The lizard fauna of sand‐mined dunes of the central coast of New South Wales, Australia has been shown to be dominated by Ctenotus robustus and Ctenotus taeniolatus (Scincidae), with relative abundance changing with time since mining. However, there is little published information on how this lizard fauna compares to that of the undisturbed open forest that previously grew on these sites. Here, existing data are added to in order to produce a longer chronosequence of times since sand‐mining (4, 8, 14 and 20 years) than has been examined previously. The new data are compared to those from unmined forests. Ctenotus robustus and C. taeniolatus dominated lizard captures on mined areas, with peak abundances at 8 and 14 years, respectively. Lampropholis guichenoti (Scincidae) was at low abundance until 20 years post‐mining and L. delicata was present only at 20 years post‐mining. Unmined forest burned 4, 8 or 14 years ago had a significantly different lizard community from that of sand‐mined areas. Ctenotus robustus and C. taeniolatus were absent from unmined forest at all post‐fire periods. Lampropholis guichenoti and Lampropholis delicata were numerically dominant in forest, with increasing abundance of L. guichenoti with time since fire. Thus the composition of the lizard community on these coastal dunes is not solely determined by time since disturbance per se. Comparisons of sites on the basis of accumulated leaf litter showed a significant relationship between Lampropholis abundance and litter density. On sand‐mined sites and forested sites with similar leaf litter densities, the abundances of L. guichenoti were similar. As Ctenotus were absent from unmined forest, we could not compare their distribution in unmined and mined areas. However, negative correlations of Ctenotus abundance with canopy cover and understorey vegetation density offer a possible explanation for the absence of these species from forest. 相似文献

Insulin-stimulated diacylglycerol production results from the hydrolysis of a novel phosphatidylinositol glycan 总被引:15,自引:0,他引:15
We recently described the insulin-dependent release of a carbohydrate substance from plasma membranes which regulated certain intracellular enzymes (Saltiel, A. R., and Cuatrecasas, P. (1986) Proc. Natl. Acad. Sci. U. S. A. 83, 5793-5797). This enzyme-modulating substance appeared to arise from the phosphodiesterase hydrolysis of a novel inositol-containing glycolipid. This is supported by observations that insulin stimulated the rapid generation of [3H]myristate-labeled diacylglycerol in cultured BC3Hl myocytes. Myristoyl diacylglycerol production in these cells was unaffected by epinephrine, although arachidonate-labeled diacylglycerol was rapidly produced in response to stimulation by this alpha-1 adrenergic agent. The production of distinct species of diacylglycerol was apparently due to hormonally specific hydrolysis of different precursors. A novel glycolipid was identified on silica TLC or high pressure liquid chromatography which served as a substrate for the insulin-stimulated phosphodiesterase reaction. This glycolipid was metabolically labeled with radioactive inositol, glucosamine, and myristic acid, suggesting a phosphatidylinositol (PI)-glycan structure. Treatment of this glycolipid with a PI-specific phospholipase C resulted in the generation of two products: an inositol phosphate-glycan which modulated the activity of the low Km cAMP phosphodiesterase and myristoyl diacylglycerol. Insulin caused the rapid hydrolysis of the PI-glycan, which was then apparently resynthesized. These data further suggest that insulin stimulates the activity of a phospholipase C which selectively hydrolyzes a novel PI-glycan, releasing a carbohydrate enzyme modulator as well as a unique species of diacylglycerol. 相似文献

Iron deficiency anemia (IDA) affects > 500 million people worldwide, and is linked to impaired cognitive development and function in children. Helicobacter pylori, a class 1 carcinogen, infects about half of the world’s population, thus creating a high likelihood of overlapping risk. This study determined the effect of H. pylori infection on iron homeostasis in INS-GAS mice. Two replicates of INS-GAS/FVB male mice (n = 9-12/group) were dosed with H. pylori (Hp) strain SS1 or sham dosed at 6–9 weeks of age, and were necropsied at 27–29 weeks of age. Hematologic and serum iron parameters were evaluated, as was gene expression in gastric and brain tissues. Serum ferritin was lower in Hp SS1-infected mice than uninfected mice (p < 0.0001). Infected mice had a lower red blood cell count (p<0.0001), hematocrit (p < 0.001), and hemoglobin concentration (p <0.0001) than uninfected mice. Relative expression of gastric hepcidin antimicrobial peptide (Hamp) was downregulated in mice infected with Hp SS1 compared to sham-dosed controls (p<0.001). Expression of bone morphogenic protein 4 (Bmp4), a growth factor upstream of hepcidin, was downregulated in gastric tissue of Hp SS1-infected mice (p<0.001). Hp SS1-infected mice had downregulated brain expression of tyrosine hydroxylase (Th) (p = 0.02). Expression of iron-responsive genes involved in myelination (myelin basic protein (Mbp) and proteolipid protein 2 (Plp2)) was downregulated in infected mice (p = 0.001 and p = 0.02). Expression of synaptic plasticity markers (brain derived neurotrophic factor 3 (Bdnf3), Psd95 (a membrane associated guanylate kinase), and insulin-like growth factor 1 (Igf1)) was also downregulated in Hp SS1-infected mice (p = 0.09, p = 0.04, p = 0.02 respectively). Infection of male INS-GAS mice with Hp SS1, without concurrent dietary iron deficiency, depleted serum ferritin, deregulated gastric and hepatic expression of iron regulatory genes, and altered iron-dependent neural processes. The use of Hp SS1-infected INS-GAS mice will be an appropriate animal model for further study of the effects of concurrent H. pylori infection and anemia on iron homeostasis and adult iron-dependent brain gene expression. 相似文献

Background
Manufacturing large quantities of recombinant RNAs by overexpression in a bacterial host is hampered by their instability in intracellular environment. To overcome this problem, an RNA of interest can be fused into a stable bacterial RNA for the resulting chimeric construct to accumulate in the cytoplasm to a sufficiently high level. Being supplemented with cost-effective procedures for isolation of the chimera from cells and recovery of the recombinant RNA from stabilizing scaffold, this strategy might become a viable alternative to the existing methods of chemical or enzymatic RNA synthesis. 相似文献939.
Of the non-native fishes introduced to the U.K., the pumpkinseed is one of six species predicted to benefit from the forecasted climate warming conditions. To demonstrate the potential response of adults and their progeny to a water temperature increase, investigations of parental pumpkinseed acclimatization, reproduction and YOY over-wintering were carried out in outdoor experimental ponds under ambient and elevated water temperature regimes. No temperature effects were observed on either adult survivorship and growth, and none of the assessed reproductive activity variables (total spawning time, spawning season length, number of spawning bouts) appeared to be responsible for the large differences observed in progeny number and biomass. However, it was demonstrated in a previous study [Zięba G. et al., 2010] that adults in the heated ponds began spawning earlier than those of the ambient ponds. Ambient ponds produced 2.8× more progeny than the heated ponds, but these progeny were significantly smaller, probably due to their late hatching date, and subsequently suffered very high mortality over the first winter. Pumpkinseed in the U.K. will clearly benefit from climate warming through earlier seasonal reproduction, resulting in larger progeny going into winter, and as a result, higher over-winter survivorship would be expected relative to that which occurs under the present climatic regime. 相似文献
