A duct system was arranged through which flowed air of controlled ion concentration. In series was a chamber with a flexible dielectric floor which made possible electrical detection of activity.Exposure to various ion concentrations of either polarity produced pronounced effects.In addition to gross motor activity six other behavioural parameters were recorded. The lowest ion concentration were most effective. In addition to presenting a controlled ambient ion concertration the ion current drawn by the animal was measured. This varied markedly from rat to rat.

Zusammenfassung Ein Kanalsystem zum Durchfluss von Luft bekannter Ionenkonzentration wurde eingerichtet. Angeschlossen war eine Kammer mit einem beweglichen dielektrischen Boden, der die Registrierung der Aktivität von Ratten ermöglichte. Neben der motorischen Aktivität wurden 6 andere Verhaltensformen der Tiere registriert.Die niedrigste Ionenkonzentration war am wirksamsten zur Anregung der Aktivität. Neben der Ionenkonzentration in der Kammer wurde der von den Tieren eingeatmete Ionenstrom gemessen; er variierte stark von Tier zu Tier.
Resume On a construit un canal dans lequel il est possible de faire circuler de l'air dont la concentration en ions est contrôlable. On y a adjoint une cellule dont le sol est constitué par un diélectrique flexible permettant de déceler électriquement l'activité de petits animaux.En exposant des rats à des concentrations ioniques variables et de polarité différente, on peut constater des effets prononcés sur le comportement des animaux. En effet, en plus de l'activité motrice, on a enregistré six autres formes de ce comportement. Ce sont les plus faibles concentrations qui produisent le plus d'effets. A part la concentration en ions à l'intérieur de la cellule, on a également mesuré la quantité d'ions absorbés par les bêtes par voie respiratoire. Cette quantité varie fortement d'un individu à l'autre.

Neuropeptides such as substance P are implicated in inflammation mediated by sensory nerves (neurogenic inflammation), but the roles in disease of these peptides and the peptidases that degrade them are not understood. It is well established that inflammation is a prominent feature of several airway diseases, including viral infections, asthma, bronchitis, and cystic fibrosis. These diseases are characterized by cough, airway edema, and abnormal secretory and bronchoconstrictor responses, all of which can be elicited by substance P. The effects of substance P and other peptides that may be involved in inflammation are decreased by endogenous neutral endopeptidase (NEP; also called enkephalinase, EC 3.4.24.11), which is a peptidase that degrades substance P and other peptides. In the present study, we report that rats with histories of infections caused by common respiratory tract pathogens (parainfluenza virus type 1, rat corona-virus, and Mycoplasma pulmonis) not only have greater susceptibility to neurogenic inflammatory responses than do pathogen-free rats but also have a lower activity of NEP in the trachea. This reduction in NEP activity may cause the increased susceptibility to neurogenic inflammation by allowing higher concentrations of substance P to reach tachykinin receptors in the trachea. Thus decreased NEP activity may exacerbate some of the pathological responses in animals with respiratory tract infections. 相似文献

The effect of palmitic acid on basal and insulin-stimulated incorporation of glucose into rat adipocytes was studied. Palmitic acid (2.40 mM) stimulated basal as well as insulin-stimulated glucose incorporation in rat adipocytes three and twofold, respectively. Similar degrees of stimulation of basal glucose oxidation by palmitate were also observed. The ability of palmitic acid to stimulate glucose uptake was additive with respect to the stimulation induced by insulin and was proportional to the palmitic acid concentration between 0.15 mM and 2.40 mM. Stimulation of glucose incorporation by palmitic acid was inhibited by preincubating the cells with quin2-AM, which accumulates intracellularly yielding the trapped chelator form. quin2, which binds intracellular Ca2+.The concentration of quin2-AM required for half-maximal inhibition of palmitic acid stimulated glucose incorporation was 3.8 +/- 1.2 microM (mean +/- SEM). The inhibition of palmitic acid-stimulated glucose incorporation by quin2-AM (10 microM) was overcome by incubating cells with the Ca2+ ionophore, A23187, in the presence of extracellular Ca2+ (2.6 mM). Chelation of extracellular Ca2+ with EGTA did not significantly affect the magnitude of palmitic acid-stimulated glucose incorporation. Dantrolene (12.5-100 microM) failed to affect basal or palmitic acid-stimulated glucose incorporation. These findings suggest that palmitic acid stimulates incorporation of glucose in the adipocyte by a mechanism dependent upon intracellular but not extracellular Ca2+. 相似文献

Transposon mutagenesis of baculoviruses: analysis of TFP3 lepidopteran transposon insertions at the FP locus of nuclear polyhedrosis viruses 总被引:3,自引:0,他引:3
We report the complete sequences of two representatives of the TFP3 transposable element family of the lepidopteran, Trichoplusia ni. These elements were isolated as insertions mobilized from the Lepidopteran host genome into two closely related nuclear polyhedrosis viruses (NPV) during infection. Both elements inserted within the 500-bp FP locus of the respective viral genomes (map units 36.0 to 37.0), causing a distinctive plaque morphology phenotype and the loss of a 25-kDa viral-specific protein. Both insertions occurred at the identical TTAA target site in the respective genomes, in the same relative orientation, and are flanked by 15-bp imperfect inverted repeats. The inserted elements interrupt the 25K open reading frame (ORF). One of these FP mutants undergoes spontaneous reversion. Sequence analysis at the excision site of a spontaneous revertant demonstrates that the TFP3 elements are capable of precise excision, restoring the expression of the 25-kDa protein. We also compare the sequences of the 25K genes of the Autographa californica and Galleria mellonella viruses (AcMNPV and GmMNPV, respectively). The 25K gene sequences diverge in five areas, resulting in an additional EcoRV and TaqI site within the GmMNPV 25K gene, and extension of the ORF for an additional 2 amino acids at the C-terminus of the predicted GmMNPV 25 kDa protein. The phenomenon of transposon mutagenesis of Baculovirus genomes provides a unique opportunity for analysis of transposon mobility. 相似文献
