Proton-gated TASK-3 K+ channel belongs to the K2P family of proteins that underlie the K+ leak setting the membrane potential in all cells. TASK-3 is under cooperative gating control by extracellular [H+]. Use of recently solved K2P structures allows us to explore the molecular mechanism of TASK-3 cooperative pH gating. Tunnel-like side portals define an extracellular ion pathway to the selectivity filter. We use a combination of molecular modeling and functional assays to show that pH-sensing histidine residues and K+ ions mutually interact electrostatically in the confines of the extracellular ion pathway. K+ ions modulate the pKa of sensing histidine side chains whose charge states in turn determine the open/closed transition of the channel pore. Cooperativity, and therefore steep dependence of TASK-3 K+ channel activity on extracellular pH, is dependent on an effect of the permeant ion on the channel pHo sensors. 相似文献

Molecular dynamics simulations of triclinic hen egg white lysozyme in aqueous solution were performed to calculate the intrinsic pKas of 14 ionizable residues. An all-atom model was used for both solvent and solute, and a single 180 ps simulation in conjunction with a Gaussian fluctuation analysis method was used. An advantage of the Gaussian fluctuation method is that it only requires a single simulation of the system in a reference state to calculate all the pKas in the protein, in contrast to multiple simulations for the free energy perturbation method. pKint shifts with respect to reference titratable residues were evaluated and compared to results obtained using a finite difference Poisson-Boltzmann (FDPB) method with a continuum solvent model; overall agreement with the direction of the shifts was generally observed, though the magnitude of the shifts was typically larger with the explicit solvent model. The contribution of the first solvation shell to the total charging free energies of the titratable groups was explicitly evaluated and found to be significant. Dielectric shielding between pairs of titratable groups was examined and found to be smaller than expected. The effect of the approximations used to treat the long-range interactions on the pKint shifts is discussed. © 1994 Wiley-Liss, Inc. 相似文献

The striatum integrates motor behavior using a well‐defined microcircuit whose individual components are independently affected in several neurological diseases. The glial cell line‐derived neurotrophic factor (GDNF), synthesized by striatal interneurons, and Sonic hedgehog (Shh), produced by the dopaminergic neurons of the substantia nigra (DA SNpc), are both involved in the nigrostriatal maintenance but the reciprocal neurotrophic relationships among these neurons are only partially understood. To define the postnatal neurotrophic connections among fast‐spiking GABAergic interneurons (FS), cholinergic interneurons (ACh), and DA SNpc, we used a genetically induced mouse model of postnatal DA SNpc neurodegeneration and separately eliminated Smoothened (Smo), the obligatory transducer of Shh signaling, in striatal interneurons. We show that FS postnatal survival relies on DA SNpc and is independent of Shh signaling. On the contrary, Shh signaling but not dopaminergic striatal innervation is required to maintain ACh in the postnatal striatum. ACh are required for DA SNpc survival in a GDNF‐independent manner. These data demonstrate the existence of three parallel but interdependent neurotrophic relationships between SN and striatal interneurons, partially defined by Shh and GDNF. The definition of these new neurotrophic interactions opens the search for new molecules involved in the striatal modulatory circuit maintenance with potential therapeutic value. 相似文献

Auxin modulates a range of plant developmental processes including embryogenesis, organogenesis, and shoot and root development. Recent studies have shown that plant hormones also strongly influence metabolic networks, which results in altered growth phenotypes. Modulating auxin signalling pathways may therefore provide an opportunity to alter crop performance. Here, we performed a detailed physiological and metabolic characterization of tomato (Solanum lycopersicum) mutants with either increased (entire) or reduced (diageotropica—dgt) auxin signalling to investigate the consequences of altered auxin signalling on photosynthesis, water use, and primary metabolism. We show that reduced auxin sensitivity in dgt led to anatomical and physiological modifications, including altered stomatal distribution along the leaf blade and reduced stomatal conductance, resulting in clear reductions in both photosynthesis and water loss in detached leaves. By contrast, plants with higher auxin sensitivity (entire) increased the photosynthetic capacity, as deduced by higher Vcmax and Jmax coupled with reduced stomatal limitation. Remarkably, our results demonstrate that auxin‐sensitive mutants (dgt) are characterized by impairments in the usage of starch that led to lower growth, most likely associated with decreased respiration. Collectively, our findings suggest that mutations in different components of the auxin signalling pathway specifically modulate photosynthetic and respiratory processes. 相似文献

Paracoccidioidomycosis, a deep mycosis endemic in Latin America, is a chronic granulomatous disease caused by the fungus Paracoccidioides brasiliensis. Phagocytic cells play a critical role against this fungus, and several studies have shown the effects of activator and suppressive cytokines on macrophage and monocyte functions. However, studies on polymorphonuclear neutrophils (PMNs), that are the first cells recruited to the infection sites, are scarcer. Thus, the objective of this paper was to assess whether interleukin-10 (IL-10), a potent anti-inflammatory cytokine, is able to block the activity of IFN-gamma-activated human PMNs upon P. brasiliensis intracellular killing, in vitro. The results showed that IFN-gamma-activated PMNs have an effective fungicidal activity against the fungus. This activity was associated with the release of high levels of H(2)O(2), the metabolite involved in phagocytic cells antifungal activities. However, the concomitant incubation of these cells with IFN-gamma and IL-10 significantly blocked IFN-gamma activation. As a consequence, PMNs killing activity and H(2)O(2) release were inhibited. Together, our results show the importance of PMNs exposure to activator or suppressor cytokines in the early stages of paracoccidioidomycosis infection. 相似文献

