Summary The permeability and partition coefficients of tetraphenylarsonium (TPA) and several other organic cations were studied in the human erythrocyte using an ion-selective electrode. The permeability constant for the different cations could be explained quite well by differences in oil/water partition coefficients. No evidence for facilitated transport could be found. Binding of the organic ions occurred to both the cell membrane and to intracellular contents. Partitioning to the membrane remained relatively constant despite variation from ion intracellular binding with blood samples from different donors. TPA flux is stimulated by substoichiometric amounts of tetraphenylboron and other organic anions, suggesting an ion-pairing mechanism. 相似文献

Encompassing some of the major hotspots of biodiversity on Earth, large mountain systems have long held the attention of evolutionary biologists. The region of the Qinghai‐Tibet Plateau (QTP) is considered a biogeographic source for multiple colonization events into adjacent areas including the northern Palearctic. The faunal exchange between the QTP and adjacent regions could thus represent a one‐way street (“out of” the QTP). However, immigration into the QTP region has so far received only little attention, despite its potential to shape faunal and floral communities of the QTP. In this study, we investigated centers of origin and dispersal routes between the QTP, its forested margins and adjacent regions for five clades of alpine and montane birds of the passerine superfamily Passeroidea. We performed an ancestral area reconstruction using BioGeoBEARS and inferred a time‐calibrated backbone phylogeny for 279 taxa of Passeroidea. The oldest endemic species of the QTP was dated to the early Miocene (ca. 20 Ma). Several additional QTP endemics evolved in the mid to late Miocene (12–7 Ma). The inferred centers of origin and diversification for some of our target clades matched the “out of Tibet hypothesis’ or the “out of Himalayas hypothesis” for others they matched the “into Tibet hypothesis.” Three radiations included multiple independent Pleistocene colonization events to regions as distant as the Western Palearctic and the Nearctic. We conclude that faunal exchange between the QTP and adjacent regions was bidirectional through time, and the QTP region has thus harbored both centers of diversification and centers of immigration. 相似文献

Female rats with complete transections of spinal cord at low thoracic levels did not perform lordosis. After treatment with quipazine, a serotonin receptor stimulant, some spinal rats were able to show hindquarter responses which resembled parts of the lordosis reflex. Five rumen bacteria, Selenomonas ruminantium, Bacteroides ruminicola, Megasphaera elsdenii, Butyrivibrio fibrisolvens, and Streptococcus bovis were grown in media containing nonlimiting concentrations of glucose, sucrose, maltose, cellobiose, xylose and/or lactate. Each bacterium was grown with every substrate that it could ferment in every possible two-way combination. Only once did a combination of substrates result in a higher maximum growth rate than that observed with either substrate alone. Such stimulations of growth rate would be expected if specific factors unique to individual substrates (transport proteins and/or enzymes) were limiting. Since such synergisms were rare, it was concluded that more general factors limit maximum growth rates in these five bacteria. 相似文献

Heterotrophic Uptake Experiments with 14C-Labeled Histidine in a Histidine-Limited Chemostat 下载免费PDF全文
The histidine uptake by bacterial strain HIS 42 was determined with [U-14C]histidine and through oxygen uptake experiments on samples taken from a histidine-limited chemostat. The uptake of [U-14C]histidine was characterized by a saturation constant of 12.8 to 78.6 nM histidine. At higher growth rates, the measured maximum uptake rate of histidine was lower than the actual uptake rate in the culture. The percentage of respired substrate (76 to 93%) was about 30 to 40% higher than the comparable value for the culture. The uptake of histidine as analyzed through the measurement of oxygen uptake rates was characterized by a saturation constant of 1.7 to 10.5 μM histidine; the maximum uptake rate was always greater than the actual histidine uptake rate in the culture. By the application of the two cited methods, set up to determine the histidine uptake kinetics, two different uptake processes were analyzed. It appeared that the determination of the histidine uptake through measurement of the oxygen uptake rate showed a better reflection of the actual uptake process of histidine in the culture. With the available data it was impossible to assess a correlation between the uptake of histidine, as determined with [U-14C]histidine, and the actual metabolism of the bacterial population. 相似文献

