Mutations in the CETP gene resulting in defective CETP activity have been shown to cause remarkable elevations of plasma HDL-C levels, with the accumulation in plasma of large, buoyant HDL particles enriched in apolipoprotein E. Genetic CETP deficiency thus represents a unique tool to evaluate how structural alterations of HDL impact on HDL atheroprotective functions. Aim of the present study was to assess the ability of HDL obtained from CETP-deficient subjects to protect endothelial cells from the development of endothelial dysfunction. HDL isolated from one homozygous and seven heterozygous carriers of CETP null mutations were evaluated for their ability to down-regulate cytokine-induced cell adhesion molecule expression and to promote NO production in cultured endothelial cells. When compared at the same protein concentration, HDL and HDL3 from carriers proved to be as effective as control HDL and HDL3 in down-regulating cytokine-induced VCAM-1, while carrier HDL2 were more effective than control HDL2 in inhibiting VCAM-1 expression. On the other hand, HDL and HDL fractions from carriers of CETP deficiency were significantly less effective than control HDL and HDL fractions in stimulating NO production, due to a reduced eNOS activating capacity, likely because of a reduced S1P content. In conclusion, the present findings support the notion that genetic CETP deficiency, by affecting HDL particle structure, impacts on HDL vasculoprotective functions. Understanding of these effects might be important for predicting the outcomes of pharmacological CETP inhibition.  相似文献

LCAT (lecithin:cholesterol acyltransferase) catalyzes the transacylation of a fatty acid of lecithin to cholesterol, generating a cholesteryl ester and lysolecithin. The knowledge of LCAT atomic structure and the identification of the amino acids relevant in controlling its structure and function are expected to be very helpful to understand the enzyme catalytic mechanism, as involved in HDL cholesterol metabolism. However - after an early report in the late ‘90 s - no recent advance has been made about LCAT three-dimensional structure. In this paper, we propose an LCAT atomistic model, built following the most up-to-date molecular modeling approaches, and exploiting newly solved crystallographic structures. LCAT shows the typical folding of the α/β hydrolase superfamily, and its topology is characterized by a combination of α-helices covering a central 7-strand β-sheet. LCAT presents a Ser/Asp/His catalytic triad with a peculiar geometry, which is shared with such other enzyme classes as lipases, proteases and esterases. Our proposed model was validated through different approaches. We evaluated the impact on LCAT structure of some point mutations close to the enzyme active site (Lys218Asn, Thr274Ala, Thr274Ile) and explained, at a molecular level, their phenotypic effects. Furthermore, we devised some LCAT modulators either designed through a de novo strategy or identified through a virtual high-throughput screening pipeline. The tested compounds were proven to be potent inhibitors of the enzyme activity.  相似文献

Summary The distribution of lanthanum on the taste disc of the frog,Rana esculenta, afteren bloc staining of the tongue with lanthanum nitrate was studied at the ultrastructural level by means of scanning electron microscopy (in the secondary electron mode or in the back scattered electron mode), energy-dispersive X-ray microanalysis, transmission electron microscopy and electron spectroscopic imaging. It was consistently found that lanthanum distribution on the surface of the taste disc is not homogeneous and that the surface of putative receptor cells is in contact with strongly lanthanum-positive material. Calcium co-localizes with lanthanum at that level. These results suggest that different microenvironments exist at the surface of the taste disc and that this could be relevant to the receptor function.  相似文献

Serum and cerebrospinal fluid (CSF) prolactin (PRL) concentrations were determined in fourteen patients of both sexes suffering from hydrocephalus, in basal conditions and after i.m. administration of 10 mg metoclopramide or 10 mg morphine. A significant increase in both serum and CSF hormone values was found after administration of both drugs. Serum and CSF PRL values after metoclopramide administration increased earlier and to a greater extent than after morphine. Furthermore, the metoclopramide induced CSF PRL increase immediately followed the serum peak, whereas after morphine administration an evident delay in the CSF hormone peak with respect to the serum increase was found. These data suggest that PRL entry in the CSF compartment is subject to a controlling mechanism which acts at the blood/brain barrier.  相似文献

Puccinia psidii sensu lato (s.l.) is the causal agent of eucalyptus and guava rust, but it also attacks a wide range of plant species from the myrtle family, resulting in a significant genetic and physiological variability among populations accessed from different hosts. The uredospores are crucial to P. psidii dissemination in the field. Although they are important for the fungal pathogenesis, their molecular characterization has been poorly studied. In this work, we report the first in-depth proteomic analysis of P. psidii s.l. uredospores from two contrasting populations: guava fruits (PpGuava) and eucalyptus leaves (PpEucalyptus). NanoUPLC-MSE was used to generate peptide spectra that were matched to the UniProt Puccinia genera sequences (UniProt database) resulting in the first proteomic analysis of the phytopathogenic fungus P. psidii. Three hundred and fourty proteins were detected and quantified using Label free proteomics. A significant number of unique proteins were found for each sample, others were significantly more or less abundant, according to the fungal populations. In PpGuava population, many proteins correlated with fungal virulence, such as malate dehydrogenase, proteossomes subunits, enolases and others were increased. On the other hand, PpEucalyptus proteins involved in biogenesis, protein folding and translocation were increased, supporting the physiological variability of the fungal populations according to their protein reservoirs and specific host interaction strategies.  相似文献

We utilized the fluorescent serotonin analogue 5,7-dihydroxytryptamine (5,7DHT) to visualize basal cells in the frog's taste organ in supravital conditions. In whole mounts of lingual mucosa, specifical and detailed morphological visualization of fluorescent basal cells was obtained in the peripheral and central region of the intact taste organ; similar results were obtained after mechanical dissociation. Preincubation with serotonin prevented any fluorescence in basal cells. Electron microscopy showed good preservation of the ultrastructural morphology of the taste disk after exposure to 5,7DHT. The advantages of the current method as compared with conventional ones are discussed. This simple, reliable procedure will be useful to further define the biology of neuroendocrine cells in taste as well as in other organs.  相似文献

One-year-old cork oak (Quercus suber) and turkey oak (Q. cerris) seedlings were exposed to ozone (110 ppb, 5 h day(-1), for 30 days) and were inoculated with Diplodia corticola and Biscogniauxia mediterranea, respectively, by spraying a suspension of spores on the leaves. Both fungi are endophytic and may act as weak parasites, contributing to oak decline. Ozone exposure stimulated leaf attacks after inoculation, although the physiological, visible, and structural responses of both oaks to O3 exposure were weak. In fact, steady-state gas exchange, leaf waxes, and wettability were not significantly affected by O3. In Q. cerris, O3 altered the structure of stomata, as observed by scanning microscopy, and reduced the leaf relative water content. No hyphal entry through stomata or growth towards stomata was, however, observed. Inoculations were performed in a humid chamber at low light; stomata were likely to be closed. When Q. cerris was inoculated in natural conditions, i.e., in a forest infected by B. mediterranea, seedlings pre-exposed to the enhanced O3 regime had a higher number of B. mediterranea isolates than the controls. This suggests that pre-exposure to O3 predisposed Q. cerris leaves to attacks by B. mediterranea independent of stomata. The hyphae of both fungi were able to enter the leaf through the cuticle, either by gradual in-growth into the cuticle or erosion of a hollow in the cuticle at the point of contact. The primary cause of increased leaf injury in O3-exposed seedlings appeared to be higher germination of spores than on control leaves.  相似文献
