Three forms of the normal human plasma fibrinogen γ-chain which differ in molecular weight have been purified. Plasma fibrinogen was separated by ion exchange chromatography on DEAE-Sephacel into three populations of molecules, each with a unique γ-chain composition. Following reduction and S-carboxymethylation, the fibrinogen polypeptide chains in each chromatographic peak were separated by ion exchange chromatography on DEAE-Sephacel and identified following sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The Aα, Bβ and smallest γ-chain (γ50) eluted at progressively higher ionic strengths, but the elution positions of Aα, Bβ and γ50 chains were identifcal for fibrinogen from each of the three different chromatographic fractions. The unique γ chain of fibrinogen in the second chromatographic peak (γ55) eluted at an ionic strength higher than that of the γ50 chain, while the largest γ-chain (γ57.5), which was contained only in the third chromatographic peak of fibrinogen, eluted at the highest ionic strength. The higher ionic strengths needed to elute fibrinogen in the second and third peaks was paralleled by the higher ionic strengths needed to elute the γ-chains unique to them, suggesting that the γ-chain composition of the three fibrinogen fractions accounted for their differential binding to the ion exchange resin. Following desialation with neuraminidase, the differences in electrophoretic mobilities between the three γ-chain forms was maintained, indicating that differential migration on SDS-polyacrylamide gel electrophoresis was not due to variation in sialic acid content.  相似文献

Use of soft‐metal (aluminum alloy) bands on gulls (Laridae) is known to result in high rates of band loss and, as a result, hard‐metal (monel, incoloy, or stainless steel) bands are superior for most studies. However, the U.S. Bird Banding Laboratory (BBL) and the Canadian Wildlife Service Bird Banding Office continue to issue soft bands for use on gulls, and the BBL does not make specific recommendations about use of hard bands so many banders continue to use soft bands. For wholly marine species of gulls banded in North America since 1996, ~20% have been banded with soft bands; the proportion of soft bands used on partially freshwater gulls was ~70% up to 2009, but has since fallen to 40%. Using hierarchical Bayesian models in program MARK, we analyzed recovery data for three gull species and found that estimates of annual survival rates derived from soft bands (0.68–0.81) were lower than those derived from hard bands (0.85–0.96). Comparison of survival rates of Herring Gulls (Larus argentatus) in the Great Lakes basin and on the Atlantic coast provided no evidence that soft bands last longer in freshwater than saltwater. Band loss compromises many types of studies, including those assessing the possible effects of climate change. We recommend that use of soft bands on gulls be discontinued, and that banders be required to use hard bands on these species in the future. The same consideration applies to other long‐lived species, including some waterfowl and all albatrosses, pelicans, cormorants, shearwaters, petrels, terns, shorebirds, and alcids. Use of hard bands should be based on expectations about a species’ longevity and evidence of band wear, rather than on whether or not it occurs in saltwater.  相似文献

Aggregatibacter actinomycetemcomitans is a Gram-negative periodontitis-associated bacterium that expresses a toxin that selectively affects leukocytes. This leukotoxin is encoded by an operon belonging to the core genome of this bacterial species. Variations in the expression of the leukotoxin have been reported, and a well-characterized specific clonal type (JP2) of this bacterium with enhanced leukotoxin expression has been isolated. In particular, the presence of the JP2 genotype significantly increases the risk for the progression of periodontal attachment loss (AL). Based on these findings we hypothesized that variations in the leukotoxicity are linked to disease progression in infected individuals. In the present study, the leukotoxicity of 239 clinical isolates of A. actinomycetemcomitans was analysed with different bioassays, and the genetic peculiarities of the isolates were related to their leukotoxicity based on examination with molecular techniques. The periodontal status of the individuals sampled for the presence of A. actinomycetemcomitans was examined longitudinally, and the importance of the observed variations in leukotoxicity was evaluated in relation to disease progression. Our data show that high leukotoxicity correlates with an enhanced risk for the progression of AL. The JP2 genotype isolates were all highly leukotoxic, while the isolates with an intact leukotoxin promoter (non-JP2 genotypes) showed substantial variation in leukotoxicity. Genetic characterization of the non-JP2 genotype isolates indicated the presence of highly leukotoxic genotypes of serotype b with similarities to the JP2 genotype. Based on these results, we conclude that A. actinomycetemcomitans harbours other highly virulent genotypes besides the previously described JP2 genotype. In addition, the results from the present study further highlight the importance of the leukotoxin as a key virulence factor in aggressive forms of periodontitis.  相似文献

Monterey Bay, CA is an Eastern boundary upwelling system that is nitrogen limited much of the year. In order to resolve population dynamics of microorganisms important for nutrient cycling in this region, we deployed the Environmental Sample Processor with quantitative PCR assays targeting both ribosomal RNA genes and functional genes for subclades of cyanobacteria (Synechococcus) and ammonia-oxidizing Archaea (Thaumarchaeota) populations. Results showed a strong correlation between Thaumarchaea abundances and nitrate during the spring upwelling but not the fall sampling period. In relatively stratified fall waters, the Thaumarchaeota community reached higher numbers than in the spring, and an unexpected positive correlation with chlorophyll concentration was observed. Further, we detected drops in Synechococcus abundance that occurred on short (that is, daily) time scales. Upwelling intensity and blooms of eukaryotic phytoplankton strongly influenced Synechococcus distributions in the spring and fall, revealing what appear to be the environmental limitations of Synechococcus populations in this region. Each of these findings has implications for Monterey Bay biogeochemistry. High-resolution sampling provides a better-resolved framework within which to observe changes in the plankton community. We conclude that controls on these ecosystems change on smaller scales than are routinely assessed, and that more predictable trends will be uncovered if they are evaluated within seasonal (monthly), rather than on annual or interannual scales.  相似文献
