We have investigated the importance of carotenoids on the accumulation and function of the photosynthetic apparatus using a mutant of the green alga Chlamydomonas reinhardtii lacking carotenoids. The FN68 mutant is deficient in phytoene synthase, the first enzyme of the carotenoid biosynthesis pathway, and therefore is unable to synthesize any carotenes and xanthophylls. We find that FN68 is unable to accumulate the light-harvesting complexes associated with both photosystems as well as the RC subunits of photosystem II. The accumulation of the cytochrome b6f complex is also strongly reduced to a level approximately 10% that of the wild type. However, the residual fraction of assembled cytochrome b6f complexes exhibits single-turnover electron transfer kinetics comparable to those observed in the wild-type strain. Surprisingly, photosystem I is assembled to significant levels in the absence of carotenoids in FN68 and possesses functional properties that are very similar to those of the wild-type complex.Carotenoids (Cars) are fundamental components of the photosynthetic apparatus (Young and Britton, 1993, and refs. therein). The vast majority of Cars are noncovalently bound to either the core or the antenna subunits of PSI or PSII (Siefermann-Harms, 1985; Bassi et al., 1993). The most abundant Car bound to the core subunits of both photosystems is β-carotene, which is found in the vast majority of oxygenic organisms (Siefermann-Harms, 1985; Bassi et al., 1993). The light-harvesting complexes (LHCs) that act as the outer antenna in plants and green algae bind a wider range of oxygenated Cars, known as xanthophylls, the most abundant of which is lutein (Siefermann-Harms, 1985; Bassi et al., 1993; Jennings et al., 1996). The stoichiometry of xanthophylls binding to LHC complexes depends on the particular complexes and often on the illumination conditions during the organism’s growth (Siefermann-Harms, 1985; Demmig-Adams, 1990; Horton et al., 1996). Intriguingly, a molecule of β-carotene (as well as a molecule of chlorophyll [Chl] a) is found also in the cytochrome (Cyt) b6f complex (Kurisu et al., 2003; Stroebel et al., 2003).Cars have multiple functions in the photosynthetic process; they act as light-harvesting pigments (Frank and Cogdell, 1993), enlarging the optical cross section to radiation that is poorly absorbed by Chl. Moreover, Cars play a crucial role in processes such as nonphotochemical quenching that control the efficiency of light harvesting in response to the intensity of the incident radiation (for review, see Demmig-Adams, 1990; Horton et al., 1996; Niyogi, 1999). Probably the most important role of Cars in photosynthesis is the quenching of the excited triplet state of Chl (for review, see Frank and Cogdell, 1993; Giacometti et al., 2007), preventing the formation of highly reactive singlet oxygen, which represents the principal species active under high light stress (Hideg et al., 1994; Krieger-Liszkay, 2005). The importance of Cars is demonstrated by the observation that disruption of their biosynthesis through mutation, or by inhibition of a key enzyme in the pathway, leads to either lethal phenotypes or to rapid photobleaching of the photosynthetic tissue (Claes, 1957; Faludi-Dániel et al., 1968, 1970; Bolychevtseva et al., 1995; Trebst and Depka, 1997).Moreover, it has been shown that the presence of xanthophylls is absolutely necessary for refolding in vitro of LHC I and LHC II antenna complexes (Plumley and Schmidt, 1987; Paulsen et al., 1993; Sandonà et al., 1998). Such Cars, therefore, have a structural role, as well as their involvement in light harvesting, nonphotochemical quenching regulation, and the quenching of the Chl triplet state. Whether Cars also play a key structural role in the formation and stability of the core complexes of both PSI and PSII has not been systematically explored, since assembly of these complexes in vitro is not feasible. Studies in vivo using higher plants are complicated by the fact that Car deficiency is lethal and can be studied only during the early stages of greening and leaf development (Faludi-Dániel et al., 1968, 1970; Inwood et al., 2008). In these studies, it was shown that the accumulation of PSII complexes was greatly impaired in mutants of maize (Zea mays; Faludi-Dániel et al., 1968, 1970; Inwood et al., 2008), while the assembly of PSI appeared to be less sensitive to Car availability. In mutants of the cyanobacterium Synechocystis sp. PCC 6803 lacking the genes for phytoene desaturase or ζ-carotene desaturase, there was a complete loss of PSII assembly, while functional PSI complexes were assembled, albeit with slightly altered electron transfer kinetics with respect to the wild-type complex (Bautista et al., 2005). In agreement with the higher sensitivity of PSII assembly to Car availability, Trebst and Depka (1997) reported a specific effect on the synthesis of the D1 subunit of PSII RC upon treatment with phytoene desaturase inhibitors. On the other hand, it has recently been reported that in lycopene-β-cyclase mutants of Arabidopsis (Arabidopsis thaliana) that have a decreased amount of β-carotene (bound to the RC) with respect to most of the xanthophyll pool pigments (bound to the LHCs), the level of accumulation of PSI complexes, particularly that of the LHC I complement, was more affected that that of PSII, probably also because of an increased sensitivity to photodamage of mutated PSI RC (Cazzaniga et al., 2012; Fiore et al., 2012).In this investigation, we have studied the accumulation and functionality of the major chromophore-binding complexes of the photosynthetic apparatus, PSI, PSII, and Cyt b6f, in a Car-less mutant of the green alga Chlamydomonas reinhardtii (FN68) that is blocked at the first committed step of Car biosynthesis, namely, phytoene synthesis (McCarthy et al., 2004). Although the mutant is incapable of growing under phototrophic or photomixotrophic conditions, it can grow in complete darkness on a medium supplemented with a carbon source. Here, we show that the PSII core and antenna complexes fail to accumulate in the mutant and that the Cyt b6f complex accumulates to approximately one-tenth of the wild-type level. On the other hand, the PSI reaction center accumulates in FN68 and possesses electron transfer properties that are remarkably similar to those of wild-type PSI. Interestingly, we find that the level of PSI accumulation differs in other phytoene synthase null mutants, suggesting that additional mutations in one or other of these strains affect PSI stability. Nevertheless, our findings demonstrate that Cars are not required for either the assembly or the functionality of PSI in vivo.  相似文献

