The new epidemiological scenario of orally transmitted Chagas disease that has emerged in Brazil, and mainly in the Amazon region, needs to be addressed with a new and systematic focus. Belém, the capital of Pará state, reports the highest number of acute Chagas disease (ACD) cases associated with the consumption of açaí juice.
Methodology/Principal Findings
The wild and domestic enzootic transmission cycles of Trypanosoma cruzi were evaluated in the two locations (Jurunas and Val-de Cães) that report the majority of the autochthonous cases of ACD in Belém city. Moreover, we evaluated the enzootic cycle on the three islands that provide most of the açaí fruit that is consumed in these localities. We employed parasitological and serological tests throughout to evaluate infectivity competence and exposure to T. cruzi. In Val-de-Cães, no wild mammal presented positive parasitological tests, and 56% seroprevalence was observed, with low serological titers. Three of 14 triatomines were found to be infected (TcI). This unexpected epidemiological picture does not explain the high number of autochthonous ACD cases. In Jurunas, the cases of ACD could not be autochthonous because of the absence of any enzootic cycle of T. cruzi. In contrast, in the 3 island areas from which the açaí fruit originates, 66.7% of wild mammals and two dogs displayed positive hemocultures, and 15.6% of triatomines were found to be infected by T. cruzi. Genotyping by mini-exon gene and PCR-RFLP (1f8/Akw21I) targeting revealed that the mammals and triatomines from the islands harbored TcI and Trypanosoma rangeli in single and mixed infections.
Conclusion/Significance
These findings show that cases of Chagas disease in the urban area of Belém may be derived from infected triatomines coming together with the açaí fruits from distant islands. We term this new epidemiological feature of Chagas disease as “Distantiae transmission”. 相似文献
Antigen-specific T cell blasts to poly-18, a polypeptide antigen of defined sequence and conformation, were generated from lymph nodes of antigen-primed BALB/cCr mice. These blasts were fused with the BW5147 thymoma to obtain anti-poly-18-reactive T cell hybridomas. All of the hybridomas were IAd-restricted and secreted IL2 in the presence of IAd/poly-18. On the basis of fine specificity analysis, these hybridomas were classified into two groups. Group A hybridomas recognized a minimal peptide sequence of Glu-Tyr-Lys-(Glu-Tyr-Ala)3-Glu-Tyr-Lys, whereas Group B needed the sequence Glu-Tyr-Ala-(Glu-Tyr-Ala)3-Glu-Tyr-Lys/Ala for activation. Three critical residues were identified in Group A hybridomas: the alanine residue at position 9, the carboxy terminal lysine, and the lysine at position 3. In Group B hybridomas, the alanine at position 3 was found to be the critical residue. We suggest that the amino acid residue at position 3 (lysine/alanine) is the T cell receptor contact residue on the poly-18 antigen in BALB/cCr mice. 相似文献
Knowledge about short-term climate change adaptation strategies for Mediterranean vineyards is needed in order to improve grapevine physiology and yield-quality attributes. We investigated effects of kaolin-particle film suspension on water relations, photosynthesis and oxidative stress of field-grown grapevines in the Douro region (northern Portugal) in 2012 and 2013. Kaolin suspension decreased leaf temperature by 18% and increased leaf water potential (up to 40.7% in 2013). Maximum photochemical quantum efficiency of PSII was higher and the minimal chlorophyll fluorescence was lower in the plants sprayed by kaolin. Two months after application, net photosynthesis and stomatal conductance at midday increased by 58.7 and 28.4%, respectively, in treated plants. In the same period, kaolin treatment increased photochemical reflectance, photosynthetic pigments, soluble proteins, soluble sugars, and starch concentrations, while decreased total phenols and thiobarbituric acid-reactive substances. Kaolin application can be an operational tool to alleviate summer stresses, which ameliorates grapevine physiology and consequently leads to a higher yield. 相似文献
A new euryhaline and eurythermal dinoflagellate species, Biecheleria tirezensis sp. nov., is described based on samples taken from an extreme environment, the athalassohaline and particularly sulphate-rich Tirez natural pond (Spain). This species is able to survive in salinities from almost fresh water up to 56 and over a 5–25°C temperature range. Thus, the ecological characteristics of this isolate differentiate it from other species of the same genus. Its morphology, as examined by light, scanning and transmission electron microscopy, shows that the cells are almost spherical, with several series of amphiesmal vesicles. It also has a single elongate amphiesmal vesicle (EAV) observed by SEM and the eyespot has a type E structure, typical of Biecheleria. Spherical and smooth cysts were observed in old cultures. The pigment composition is typical of a peridinin-containing dinoflagellate. Phylogeny inferred from nuclear rDNA SSU, ITS and LSU sequences showed the isolate belongs to the genus Biecheleria, closest to B. cincta and B. brevisulcata. Modelling and analysis of the secondary structure of its ITS2 region, and that of other species of the same genus and some representatives of the most closely related genera, indicated that the isolate represents a new species clearly separated from but related to B. cincta. The criterion of the presence of Compensatory Base Changes (CBCs) in the secondary structure of the ITS2 region as an indicator of species differentiation confirmed this, supporting the establishment of the Tirez pond isolate as a new species of Biecheleria. 相似文献
