A rapid and sensitive assay for the detection of lipase activity is described. The method is based upon the increase in absorbance at 360 nm due to the formation of the 2,4-dinitrophenolate anion during the enzymatic hydrolysis of 2,4-dinitrophenyl butyrate. The substrate is used in an emulsified form. Using a diode array spectrophotometer with internal referencing a correction can be made for absorbance changes due to clearance of the emulsion during hydrolysis. The small reaction volume and the high extinction coefficient of the product makes the method applicable for detection of both low substrate and low enzyme concentration.
Four lipases were tested: lipase from porcine pancreas, Candida cylindracea, Pseudomonas sp. and Aspergillus niger. All enzymes are readily able to catalyse the hydrolysis of 2,4-dinitrophenyl butyrate. 相似文献

The signal produced by fluorescence in situ hybridization (FISH) often is inconsistent among cells and sensitivity is low. Small DNA targets on the chromatin are difficult to detect. We report here an improved nick translation procedure for Texas red and Alexa Fluor 488 direct labeling of FISH probes. Brighter probes can be obtained by adding excess DNA polymerase I. Using such probes, a 30 kb yeast transgene, and the rp1, rp3 and zein multigene clusters were clearly detected. 相似文献

A goal of behavioral neuroscience is to identify underlying neurobiological factors that regulate specific behaviors. Using animal models to accomplish this goal, many methodological strategies require invasive techniques to manipulate the intensity of the behavior of interest (e.g., lesion methods, pharmacological manipulations, microdialysis techniques, genetically-engineered animal models). The utilization of a comparative species approach allows researchers to take advantage of naturally occurring differences in response strategies existing in closely related species. In our lab, we use two species of the Peromyscus genus that differ in paternal responses. The male California deer mouse (Peromyscus californicus) exhibits the same parental responses as the female whereas its cousin, the common deer mouse (Peromyscus maniculatus) exhibits virtually no nurturing/parental responses in the presence of pups. Of specific interest in this article is an exploration of the neurobiological factors associated with the affiliative social responses exhibited by the paternal California deer mouse. Because the behavioral neuroscience approach is multifaceted, the following key components of the study will be briefly addressed: the identification of appropriate species for this type of research; data collection for behavioral analysis; preparation and sectioning of the brains; basic steps involved in immunocytochemistry for the quantification of vasopressin-immunoreactivity; the use of neuroimaging software to quantify the brain tissue; the use of a microsequencing video analysis to score behavior and, finally, the appropriate statistical analyses to provide the most informed interpretations of the research findings. 相似文献

The Aschelminthes is a collection of at least eight animal phyla,
historically grouped together because the absence of a true body cavity was
perceived as a pseudocoelom. Analyses of 18S rRNA sequences from six
Aschelminth phyla (including four previously unpublished sequences) support
polyphyly for the Aschelminthes. At least three distinct groups of
Aschelminthes were detected: the Priapulida among the protostomes, the
Rotifera-Acanthocephala as a sister group to the protostomes, and the
Nematoda as a basal group to the triploblastic Eumetazoa.

Over evolutionary time, predator-prey interactions have shaped and constrained functional and behavioral traits of piscivorous fishes. The endangered Colorado Pikeminnow Ptychocheilus lucius, a large endemic piscivore of the Colorado River Basin, encounters a substantially altered prey base that differs in behaviors and morphologies compared to the historical suite of native prey. To assess physical limitations of Colorado Pikeminnow predation, we conducted a feeding experiment with two species of nonnative prey (spined and despined Channel Catfish Ictalurus punctatus and Red Shiner Cyprinella lutrensis) and quantified scaling of cranial morphology in this predator. In our predation experiments, Colorado Pikeminnow (215–312 mm total length) consumed both spined and despined Channel Catfish as well as Red Shiner but only consumed prey less than 20% of the predator’s total length. Previous feeding trials using smaller Colorado Pikeminnow, with native and nonnative prey species, indicated they consumed prey up to 35% of their total length, suggesting relative prey size limits may decrease as this predator grows. Morphological measurements also suggested relative prey size suitability may decrease as Colorado Pikeminnow become larger, with head depth and width demonstrating isometric scaling at small sizes and shifting to negative allometry as fish get larger. Together, these data suggest an ontogenetic shift in the head morphology of Colorado Pikeminnow may decrease the relative size of prey available to these predators. In severely altered systems, understanding trophic characteristics that limit overall predator resource availability will be critical for conservation of piscivorous fishes. 相似文献
