Pseudomonas aeruginosa is the most common cause of hospital-acquired pneumonia and a killer of immunocompromised patients. We and others have demonstrated that the type III secretion system (T3SS) effector protein ExoT plays a pivotal role in facilitating P. aeruginosa pathogenesis. ExoT possesses an N-terminal GTPase-activating protein (GAP) domain and a C-terminal ADP-ribosyltransferase (ADPRT) domain. Because it targets multiple non-overlapping cellular targets, ExoT performs several distinct virulence functions for P. aeruginosa, including induction of apoptosis in a variety of target host cells. Both the ADPRT and the GAP domain activities contribute to ExoT-induced apoptosis. The ADPRT domain of ExoT induces atypical anoikis by transforming an innocuous cellular protein, Crk, into a cytotoxin, which interferes with integrin survival signaling. However, the mechanism underlying the GAP-induced apoptosis remains unknown. In this study, we demonstrate that the GAP domain activity is both necessary and sufficient to induce mitochondrial (intrinsic) apoptosis. We show that intoxication with GAP domain results in: (i) JNK1/2 activation; (ii) substantial increases in the mitochondrial levels of activated pro-apoptotic proteins Bax and Bid, and to a lesser extent Bim; (iii) loss of mitochondrial membrane potential and cytochrome c release; and (iv) activation of initiator caspase-9 and executioner caspase-3. Further, GAP-induced apoptosis is partially mediated by JNK1/2, but it is completely dependent on caspase-9 activity. Together, the ADPRT and the GAP domains make ExoT into a highly versatile and potent cytotoxin, capable of inducing multiple forms of apoptosis in target host cells.  相似文献

The FtsH2 protease, encoded by the slr0228 gene, plays a key role in the selective degradation of photodamaged D1 protein during the repair of Photosystem II (PSII) in the cyanobacterium Synechocystis sp. PCC 6803. To test whether additional proteases might be involved in D1 degradation during high rates of photodamage, we have studied the synthesis and degradation of the D1 protein in ΔPsbO and ΔPsbV mutants, in which the CaMn4 cluster catalyzing oxygen evolution is less stable, and in the D1 processing mutants, D1-S345P and ΔCtpA, which are unable to assemble a functional cluster. All four mutants exhibited a dramatically increased rate of D1 degradation in high light compared to the wild-type. Additional inactivation of the ftsH2 gene slowed the rate of D1 degradation dramatically and increased the level of PSII complexes. We conclude that FtsH2 plays a major role in the degradation of both precursor and mature forms of D1 following donor-side photoinhibition. However, this conclusion concerned only D1 assembled into larger complexes containing at least D2 and CP47. In the ΔpsbEFLJ deletion mutant blocked at an early stage in PSII assembly, unassembled D1 protein was efficiently degraded in the absence of FtsH2 pointing to the involvement of other protease(s). Significantly, the ΔPsbO mutant displayed unusually low levels of cellular chlorophyll at extremely low-light intensities. The possibilities that PSII repair may limit the availability of chlorophyll for the biogenesis of other chlorophyll-binding proteins and that PsbO might have a regulatory role in PSII repair are discussed.  相似文献

Zusammenfassung Nun könnte man aber einwenden, es wäre wohl denkbar, daß die Chromosomenzahl in der Plumula und in Wurzelspitzen 18, in Pollenmutterzellen 9 beträgt, ferner daß die Keimpflanzen und außerdem jene geschoßten Exemplare, die bekanntlich schon im ersten Jahre blühen, ohne daß es zur Kopfbildung kommt, in den Körperzellen normal diploid sind, damit sei aber noch nicht bewiesen, daß dies auch bei den Chromosomengarnituren des oft riesigen Krautkopfes der Fall ist. Vielmehr könnte im Verlaufe der ontogenetischen Entwicklung auf ähnliche Weise, wie sie H.Winkler (1916) in Körperzellen geschildert hat (siehe Zitat auf Seite 107), Tetraploidie entstehen, welche mit der Ausbildung des Krautkopfes in Beziehung steht, so daß dieser normalerweise in allen Zellen eine Verdoppelung der diploiden Chromosomenzahl aufweist, mithin also die wichtigste Eigentümlichkeit einer Gigas-Form besitzt. Zu dieser Annahme verleitet vor allem Fig. 37, die im Dauergewebe des Keimblattstieles gefunden wurde. Da die ausgewachsenen Blätter der Kulturrassen ebenfalls Dauergewebe vorstellen, wäre die Möglichkeit vorhanden, daß Tetraploidie dort normal sei.Die Wahrscheinlichkeit dieser Annahme wird aber vermindert, wenn man folgendes bedenkt:Wäre der Krautkopf tetraploid, dann müßten die Reduktionsteilungen der Pollenmutterzellen die doppelte haploide Zahl, also 18 Chromosomen zeigen, da doch die Blüten im zweiten Jahr aus dem Kopf des ersten Jahres hervorgehen. N.Shimotomai beobachtete in Pollenmutterzellen jedoch nur neun Chromosomen, daher dürfte der Krautkopf die normale diploide Zahl aufweisen. Allerdings wäre noch möglich, daß die Körperzellen des nächstjährigen Blütenstandes infolge somatischer Reduktionsteilungen (R. R.Gates 1912, B.Nemec 1910) wieder diploid und die aus ihnen hervorgehenden Pollenmutterzellen haploid werden, doch glaube ich, die Annahme ist zu gezwungen, um auch nur eine geringe Wahrscheinlichkeit für sich zu haben. Eine erhöhte Chromosomenzahl ist in den großen Blättern des Kopfes von Kraut und Wirsingkohl ferner deswegen kaum anzunehmen, weil die Zellen der Blattepidermis vonBrassica montana und auch vom Helgoländer Wildkohl im Durchschnitt sogar etwas größer sind, als die der Kulturrassen.Wir dürfen demnach abschließend sagen:Brassica oleracea L. var.capitata L., var.sabauda L., var.acephala DC. und var.gongylodes L. sind nach diesen Untersuchungen nichtals Riesenformen im Sinne der modernen Genetik anzusehen, nicht als Riesenformen mit vermehrter Chromosomenzahl oder vergrößerten Chromosomen.  相似文献

