Experimental data suggest that halothane anesthesia is associated with significant changes in dopamine (DA) concentration
in some brain regions but the mechanism of this effect is not well known. Rat brain cortical slices were labeled with [3H]DA to further characterize the effects of halothane on the release of this neurotransmitter from the central nervous system.
Halothane induced an increase on the release of [3H]DA that was dependent on incubation time and anesthetic concentration (0.012, 0.024, 0.048, 0.072 and 0.096 mM). This effect
was independent of extracellular or intracellular calcium. In addition, [3H]DA release evoked by halothane was not affected by TTX (blocker of voltage-dependent Na+ channels) or reserpine (a blocker of vesicular monoamine transporter). These data suggest that [3H]DA release induced by halothane is non-vesicular and would be mediated by the dopamine transporter (DAT) and norepinephrine
transporter (NET). GBR 12909 and nomifensine, inhibitors of DAT, decreased the release of [3H]DA evoked by halothane. Nisoxetine, a blocker of NET, reduced the release of [3H]DA induced by halothane. In addition, GBR 12909, nisoxetine and, halothane decrease the uptake of [3H]DA into rat brain cortical slices. A decrease on halothane-induced release of [3H]DA was also observed when the brain cortical slices were incubated at low temperature and low extracellular sodium, which
are known to interfere with the carrier-mediated release of the neurotransmitter. Ouabain, a Na+/K+ ATPase pump inhibitor, which induces DA release through reverse transport, decreased [3H]DA release induced by halothane. It is suggested that halothane increases [3H]DA release in brain cortical slices that is mediated by DAT and NET present in the plasma membrane. 相似文献

Two new species of Dalyellidae, Dalyellia callvucurai n. sp. and Gieysztoria namuncurai n. sp., are described from temporary freshwater environments in central Buenos Aires Province, Argentina. The ultrastructure of the stylets of both species is also described. The new species of the genus Dalyellia is the second species of the genus found in the neotropics and the first whose stylet has been studied with scanning electron microscopy. Gieysztoria namuncurai n. sp. joins the seven species of the genus known in Argentina. The ultrastructural characteristics of the stylet place it in the Inaequales group, with complex stylets. 相似文献

The development of strategies to ameliorate post-myocardial infarction (MI) remodeling and improve function continues to be
an area of clinical importance. Use of steroids for this purpose is controversial since the effects of timed treatment on
relevant inflammatory, biochemical and structure/function endpoints are unclear. In a previous report, we demonstrated that
use of doxycycline pre-treatment improves post-MI remodeling and passive left ventricular (LV) function. However, the effects
of timed doxycyline post-MI treatment are unknown. To examine these issues, we performed a study using a rat MI model. Animals
were administered one of the following: doxycycline (DOX), the corticosteroid methylprednisolone (MP), or aqueous vehicle.
Treatment was given early, short-term (at time of MI to 24 h post-MI) or late, long term (2–7 days post-MI). Animals were
sacrificed at 3, 7 or 42 days post-surgery. We assessed LV hemodynamics, pressure–volume, and pressure–scar strains, histomorphometry,
inflammation via measurements of myeloperoxidase activity, and matrix metalloproteinase (MMP) activity. Late MP treatment
yielded a robust right-shifted pressure–volume curve, which was accompanied by increased scar strains. Late DOX treatment
yielded reduced average heart weight and size and preserved scar thickness. DOX treatment did not suppress inflammation, which
contrasts with the suppressive effects of MP. Use of early or late MP yielded increased MMP activity in infarcted and non-infarcted
regions. Early and late treatment with DOX yielded infarct–associated MMP activity levels comparable to those of vehicle–treated
animals. In conclusion, results indicate that late use of MP yields adverse post-MI structure/function outcomes that correlate
with suppression of inflammation and increased MMP activity. These observations contrast with those of DOX, in particular,
late treatment where improved outcomes were observed in LV structure and were accompanied by the lack of suppression of inflammation. 相似文献