Members of the Psb28 family of proteins are accessory factors implicated in the assembly and repair of the photosystem II complex. We present here the crystal structure of the Psb28 protein (Tlr0493) found in the thermophilic cyanobacterium Thermosynechococcus elongatus at a resolution of 2.3 Å. Overall the crystal structure of the Psb28 monomer is similar to the solution structures of C-terminally His-tagged Psb28-1 from Synechocystis sp. PCC 6803 obtained previously by nuclear magnetic resonance spectroscopy. One new aspect is that Escherichia coli-expressed T. elongatus Psb28 is able to form dimers in solution and packs as a dimer of dimers in the crystal. Analysis of wild type and mutant strains of Synechocystis 6803 by blue native-polyacrylamide gel electrophoresis suggests that Psb28-1, the closest homologue to T. elongatus Psb28 in this organism, also exists as an oligomer in vivo, most likely a dimer. In line with the prediction based on the crystal structure of T. elongatus Psb28, the addition of a 3× Flag-tag to the C-terminus of Synechocystis 6803 Psb28-1 interferes with the accumulation of the Psb28-1 oligomer in vivo. In contrast, the more distantly related Psb28-2 protein found in Synechocystis 6803 lacks the residues that stabilize dimer formation in the T. elongatus Psb28 crystal and is detected as a monomer in vivo. Overall our data suggest that the dimer interface in the Psb28 crystal might be physiologically relevant.  相似文献

The role of human intraepithelial Vδ1+ γδ T cell cytotoxic effectors in the immune surveillance against metastatic colon cancer has never been addressed, despite their reported capacity to infiltrate colon carcinomas and to kill colonic cancer cells in vitro. We previously showed that Vδ1+ γδ T cells are enriched in blood in response to cytomegalovirus (CMV) infection, and that such increase may be protective against epithelial cancers. The objective of the present study was to investigate whether CMV-induced Vδ1+ γδ T lymphocytes could inhibit the propagation of human colon tumors in vivo, in order to evaluate their immunotherapeutic potential in this context. Even though metastases are an important cause of death in various cancers including colorectal cancer (CRC), the anti-metastatic effect of immune effectors has been poorly analyzed. To this purpose, we set up a reliable model of metastatic colon cancer through orthotopic implantation of luciferase-expressing human HT29 cells in immunodeficient mice. Using bioluminescence imaging to follow the outcome of colonic cancer cells, we showed that a systemic treatment with CMV-induced Vδ1+ γδ T cells could not only inhibit primary colon tumor growth but also the emergence of secondary tumor foci in the lungs and liver. Finally, our data lead to propose that Vδ1+ γδ T lymphocytes may directly influence the appearance of metastases independently from their control of primary tumor size. These findings, which extend our previous work, pave the road for the potential manipulation of Vδ1+ γδ T lymphocytes in novel anti-CRC immunotherapeutic protocols.  相似文献