A persistent and unusual bloom of Dinophysic acuta Ehrenbergin the RIas Bajas (Northwest Spain) from early July to mid-November1989, was associated with a diarshetic shellfish poisoning (DSP)outbreak that prevented mussel extraction in some areas fromearly August to mid-December. Integrated samples (05,510 and 1015 m) were taken once or twice a weekat six stations. Maximum numbers of D.acuta (up to 2.3 104intheintegratedsamples)andpercentageofobserveddMdingcells(upto3l%),appearedin August, and formed a maximum in the thermodine. Cellnumbers were minimal when upwelling caused a breakdown of stratification.Fligh numbers of D.acuta (up to 1.8 104 cells 11 appearedagain when persistent south winds in late October caused a reversalof the circulation. Thus, the two cell maxima occurred duringtwo distinct hydrographic regimes, but in both cases at temperaturesof 1517C. Numerous cells with one typical D.acuta valve,but the other valve reduced in size, were seen during the bloom,as well as Dinophysts dens Pavillard cells that might be a lifecycle stage of D.acuta. Other observations include double- walled,unwinged forms that are perhaps some kind of cyst, and a hypotheticalfertilizing tube emerging from the larger cellof a D.acuta/D.dens couplet. It is suggested that the maximaduring stratification and during downwelling episodes couldcorrespond to periods with a predominance of in situ growthand physical accumulation, respectively, and that the exceptionalmeteorological conditions during 1989 provided the optimum scenariofor the unusual occurrence of D.acuta. 相似文献
Bioabsorbable hemostatic agents such as oxidized regenerated cellulose are widely used to control intraoperative diffuse capillary bleeding. Compared with electrocautery or ligation, oxidized regenerated cellulose has the advantage of controlling bleeding without occluding the vessel lumen or causing thermal injuries to adjacent tissue. Although the manufacturer recommends removal of the material once hemostasis is achieved, oxidized regenerated cellulose is a bioabsorbable hemostatic agent and is often left in the surgical bed to prevent subsequent bleeding after surgical closure. However, noninvasive imaging techniques have revealed granulomatous foreign-body reactions that mimic infection or tumor recurrence. We present a case report of sterile peritonitis and granuloma formation secondary to the presence of oxidized regenerated cellulose after intestinal resection to excise a colonic adenocarcinoma in an aged rhesus macaque.Bioabsorbable hemostatic agents such as oxidized regenerated cellulose (for example, Surgicel) are widely used to control intraoperative diffuse capillary bleeding. This fiber is then woven into a gauze and oxidized.17,22 Oxidized regenerated cellulose is supplied as a substrate that is flexible, malleable, and trimable.16The mechanism of hemostasis of oxidized regenerated cellulose is reportedly associated with its caustic activity.2 The oxidation of cellulose produces a low-pH organic acid that reacts with blood, thus forming an artificial clot and causing platelet aggregation.18Although the manufacturer recommends the removal of oxidized regenerated cellulose once hemostasis is achieved,8 the product, a bioabsorbable hemostatic agent, is often left in situ within the surgical bed to prevent bleeding after surgical procedures. The biodegradation and elimination of oxidized regenerated cellulose from the tissue occurs in 2 phases.14 Polyanhydroglucuronic acid, the major functional unit of oxidized regenerated cellulose, is readily soluble. This acid is degraded extracellulary and systematically cleared from the system approximately 18 h after implantation.13,14 The remaining fibrous residue, however, requires macrophage phagocytosis for clearance and can be observed within macrophages for at least 48 h after implantation.13 Unfortunately, these fibrous residues have a prolonged degradation, and their persistence for as long as 7 mo after surgery has been confirmed histologically.7Despite the biocompatibility of oxidized regenerated cellulose, granulomatous foreign-body reactions that imitate infection or tumor recurrence have been revealed by using noninvasive imaging techniques.1,11,12,15,17,18,22 Here we describe a case of peritonitis and granuloma formation secondary to the presence of oxidized regenerated cellulose after an intestinal resection to excise a colonic adenocarcinoma in an aged rhesus macaque. 相似文献
Here we evaluated and characterized the growth dynamics of A. angustifolia embryogenic cultures (EC) submitted to different cryotreatment incubation times through morphological and time-lapse cell tracking analyzes. The EC submitted to cryopreservation protocol were evaluated by regrowth rates, and ultrastructural characterization by transmission electron microscopy (TEM). The results indicated that A. angustifolia EC support all the cryoprotection times evaluated, without cell proliferation inhibition, but with noticeable genotype-dependent response in all tested cell lines. The use of 1M DMSO showed non-inhibitory effects to EC regrowth independent of cell line or cryotreatment incubation time. However, after cryopreservation, Cr01 cell line regrowth was 100 % for all cryotreatments incubation times evaluated (30, 60, 120, 240 min), while Cr02 cell line only showed 100 % regrowth in 240 min of cryotreatment. The 100 % cell regrowth obtained in both cell lines indicates that the proposed protocol can be successful applied to A. angustifolia EC cryopreservation. Cell tracking analysis showed a survival and initial proliferation of embryogenic cells, with the first cell regrowth signs after 30 days in culture. TEM analysis revealed a conspicuous cell wall thickening in embryogenic cells after cryotreatment and after thawing, which may be related to osmotic stress response caused by the cryopreservation process. An increased heterochromatin presence was also observed in cryotreated or after thawing cells, may possibly be acting as a cell defense mechanism, decreasing the DNA vulnerability to cleavage and preserving the cell integrity.