Botrytis cinerea is a phytopathogenic fungus causing disease in a substantial number of economically important crops. In an attempt to identify putative fungal virulence factors, the two-dimensional gel electrophoresis (2-DE) protein profile
from two B. cinerea strains differing in virulence and toxin production were compared. Protein extracts from fungal mycelium obtained by tissue
homogenization were analyzed. The mycelial 2-DE protein profile revealed the existence of qualitative and quantitative differences
between the analyzed strains. The lack of genomic data from B. cinerea required the use of peptide fragmentation data from MALDI-TOF/TOF and ESI ion trap for protein identification, resulting
in the identification of 27 protein spots. A significant number of spots were identified as malate dehydrogenase (MDH) and
glyceraldehyde-3-phosphate dehydrogenase (GAPDH). The different expression patterns revealed by some of the identified proteins
could be ascribed to differences in virulence between strains. Our results indicate that proteomic analysis are becoming an
important tool to be used as a starting point for identifying new pathogenicity factors, therapeutic targets and for basic

BACKGROUND: A huge variety of plant forms can be found in nature. This is particularly noticeable for inflorescences, the region of the plant that contains the flowers. The architecture of the inflorescence depends on its branching pattern and on the relative position where flowers are formed. In model species such as Arabidopsis thaliana or Antirrhinum majus the key genes that regulate the initiation of flowers have been studied in detail and much is known about how they work. Studies being carried out in other species of higher plants indicate that the homologues of these genes are also key regulators of the development of their reproductive structures. Further, changes in these gene expression patterns and/or function play a crucial role in the generation of different plant architectures. SCOPE: In this review we aim to present a summarized view on what is known about floral initiation genes in different plants, particularly dicotyledonous species, and aim to emphasize their contribution to plant architecture. 相似文献

BACKGROUND AND AIMS: Atriplex (Halimione) portulacoides is a halophytic, C(3) shrub. It is virtually confined to coastal salt marshes, where it often dominates the vegetation. The aim of this study was to investigate its growth responses to salinity and the extent to which these could be explained by photosynthetic physiology. METHODS: The responses of young plants to salinity in the range 0-700 mol m(-3) NaCl were investigated in a glasshouse experiment. The performance of plants was examined using classical growth analysis, measurements of gas exchange (infrared gas analysis), determination of chlorophyll fluorescence characteristics (modulated fluorimeter) and photosynthetic pigment concentrations; total ash, sodium, potassium and nitrogen concentrations, and relative water content were also determined. KEY RESULTS: Plants accumulated Na(+) approximately in proportion to external salinity. Salt stimulated growth up to an external concentration of 200 mol m(-3) NaCl and some growth was maintained at higher salinities. The main determinant of growth response to salinity was unit leaf rate. This was itself reflected in rates of CO(2) assimilation, which were not affected by 200 mol m(-3) but were reduced at higher salinities. Reductions in net photosynthetic rate could be accounted for largely by lower stomatal conductance and intercellular CO(2) concentration. Apart from possible effects of osmotic shock at the beginning of the experiment, salinity did not have any adverse effect on photosystem II (PSII). Neither the quantum efficiency of PSII (Phi(PSII)) nor the chlorophyll fluorescence ratio (F(v)/F(m)) were reduced by salinity, and lower mid-day values recovered by dawn. Mid-day F(v)/F(m) was in fact depressed more at low external sodium concentration, by the end of the experiment. CONCLUSIONS: The growth responses of the hygro-halophyte A. portulacoides to salinity appear largely to depend on changes in its rate of photosynthetic gas exchange. Photosynthesis appears to be limited mainly through stomatal conductance and hence intercellular CO(2) concentration, rather than by effects on PSII; moderate salinity might stimulate carboxylation capacity. This is in contrast to more extreme halophytes, for which an ability to maintain leaf area can partially offset declining rates of carbon assimilation at high salinity. 相似文献