The Ecouflant area (Maine-et-Loire, France) shows the last outcrops from the Anjou region that allow the study of early Cenomanian lignites and the associated amber. The quarry of Le Brouillard was historically the most studied locality, but it is now partly covered by bulky waste. No fossil arthropod has been found in the amber collected in this locality, and only a few bacterial filaments and plant fragments were detected among the microinclusions. The quarry of Hucheloup shows a wider exposure. Amber is poor, but fossil plants and bivalve are frequent. Based on the sedimentological series and the palaeontological contents of the lignitic clay, we suggest that it corresponds to estuarine to coastal depositional environments with variable salinity.  相似文献

The neutral theory of biodiversity challenges the classical niche-based view of ecological communities, where species attributes and environmental conditions jointly determine community composition. Functional equivalence among species, as assumed by neutral ecological theory, has been recurrently falsified, yet many patterns of tropical tree communities appear consistent with neutral predictions. This may mean that neutral theory is a good first-approximation theory or that species abundance data sets contain too little information to reject neutrality. Here we present a simple test of neutrality based on species abundance distributions in ecological communities. Based on this test, we show that deviations from neutrality are more frequent than previously thought in tropical forest trees, especially at small spatial scales. We then develop a nonneutral model that generalizes Hubbell's dispersal-limited neutral model in a simple way by including one additional parameter of frequency dependence. We also develop a statistical method to infer the parameters of this model from empirical data by approximate Bayesian computation. In more than half of the permanent tree plots, we show that our new model fits the data better than does the neutral model. Finally, we discuss whether observed deviations from neutrality may be interpreted as the signature of environmental filtering on tropical tree species abundance distributions.  相似文献

Although metacommunity ecology has been a major field of research in the last decades, with both conceptual and empirical outputs, the analysis of the temporal dynamics of metacommunities has only emerged recently and consists mostly of repeated static analyses. Here we propose a novel analytical framework to assess metacommunity processes using path analyses of spatial and temporal diversity turnovers. We detail the principles and practical aspects of this framework and apply it to simulated datasets to illustrate its ability to decipher the respective contributions of entangled drivers of metacommunity dynamics. We then apply it to four empirical datasets. Empirical results support the view that metacommunity dynamics may be generally shaped by multiple ecological processes acting in concert, with environmental filtering being variable across both space and time. These results reinforce our call to go beyond static analyses of metacommunities that are blind to the temporal part of environmental variability.  相似文献

Background

The analysis of humoral responses directed against the saliva of blood-sucking arthropods was shown to provide epidemiological biomarkers of human exposure to vector-borne diseases. However, the use of whole saliva as antigen presents several limitations such as problems of mass production, reproducibility and specificity. The aim of this study was to design a specific biomarker of exposure to tsetse flies based on the in silico analysis of three Glossina salivary proteins (Ada, Ag5 and Tsgf1) previously shown to be specifically recognized by plasma from exposed individuals.

Methodology/Principal Findings

Synthetic peptides were designed by combining several linear epitope prediction methods and Blast analysis. The most specific peptides were then tested by indirect ELISA on a bank of 160 plasma samples from tsetse infested areas and tsetse free areas. Anti-Tsgf118–43 specific IgG levels were low in all three control populations (from rural Africa, urban Africa and Europe) and were significantly higher (p<0.0001) in the two populations exposed to tsetse flies (Guinean HAT foci, and South West Burkina Faso). A positive correlation was also found between Anti-Tsgf118–43 IgG levels and the risk of being infected by Trypanosoma brucei gambiense in the sleeping sickness foci of Guinea.

Conclusion/Significance

The Tsgf118–43 peptide is a suitable and promising candidate to develop a standardize immunoassay allowing large scale monitoring of human exposure to tsetse flies in West Africa. This could provide a new surveillance indicator for tsetse control interventions by HAT control programs.  相似文献

In French Guiana, from 1984 to 2011, 14 animal rabies cases and 1 human rabies case (2008) were diagnosed. In January 2011, vampire-bat attacks occurred in 2 isolated villages. In mid-January, a medical team from the Cayenne Centre for Anti-Rabies Treatment visited the sites to manage individuals potentially exposed to rabies and, in April, an anti-rabies vaccination campaign for dogs was conducted. Twenty individuals were bitten by bats in 1 month, most frequently on the feet. The median time to start management was 15 days. The complete Zagreb vaccination protocol (2 doses on day 0 and 1 dose on days 7 and 21) was administered to 16 patients, 12 also received specific immunoglobulins. The antibody titration was obtained for 12 patients (different from those who received immunoglobulins). The antibody titers were ≥0.5 EU/mL for all of them. The serology has not been implemented for the 12 patients who received immunoglobulins. Accidental destruction of a vampire-bat colony could be responsible for the attacks. The isolation and absence of sensitization of the populations were the main explanations for the management difficulties encountered. Sensitization programs should be conducted regularly.  相似文献