We report the purification of a presynaptic "particle web" consisting of approximately 50 nm pyramidally shaped particles interconnected by approximately 100 nm spaced fibrils. This is the "presynaptic grid" described in early EM studies. It is completely soluble above pH 8, but reconstitutes after dialysis against pH 6. Interestingly, reconstituted particles orient and bind PSDs asymmetrically. Mass spectrometry of purified web components reveals major proteins involved in the exocytosis of synaptic vesicles and in membrane retrieval. Our data support the idea that the CNS synaptic junction is organized by transmembrane adhesion molecules interlinked in the synaptic cleft, connected via their intracytoplasmic domains to the presynaptic web on one side and to the postsynaptic density on the other. The CNS synaptic junction may therefore be conceptualized as a complicated macromolecular scaffold that isostatically bridges two closely aligned plasma membranes. 相似文献

Subcellular distribution of 14-3-3 proteins in the unicellular green alga Chlamydomonas reinhardtii.
A polyclonal antibody was raised against a recombinant Chlamydomonas 14-3-3-beta-galactosidase (beta-Gal) fusion protein and characterized for its epitope specificity towards the corresponding Chlamydomonas 14-3-3 protein by scan-peptide analysis. This antibody recognized four Chlamydomonas polypeptides with apparent molecular masses 32, 30, 27, and 24 kDa, which also reacted with the antiserum depleted of anti-(Escherichia coli beta-Gal) IgG, but not with the corresponding preimmune serum or the antiserum preincubated with purified 14-3-3 proteins. Western-blot analyses performed with the antibody depleted of anti-(beta-Gal) IgG revealed that more or less pronounced levels of 14-3-3 proteins were present in all subcellular fractions of Chlamydomonas reinhardtii except the nuclei. The highest levels of 14-3-3 protein were observed in the cytosol and microsomal fraction. The 30-kDa isoform was predominant in the cytosol, whereas the 27-kDa isoform was prevalent in the microsomes. When microsomal membranes were separated by sucrose-density-gradient centrifugation, Western-blot analysis revealed distinct patterns of 14-3-3 isoforms in the endoplasmic reticulum, dictyosome, and plasma membrane fractions identified by marker enzyme activities. These findings indicate that the four 14-3-3 proteins of C. reinhardtii differentially interact with endoplasmic reticulum, dictyosomes, and plasma membrane. 相似文献

Two experiments were carried out to study the effects of boron (B) deficiency on 7-day-old pea plants for 6 or 9 days under controlled growth chamber conditions. Growth and apical dominance (AD) of the plants and their B concentration and compartmentation were followed throughout the starvation period. Additionally, auxin (indoleacetic acid, IAA) concentration in the shoot apex and polar transport from it were measured along with the cytokinin (CK) concentration in the shoot apex and the roots. The results demonstrate that during a 6-day B-deficiency period, B concentration in the water-insoluble residue of the roots was very stable and could not easily be reduced. In contrast, B concentration in the cell sap fraction was very sensitive to external B supply. Twelve hours after transferring the plants from B-sufficient to B-deficient solutions, the B concentration in root cell sap declined to half the concentration of the control plants. In addition, B concentration in the new aerial plant parts, which developed after the onset of the B-deficiency treatment, was extremely low. A decline in elongation growth could be observed as soon as about 4 days after the imposition of B deficiency. This preceded the first measurable growth of lateral buds (release from AD). Before the onset of these morphological changes, there was a considerable decline in CK concentration, accompanied by a dramatic decrease in IAA export out of the shoot apex, a decline in IAA concentration in the shoot apex and the roots and a reduced capacity for polar IAA-transport. These changes are discussed as possible reasons for the observed reduction in elongation growth and AD. These hormonal changes themselves are possibly the result of the decreased symplasmic B concentration, which in turn may be responsible for the reduced concentration in apical CKs. A sequence of events, which may be causally related, is suggested to explain the effects of B deficiency on the growth and AD of pea plants. 相似文献
