were presented to four species of vertebrate predators that commonly occur in the summer and/or winter habitats of D. u. howardi: Bufo americanus, B. fowleri, Peromyscus maniculatus, Colinus virginianus. None of the four species were significantly deterred from preying on beetles that had eaten Marketmore 70 cucumber. These results do not support either of the two hypotheses. The limitations of these negative results as evidence for refutation of the first hypothesis are discussed.
Résumé Deux hypothèses concernant les relations entre les Diabroticina et les Cucurbitaceae ont été examinées: (1) l'évolution chez les Diabroticina des récepteurs pour les Cucurbitacines est due en partie à la protection contre les prédateurs apportée par la consommation de ces produits (2) les variétés modernes de concombres produisant de la cucurbitacine fournissent à D. undecimpunctata howardi Barber une protection chimique contre quelques sources potentielles de lutte biologique.Des D. undecimpunctata howardi, ayant consommé soit des concombres Marketmore 70 contenant de la cucurbitacine C, soit des concombres Marketmore 72 qui ont totalement perdu leur cucurbitacine, ont été présentés à quatre espèces de vertébrés prédateurs, qui s'observent fréquemment dans les habitats d'été ou d'hiver de D. undecimpunctata howardi soient Bufo americanus, B. fowleri, Peromyscus maniculatus, Colinus virginianus. Aucune de ces quatre espèces n'est significativement dissuadée d'attaquer les coléoptères qui on consommé les concombres Marketmore 70. Ces résultats n'étayent aucune des deux hypothèses.