Fragmentarily preserved shells – mainly pro-ostraca, in several cases also phragmocones – occurring together with arm hooks and the ink sac of the Carnian (Late Triassic) coleoid cephalopod Phragmoteuthis bisinuata (Bronn) from Lunz (Austria) are examined with the scanning electron microscope and energy-dispersive spectrometer. The pro-ostracum bears black, shiny, pitch-like sheets. The black sheets, the ink sac content and the arm hooks have a granular ultrastructure of 0.1–1 μm grain size. The arm hooks and black sheets are micro-laminated; each lamina consists of fibres. The ink consists of an agglomerate of grains. On the ventral (internal) side of the pro-ostracum, the black sheets occasionally bear agglomerates of homogeneous, ink-like material along with heterogeneous structures. The pro-ostracum has crystal-shaped units with lamello-columnar ultrastructure of the inner layer and plate ultrastructure of the outer layer. This resembles the Late Triassic Lunzoteuthis [Doguzhaeva, L.A., Mutvei, H., Summesberger, H., 2005a. A Late Triassic coleoid from the Austrian Alps: the pro-ostracum viewpoint. The black sheets, the material on their inner surface, the ink and the arm hooks consist of carbon, occasionally with minor amounts of sulfur. The shell is of calcium carbonate.Based on their organic composition, position in the shell and lamello-fibrillar ultrastructure, the black sheets are considered to be remains of the mantle, sometimes with ink sac and soft body debris. The carbon composition and granular ultrastructure of arm hooks, ink, and soft tissue remains indicate that the non-mineralized structures are pseudomorphosed by carbon (carbonization), possibly due to C-accumulating bacteria.  相似文献

Isolated rat pancreatic islets were perifused and analyzed for phosphate content immediately following the transient increase in the efflux of orthophosphate which occurs when insulin secretion is stimulated by glucose. In some instances, islets were perifused directly following isolation to minimize preparative delay; in others, islets were prelabeled during incubation with [32P]orthophosphate for 90 min prior to perifusion. In both experimental situations, total islet phosphate content declined 40–50% following exposure to stimulating concentrations of glucose and initiation of enhanced insulin release. In the experiments with prelabeled islets, tissue content of [32P]orthophosphate fell to a similar extent so that the specific radioactivity of islet orthophosphate was unaffected. Inhibition of heightened insulin release with Ni2+ did not modify the decrements in total or radioactive tissue orthophosphate, thus indicating that these responses to islet stimulation reflect events which are proximal to activated exocytosis. Simultaneous analyses for tissue ATP and ADP demonstrated that the efflux in orthophosphate and reduction in tissue orthophosphate content were not mediated via net changes in islet adenine nucleotides. The observations represent the first documentation that a net reduction of tissue inorganic phosphate is one of the early components of stimulus-secretion coupling in isolated pancreatic islets.  相似文献

Cyanide-insensitive mitochondria from Saccharomycopsis lipolytica possess an exogenous NADH dehydrogenase, located outside the inner mitochondrial membrane, and linked to coupling site II. These mitochondria are able to oxidize exogenous NADH via two pathways: (1) a cyanide- and antimycin-sensitive pathway, or cytochrome pathway, and (2) a cyanide- and antimycin-insensitive pathway, or alternative pathway. Although the oxidation of exogenous NADH through the cytochrome pathway occurs with an ATP/0 ratio tending to 2, it proceeds, per molecule of NADH oxidized, with the apparent ejection in the outer medium of only 3 protons instead of 4 protons, as is the case with glycerol 3-phosphate as control substrate, but leaves 1 hydroxyl ion in the outer medium after decay of the protonmotive force. These properties were used to demonstrate the non electrogenic function of the alternative pathway. Indeed, the oxidation of exogenous NADH via the alternative pathway proceeds without apparent ejection of protons, although this oxidation generates an electron flux in the alternative pathway as demonstrated by the net appearance in the outer medium of 1 hydroxyl ion per atom of oxygen reduced, appearance which proves sensitive to benhydroxamic acid, a specific inhibitor of the alternative pathway. The non electrogenicity of the alternative pathway is accompanied by the absence of ATP synthesis as expected from Mitchell's chemiosmotic model. The absence of energy conservation when the electron transfer proceeds via the alternative pathway is not the result of an uncoupling property of an active alternative pathway, as the oxidation of malate plus pyruvate via coupling site I and the alternative pathway occurs with an ATP/0 ratio tending to 1.  相似文献
