The plasma membrane of cells has a complex architecture based on the bidimensional liquid-crystalline bilayer arrangement of phospho- and sphingolipids, which in turn embeds several proteins and is connected to the cytoskeleton. Several studies highlight the spatial membrane organization into more ordered (Lo or lipid raft) and more disordered (Ld) domains. We here report on a fluorescent analog of the green fluorescent protein chromophore that, when conjugated to a phospholipid, enables the quantification of the Lo and Ld domains in living cells on account of its large fluorescence lifetime variation in the two phases. The domain composition is straightforwardly obtained by the phasor approach to confocal fluorescence lifetime imaging, a graphical method that does not require global fitting of the fluorescence decay in every spatial position of the sample. Our imaging strategy was applied to recover the domain composition in human oligodendrocytes at rest and under treatment with galactosylsphingosine (psychosine). Exogenous psychosine administration recapitulates many of the molecular fingerprints of a severe neurological disease, globoid cell leukodystrophy, better known as Krabbe disease. We found out that psychosine progressively destabilizes plasma membrane, as witnessed by a shrinking of the Lo fraction. The unchanged levels of galactosyl ceramidase, i.e., the enzyme lacking in Krabbe disease, upon psychosine treatment suggest that psychosine alters the plasma membrane structure by direct physical effect, as also recently demonstrated in model membranes. 相似文献

The HNK-1 carbohydrate epitope is a 3-sulfo-glucuronyl residue attached to lactosamine structures on glycoproteins, proteoglycans, or glycolipids mostly expressed in the nervous system. Here, using monoclonal antibodies against the sulfated HNK-1 carbohydrate epitope, we first examined its distribution in developing and adult kidneys, then its expression in kidneys with tubular necrosis and renal neoplasms. This HNK-1 epitope was expressed in the human, rabbit, and rat, but not mouse kidney. It was detected within a subset of epithelial cells in the renal vesicle and in comma- and S-shaped bodies during early stages of nephrogenesis. In ureteral bud derivatives, the epitope was present transiently in the area where the collecting duct fused with the nephron. In the adult kidney, expression of the HNK-1 epitope became mainly restricted to the thin ascending loop of Henle where this epitope was carried by heparan- and chondro-proteoglycan. In pathological conditions, HNK-1 epitope expression increased dramatically in proximal epithelial tubule cells in kidneys with acute tubular necrosis. In tumors, the HNK-1 epitope was expressed in the epithelial component of nephroblastomas and in a subgroup of papillary renal cell carcinomas. These data suggest that molecules carrying the sulfated HNK-1 carbohydrate epitope may play an important role in critical stages of renal development and in the physiology of thin ascending loop of Henle. 相似文献

Pair formation in the bushcricket Gampsocleis gratiosa is achieved through acoustic signalling by the male and phonotactic approaches of the female towards the calling song. On
a walking belt in the free sound field, females tracked the position of the speaker broadcasting the male calling song with
a remarkable precision, deviating by no more than 10 cm in either direction from the ideal course. Starting with stimulus
angles of 6–10° the females significantly turned to the correct side, and with stimulus angles greater than 25° no incorrect
turns were made. Using neurophysiological data on the directionality of the ear we calculated that with such stimulus angles
the available binaural intensity difference is in the order of 1–2 dB. We developed a dichotic ear stimulation device for
freely moving females with a cross-talk barrier of about 50 dB, which allowed to precisely apply small binaural intensity
differences. In such a dichotic stimulation paradigm, females on average turned to the tronger stimulated side starting with
a 1 dB difference between both ears. The significance of such a reliable lateralization behaviour with small interaural intensity

The regional distribution pattern of Bombus muscorum was studied in an agricultural landscape of central Germany, one of two remaining areas with the occurrence of this nationally
endangered species in the Land Hesse. To determine the landscape characteristics that facilitate the occurrence of B. muscorum, grid-based observation records were analysed in a GIS environment at a regional scale. A significantly negative effect of
the number of trees on the occurrence of B. muscorum and a significantly positive one of the proportion of arable land, strongly support the species’ preference for open landscapes.
Yet, apart from open landscapes additional landscape features were shown to be important. A significantly positive effect
of ditches in the final model revealed the importance of this landscape element for the occurrence of B. muscorum. This finding was additionally supported by recordings of nest-searching queens, nests, and flower visits along dithes. The
positive effects of clover and fallow land indicate the species’ need for suitable food resources throughout the season. Because
B. muscorum exhibits small foraging ranges, it is essential that landscape elements that provide nesting sites, foraging habitats and
undisturbed hibernation structures are next to each other. The low numbers of individuals of B. muscorum recorded indicate that the supply of these habitat elements may have reached a critical threshold in the study region. 相似文献