Adolescence is marked by intensive growth and development. When pregnancy occurs during this period of the mother’s growth,
there is an increase in her nutritional needs. The aim of this study was to determine the levels of calcium, iron, copper,
and zinc in maternal plasma, the placenta, and in the cord plasma of pregnant teenagers and adults. A total of 80 sets of
maternal plasma, placentas, and cord plasma (40 from teenagers and 40 from adults) were analyzed using synchrotron radiation
total reflection X-ray fluorescence. The levels of calcium, copper, and zinc in the maternal and cord plasma from teenagers
were not significantly different than those from adults. Iron levels in the teenagers maternal and cord plasma were higher
than in the adults. All of the minerals analyzed were present at higher levels in the placentas from adults than those from
teenagers. However, the low quantities of placental calcium, iron, copper, and zinc in the teenagers do not compromise the
levels of these minerals in the cord plasma. Future research regarding the placental transport of these minerals is recommended
to investigate the efficiency of mechanisms of transfer of these minerals in pregnant teenagers. 相似文献

Dense concentrations of juvenile ammonoids were recently discovered in the Upper Cretaceous Mancos Shale of east-central Utah. In this paper, we describe this remarkable fossil occurrence and propose a taphonomic model to explain it. Large accumulations of cephalopods are not uncommon in the stratigraphic record, though concentrations of juveniles are relatively rare. Lithologic, geochemical, and stratigraphic evidence suggest that the unusual fossil occurrence we document here resulted from a combination of factors. We think the cause of these dense concentrations of juvenile ammonoids involves the hypothesized semelparous reproductive strategy of ammonoids, environmentally driven mass mortality, and a peculiar taphonomic phenomenon. In our model, an important role is played by the adverse oceanographic conditions common during the extreme greenhouse global climate regime of the Cretaceous, manifested by the well-known Ocean Anoxic Events around the time when these deposits were formed. The proposed mechanisms responsible for the mass accumulations we report here have operated at other times in Earth history, and may help explain similar occurrences elsewhere in the fossil record. 相似文献

The hypothesis that nitrate versus phosphate regulates the coastal primary production has been assessed at different time scales in the northwest Alboran Sea. Time series of temperature, salinity, nutrients and chlorophyll a obtained at three stations located off Málaga city (the greatest coastal urban core along the Alboran Sea shoreline) from 1992 to 2006 have been analysed. At the decadal scale, temperature increased linearly while salinity decreased. These changes were related to a shift in the wind regime suggesting that coastal upwelling became steadily weaker. In contrast to phosphate, nitrate was positively correlated with salinity at the seasonal scale and decreased linearly from 1992 to 2006. Seasonal and decadal changes in chlorophyll a were correlated with nitrate (and uncorrelated with phosphate). However, non-regular variability in chlorophyll a was correlated with phosphate. Consequently, the results demonstrate that nitrate controls the phytoplankton biomass at the inter-annual scale while both nitrate and phosphate do so at a shorter time scale. The Bay of Málaga receives elevated entries of domestic waste waters that release high loads of phosphate compared to nitrate. Our analysis indicates that the expected impact of this pollution on chlorophyll a at the inter-annual scale is reduced in comparison with the effects of atmospheric forcing. 相似文献

Aim Our aim was to investigate the historical biogeography of the three genera of the Leucocroton alliance (i.e. Garciadelia Jestrow & Jiménez Rodr., Lasiocroton Griseb., and Leucocroton Griseb., Euphorbiaceae). Location The alliance is restricted to the Bahamas, Cuba, Hispaniola and Jamaica. Methods Members of the Leucocroton alliance, along with representatives from tribe Adelieae (Adelia L. and Philyra Klotzsch.), were included in a molecular phylogenetic analysis based upon nucleotide sequences of the internal transcribed spacer region of the nuclear ribosomal DNA and the non‐coding chloroplast regions psbM–trnD and ycf6–pcbM. The program s‐diva was used to calculate ancestral areas based on the phylogenetic trees and present species distributions. Results Phylogenetic analyses support the monophyly of the three genera. The ancestral area of the Leucocroton alliance is eastern Cuba and Hispaniola. Ancestral forms of Leucocroton arose on eastern Cuba and underwent two migrations across the island. The ancestor of Lasiocroton also originated on eastern Cuba followed by later dispersal to and speciation events on the other islands. Our study also suggests that ancestral forms of the Leucocroton alliance probably occurred on limestone soils. Main conclusions Our study concurs with previous hypotheses suggesting that the flora of serpentinite regions of the Caribbean derives from other types of soils. The serpentine endemics of the Leucocroton alliance have a single origin and represent one of the most extraordinary examples of speciation in this unique environment of the New World. The high colonization success achieved by the members of Leucocroton on serpentine soils was not attained by the other genera of the alliance, which occur on limestone areas. 相似文献