BACKGROUND AND AIMS: The genus Melocactus comprises 36 species of globose cacti with the most derived traits in the Cereeae tribe. It is the proper study system to examine what are the most derived reproductive strategies within that tribe. This study aims to characterize the reproductive biology and to estimate the mating system parameters of two Andean melocacti, Melocactus schatzlii and M. andinus. METHODS: The reproductive attributes of the two species were described, including floral morphology, anthesis patterns, floral rewards, floral visitors and visitation patterns. Levels of self-compatibility and autonomous self-pollination were estimated by hand-pollination experiments. Mating system estimates were obtained by conducting progeny array analyses using isozymes. KEY RESULTS: The flowers of the two species present the typical hummingbird-pollination syndrome. Despite their morphological resemblance, the two species differ in flower size, pollen and ovule production and anthesis pattern. Their main pollinator agents are hummingbirds, four species in M. schatzlii and one species in M. andinus. Both cacti are self-compatible and capable of self-pollination without the aid of pollen vectors. Population-level outcrossing rate was higher for M. schatzlii (t(m)=0.9) than for M. andinus (t(m)=0.4). At the family level, outcrossing rates for most mothers of M. schatzlii were higher (t(m)>0.8) than for M. andinus (t(m)<0.5). CONCLUSIONS: Although the two cacti are capable of selfing, M. schatzlii is a predominantly outcrossing species, while M. andinus behaves as a mixed-mating cactus. Hummingbirds are the only pollinators responsible for outcrossing and gene flow events in these species. In their absence, both melocacti set seeds by selfing. Based on its low population size, restricted distribution in Venezuela, low rates of floral visits, and high levels of inbreeding, M. andinus is considered to be an endangered species deserving further study to define its conservation status. 相似文献

The study of the interaction of biosurfactants with biological membranes is of great interest in order to gain insight into the molecular mechanisms of their biological actions. In this work we report on the interaction of a bacterial trehalose lipid produced by Rhodococcus sp. with phosphatidylcholine membranes. Differential scanning calorimetry measurements show a good miscibility of the glycolipid in the gel state and immiscibility in the fluid state, suggesting domain formation. These domains have been visualized and characterized, for the first time, by scanning force microscopy. Incorporation of trehalose lipid into phosphatidylcholine membranes produces a small shift of the antisymmetric stretching band toward higher wavenumbers, as shown by FTIR, which indicates a weak increase in fluidity. The CO stretching band shows that incorporation of trehalose lipid increases the proportion of the dehydrated component in mixtures with the three phospholipids at temperatures below and above the gel to liquid-crystalline phase transition. This dehydration effect is also supported by data on the phospholipid PO stretching bands. Small-angle X-ray diffraction measurements show that in the samples containing trehalose lipid the interlamellar repeat distance is larger than in those of pure phospholipids. These results are discussed within the frame of trehalose lipid domain formation, trehalose lipid/phospholipid interactions and its relevance to membrane-related biological actions. 相似文献

Integrated analysis of a 50-m long sedimentary core collected in the central part of the Odiel estuary (SW Atlantic coast of Spain) allows delineation of the main paleoenvironmental changes that occurred in this area during the Holocene. Eight sedimentary facies were deposited in the last ca. 9000 years BP, confirming a transgressive-regressive cycle that involves the transition from fluvial to salt marsh deposits with intermediate marine tidal deposits. Disruption of the seqA gene of Salmonella enterica serovar Typhimurium causes defects similar to those described in E. coli: filament formation, aberrant nucleoid segregation, induction of the SOS response, envelope instability, and increased sensitivity to membrane-damaging agents. Differences between SeqA− mutants of E. coli and S. enterica, however, are found. SeqA− mutants of S. enterica form normal colonies and do not exhibit alterations in phage plaquing morphology. Lack of SeqA causes attenuation of S. enterica virulence by the oral route but not by the intraperitoneal route, suggesting a virulence defect in the intestinal stage of infection. However, SeqA− mutants are fully proficient in the invasion of epithelial cells. We hypothesize that attenuation of SeqA− mutants by the oral route may be caused by bile sensitivity, which in turn may be a consequence of envelope instability. 相似文献
