Physical mapping of plastid DNA variation among eleven Nicotiana species   总被引:1,自引:0,他引:1  
Summary Plastid DNA of seven American and four Australian species of the genus Nicotiana was examined by restriction endonuclease analysis using the enzymes Sal I, Bgl I, Pst I, Kpn I, Xho I, Pvu II and Eco RI. These endonucleases collectively distinguish more than 120 sites on N. tabacum plastid DNA. The DNAs of all ten species exhibited restriction patterns distinguishable from those of N. tabacum for at least one of the enzymes used. All distinctive sites were physically mapped taking advantage of the restriction cleavage site map available for plastid DNA from Nicotiana tabacum (Seyer et al. 1981). This map was extended for the restriction endonucleases Pst I and Kpn I. In spite of variation in detail, the overall fragment order was found to be the same for plastid DNA from the eleven Nicotiana species. Most of the DNA changes resulted from small insertions/deletions and, possibly, inversions. They are located within seven regions scattered along the plastid chromosome. The divergence pattern of the Nicotiana plastid chromosomes was strikingly similar to that found in the genus Oenothera subsection Euoenothera (Gordon et al. 1982). The possible role of replication as a factor in the evolution of divergence patterns is discussed. The restriction patterns of plastid DNA from species within a continent resembled each other with one exception in each instance. We administered antifibrotic agent beta-aminopropionitrile (BAPN) to rats exposed to 10% O2-90% N2 for 3 wk to prevent excess vascular collagen accumulation. Groups of Sprague-Dawley rats studied were air breathing, hypoxic, and hypoxic treated with BAPN, 150 mg/kg twice daily intraperitoneally. After the 3-wk period, we measured mean right ventricular pressure (RVP), the ratio of weight of right ventricle to left ventricle plus septum (RV/LV + S), and hydroxyproline content of the main pulmonary artery (PA) trunk. Hypoxia increased RVP from 14 to 29 mmHg; RVP was 21 mmHg in hypoxic BAPN-treated animals. Hypoxia increased the RV/LV + S ratio from 0.28 to 0.41; the ratio was 0.32 in hypoxic BAPN-treated animals. Hypoxia increased PA hydroxyproline from 20 to 239 micrograms/artery; hydroxyproline was 179 micrograms/artery in hypoxic BAPN-treated animals. Thus BAPN prevented pulmonary hypertension, right ventricular hypertrophy, and excess vascular collagen produced by hypoxia. We conclude that vascular collagen contributes to the maintenance of chronic hypoxic pulmonary hypertension.  相似文献

A particular monoclonal antibody that binds to the influenza virus HA molecule inhibits HA-specific thymus-derived lymphocytes mediating cytotoxicity in the context of H-2Dd but not of H-2Kd. Another monoclonal antibody blocks both sets of HA-specific effector T cells. This observation, together with related findings from other laboratories, is considered to support the idea that T cell recognition is directed against some association of viral and H-2 glycoproteins, as proposed in the original formulation of the "altered self" concept.  相似文献

Effect of acetazolamide on cerebral blood flow and capillary patency.   总被引:2,自引:0,他引:2  
This study investigated the effects 2 h after administration of acetazolamide on cerebral blood flow and the pattern of cerebral capillary perfusion. Arterial blood pressure, heart rate, arterial blood gases, and pH were recorded in two groups of rats along with either regional cerebral blood flow or the percentage of capillary volume per cubic millimeter and number per square millimeter perfused as determined in cortical, thalamic, pontine, and medullary regions of the brain. Blood pressure, heart rate, and arterial PCO2 were not significantly different between the rats receiving acetazolamide (100 mg/kg) and the controls. Arterial blood pH was significantly lower in the acetazolamide rats. Blood flow increased significantly in the cortical (+ 102%), thalamic (+ 89%), and pontine (+ 88%) regions receiving acetazolamide. In control rats, approximately 60% of the capillaries were perfused in all of the examined regions. The percentage of capillaries per square millimeter perfused was significantly greater in the cortical (+ 52%), thalamic (+ 49%), and pontine (+ 47%) regions of acetazolamide rats compared with controls. In the medulla the increases in blood flow and percentage of capillaries perfused were not significant. Thus in the regions that acetazolamide increased cerebral blood flow, it also increased the percentage of capillaries perfused.  相似文献

Thirty endogenous proviruses belonging to the modified polytropic (Mpmv) class of murine leukemia virus (MLV) were identified by proviral-cellular DNA junction fragment segregation in several sets of recombinant inbred mice. Twenty-six Mpmv loci were mapped to chromosomal regions by matching proviral strain distribution patterns to those of previously assigned genes. Like other endogenous nonecotropic MLVs, Mpmv loci were present on several chromosomes in all strains examined. We pooled recombinant inbred strain linkage data from 110 MLV loci and selected marker genes in order to construct a chromosomal linkage map. Every mouse chromosome was found to harbor at least one proviral insertion, and several regions contained multiple integrations. However, the overall distribution of the 110 mapped proviruses did not deviate significantly from a random distribution. Because of their polymorphism in inbred strains of mice, and the ability to score as many as 57 proviruses per strain using only three hybridization probes, the nonecotropic MLVs mapped in common strains of mice offer a significant advantage over older methods (e.g., biochemical or individual restriction fragment polymorphisms) as genetic markers. These endogenous insertion elements should also be useful for assessing strain purity, and for studying the relatedness of common and not-so-common inbred strains.  相似文献

The cell surface of Tetrahymena thermophila is made up of an anterior region in which virtually all basal bodies of ciliary rows are ciliated, and the remainder in which ciliated and unciliated basal bodies are fairly irregularly interspersed. This pattern persists through interfission development until the stage of appearance of the equatorial ring of gaps in the ciliary rows that marks the fission zone. The ciliation pattern then becomes subdivided, in large part through the rapid ciliation of contiguous basal bodies located posterior to the fission zone. We interpret this process as a wave of ciliation of preexisting basal bodies that propagates posteriorly from the site of the fission zone. The location, extent, and timing of the ciliation process are the same in inverted as in normally oriented ciliary rows, in spite of the fact that in inverted rows the visible fission zone gap is tardily formed and the local configuration of ciliature around this gap is abnormal. The putative ciliation wave thus does not depend directly upon the local manifestations of the fission zone. However, in a cell-division-arrest mutant, cdaA1, analyzed under conditions in which formation of fission-zone gaps is permanently prevented in some ciliary rows but not in all, it is found that the ciliation pattern becomes subdivided in those ciliary rows that express fission-zone gaps and fails to become subdivided in neighboring rows that fail to manifest gaps. We interpret this combination of findings to indicate that a signal localized at the cell equator initiates a set of polarized developmental events that simultaneously create and demarcate two cellular fields within what was previously one. We further suggest that the characteristic tandem cell division pattern of ciliates is fundamentally a process of segmentation, which might involve mechanisms of gradient subdivision analogous to those taking place during segmentation of insects and other multicellular organisms.  相似文献

A total of 65 Schizophyllum commune dikaryons of three different genotypes were synthesized, maintained on agar plates, prevented from fruiting, and macerated. The macerate was plated. Germlings of homokaryotic morphology were isolated using no chemical or genetic selection, grown, and tested for recombinant genotypes. Crossover types were plentiful among the homokaryons with recombinant genotypes, indicating that meiotic-like processes in vegetative dikaryons are common and may be as important as conventional basidial meiosis in the origin of recombinant strains.  相似文献
