The joint effects of parental gene fixation and consanguinity of mates upon the fitness of matings was examined in Mimulus guttatus. Plants from four populations were crossed at five levels of genetic relatedness, and five viability characters were scored in progeny. Parental gene fixation at 12 polymorphic allozyme loci was partitioned into local, subpopulation, and population components. A model is proposed wherein parental gene fixation influences distance-dependent crossing success. At a fixed locus, inbreeding is favored if natural selection caused allele fixation, or is disfavored if gene fixation was random. The distance between mates required to eliminate gene fixation depends upon patch size of fixation. When selective fixation and patch size differ among loci, an optimal crossing distance is possible. In M. guttatus, progeny viability generally decreased with greater relatedness between mates, but this decrease was often heterogeneous among populations. The highest pollen viability and the lowest seed set were found at an intermediate relatedness between mates. To determine whether parental gene fixation influences these crossing patterns, a type of mutational-load analysis was performed. Progeny fitness was regressed on parent F and fitness estimated at F = 1. This was done for each component of F, for a) crosses that maintain gene fixation and b) crosses that eliminate gene fixation. A multiplicative, composite measure of fitness indicates that, in M. guttatus, genes fixed during local or population differentiation favor outbreeding, while genes fixed during subpopulation differentiation favor inbreeding. This predicts that random mating within subpopulations confers highest progeny fitness, exclusive of between-population matings. However, predictions did not fit the observed patterns of crossing success very well, perhaps because gene fixation was relatively low or was not adequately measured at loci influencing fitness.  相似文献

Peptide T, from the human immunodeficiency virus (HIV), whose sequence is Ala-Ser-Thr-Thr-Thr-Asn-Tyr-Thr, has been shown to inhibit attachment of this virus to T cells and neural cells bearing the CD4 receptor. This peptide shares extensive homology with the 19–26 segment of ribonuclease A (RNase A), whose sequence is Ala-Ala-Ser-Ser-Ser-Asn-Tyr-Cys. Based on comparison of the structures of peptides occurring in proteins of known structure that are homologous to peptide T,viz, RNase A and endothiapepsin and on conformational energy calculations, we predicted that peptide T adopts a structure much like that for residues 19–26 in RNase A. A critical feature is a bend involving residues Thr 4-Asn 7 in peptide T corresponding to Ser 22-Tyr 25 in the RNase A peptide. Our proposed structure for peptide T has recently been confirmed by Cotelleet al. (Biochem. Biophys. Res. Commun. 171, 596–602). We now show directly that the RNase A peptide, with Met replacing Cys 26 to prevent disulfide exchange reactions, strongly induces monocyte-chemotaxis that is blocked by anti-CD4 monoclonal antibody. Both peptide T and RNase A fail to induce chemotaxis, however, in neutrophils which do not express surface CD4 receptors. These results suggest that both peptides interact with the CD4 receptor in inducing monocyte chemotaxis. We have also prepared cyclo-RNase A peptide with Met 26. Using molecular dynamics and conformational energy calculations, we find that the cyclic peptide cannot form a bend structure involving Ser 22-Tyr 25 that is superimposable on the RNase A bend. Indeed, we find that this peptide is inactive in inducing monocyte chemotaxis despite the fact that its amino acid sequence is identical to that of the open chain form. This result suggests that a correlation between the -bend structure of the RNase A peptide and peptide T and their abilities to bind to the CD4 receptor.  相似文献

Summary Some limnological aspects of two small, widely fluctuating reservoirs were studied for one year. A well defined thermocline did not form in either reservoir. Annual heat budgets were about 15,000 cal/cm2 for both waters. Dissolved oxygen concentrations were low on the bottom during the summer months. Both reservoirs were alkaline with the pH ranging from 7.5 to 8.9.The mean primary production summer rates of nannoplankton were 9.9 mg C/m3/hr for Crowthers Reservoir and 6.7 mg C/m3/hr for Deep Creek Reservoir. Standing crop of zooplankton was highest in Deep Creek Reservoir. Biomass of detrital heterotrophs was highest in Crowthers.
Resumen Algunos aspectos limnológicos de dos reservorios pequeños con extensa fluctuación se estudiaron por un año. Termocline (thermocline)bien definido no se formó en uno u otro reservorio. Las acumulaciones anuales de calor fueron más o menos 15,000 cal/cm2 (calorías por centímetros cuadrados) por ambas aguas. La concentración de oxígeno disuelto fué bajo en el fondo durante los meses del verano. Ambos reservorios fueron alcalinos con el pH fluctuando desde 7.5 hasta 8.9.Les clasificaciones del pequeño plankton (nannoplankton) durante la producción en el verano fueron 9.9 mg (miligramos) C/m3/hr (carbóno por metro cúbico por hora) para el Reservorio Crowthers y 6.7 mg C/m3/hr para el Reservorio Deep Creek. La cosecha fija de zooplancton fué más fuerte en el Reservorior Deep Creek. La biomasa de heterótrofo detríticos fué más fuerte en Crowthers.

Rapid, short duration mechanical compression of lobster giant axons by a crystal-driven stylus produces a depolarization and an increase in membrane conductance which develop immediately with compression but take several seconds to recover. The conductance increase occurs even when the depolarization is prevented electrically. If sodium is removed from the external medium or if procaine is added to it, compression produces almost no depolarization. Small bundles of myelinated frog fibers are depolarized by rapid compression but recover very rapidly (milliseconds); "off" responses are occasionally seen. The results are discussed in terms of the mechanoelectric transducer behavior of an axon membrane.  相似文献

Extragenic sequences in genomes, such as microRNA and CRISPR, are vital players in the cell. Repetitive extragenic palindromic sequences (REPs) are a class of extragenic sequences, which form nucleotide stem-loop structures. REPs are found in many bacterial species at a high copy number and are important in regulation of certain bacterial functions, such as Integration Host Factor recruitment and mRNA turnover. Although a new clade of putative transposases (RAYTs or TnpAREP) is often associated with an increase in these repeats, it is not clear how these proteins might have directed amplification of REPs. We report here the structure to 2.6 Å of TnpAREP from Escherichia coli MG1655 bound to a REP. Sequence analysis showed that TnpAREP is highly related to the IS200/IS605 family, but in contrast to IS200/IS605 transposases, TnpAREP is a monomer, is auto-inhibited and is active only in manganese. These features suggest that, relative to IS200/IS605 transposases, it has evolved a different mechanism for the movement of discrete segments of DNA and has been severely down-regulated, perhaps to prevent REPs from sweeping through genomes.  相似文献

Chromium (Cr) is a heavy metal risk to human health, and a contaminant found in agricultural soils and industrial sites. Phytoremediation, which relies on phytoextraction of Cr with biological organisms, is an important alternative to costly physical and chemical methods of treating contaminated sites. The ability of the arbuscular mycorrhizal fungus (AM),Glomus intraradices, to enhance Cr uptake and plant tolerance was tested on the growth and gas exchange of sunflower (Helianthus annuus L.). Mycorrhizal-colonized (AM) and non-inoculated (Non-AM) sunflower plants were subjected to two Cr species [trivalent cation (Cr3+) Cr(III) , and divalent dichromate anion (Cr2O7) Cr(VI) ]. Both Cr species depressed plant growth, decreased net photosynthesis (A) and increased the vapor pressure difference; however, Cr(VI) was more toxic. Chromium accumulation was greatest in roots, intermediate in stems and leaves, and lowest in flowers. Greater Cr accumulation occurred with Cr(VI) than Cr(III). AM enhanced the ability of sunflower plants to tolerate and hyperaccumulate Cr. At higher Cr levels greater mycorrhizal dependency occurred, as indicated by proportionally greater growth, higherA and reduced visual symptoms of stress, compared to Non-AM plants. AM plants had greater Cr-accumulating ability than Non-AM plants at the highest concentrations of Cr(III) and Cr(VI), as indicated by the greater Cr phytoextraction coefficient. Mycorrhizal colonization (arbuscule, vesicle, and hyphae formation) was more adversely affected by Cr(VI) than Cr(III), however high levels of colonization still occurred at even the most toxic levels. Arbuscules, which play an important role in mineral ion exchange in root cortical cells, had the greatest sensitivity to Cr toxicity. Higher levels of both Cr species reduced leaf tissue phosphorus (P). While tissue P was higher in AM plants at the highest Cr(III) level, tissue P did not account for mycorrhizal benefits observed with Cr(VI) plants.  相似文献
