Mud volcanoes (MVs) emit globally significant quantities of methane into the atmosphere, however, methane cycling in such environments is not yet fully understood, as the roles of microbes and their associated biogeochemical processes have been largely overlooked. Here, we used data from high‐throughput sequencing of microbial 16S rRNA gene amplicons from six MVs in the Junggar Basin in northwest China to quantify patterns of diversity and characterize the community structure of archaea and bacteria. We found anaerobic methanotrophs and diverse sulfate‐ and iron‐reducing microbes in all of the samples, and the diversity of both archaeal and bacterial communities was strongly linked to the concentrations of sulfate, iron and nitrate, which could act as electron acceptors in anaerobic oxidation of methane (AOM). The impacts of sulfate/iron/nitrate on AOM in the MVs were verified by microcosm experiments. Further, two representative MVs were selected to explore the microbial interactions based on phylogenetic molecular ecological networks. The sites showed distinct network structures, key species and microbial interactions, with more complex and numerous linkages between methane‐cycling microbes and their partners being observed in the iron/sulfate‐rich MV. These findings suggest that electron acceptors are important factors driving the structure of microbial communities in these methane‐rich environments.  相似文献

Aim Nestedness occurs when species present in depauperate sites are subsets of those found in species‐rich sites. The degree of congruence of site nestedness among different assemblages can inform commonalities of mechanisms structuring the assemblages. Well‐nested assemblages may still contain idiosyncratic species and sites that notably depart from the typical assemblage pattern. Idiosyncrasy can arise from multiple processes, including interspecific interactions and habitat preferences, which entail different consequences for species co‐occurrences. We investigate the influence of fine‐scale habitat variation on nestedness and idiosyncrasy patterns of beetle and bird assemblages. We examine community‐level and pairwise species co‐occurrence patterns, and highlight the potential influence of interspecific interactions for assemblage structure. Location Côte‐Nord region of Québec, Canada. Methods We sampled occurrences of ground‐dwelling beetles, flying beetles and birds at sites within old‐growth boreal forest. We examined the nestedness and idiosyncrasy of sites and sought relationships to habitat attributes. We analysed non‐random species co‐occurrence patterns at pairwise and community levels, using null model analysis and five ‘association’ indices. Results All three assemblages were significantly nested. There was limited congruence only between birds and flying beetles whose nestedness was related to canopy openness. For ground‐dwelling beetles, nestedness was related to high stand heterogeneity and sapling density, whereas site idiosyncrasy was inversely related to structural heterogeneity. For birds, site idiosyncrasy increased with canopy cover, and most idiosyncratic species were closed‐canopy specialists. In all assemblages, species idiosyncrasy was positively correlated with the frequency of negative pairwise associations. Species co‐occurrence patterns were non‐random, and for flying beetles and birds positive species pairwise associations dominated. Community‐level co‐occurrence summaries may not, however, always reflect these patterns. Main conclusions Nestedness patterns of different assemblages may not correlate, even when sampled at common locations, because of different responses to local habitat attributes. We found idiosyncrasy patterns indicating opposing habitat preferences, consistent with antagonistic interactions among species within assemblages. Analysis of such patterns can thus suggest the mechanisms generating assemblage structures, with implications for biodiversity conservation.  相似文献

Regeneration and reestablishment of synaptic connections is an important topic in neurobiological research. In the present study, the regeneration of auditory afferents and the accompanying effects in the central nervous system are investigated in nymphs and adults of the bush cricket Tettigonia viridissima L. (Orthoptera: Tettigoniidae). In all animals in which the tympanal nerve is crushed, neuronal tracing shows a regrowth of the afferents into the prothoracic ganglion. This regeneration is seen in both adult and nymphal stages and starts 10–15 days after nerve crushing. Physiological recordings from the leg nerve indicate a recovery of tympanal fibres and a formation of functional connections to interneurones in the same time range. Electrophysiological recordings from the neck connective suggest additional contralateral sprouting of interneurones and the formation of aberrant connections. The regeneration processes of the tympanal nerve in nymphal stages and adults appear to be similar.  相似文献

Summary The influence of five substrates on the interaction betweenGlomus intraradices andFusarium oxysporum f.sp.radicis-lycopersici and its effect on tomato plants development was investigated. The presence ofG. intraradices decreased root necrosis in all substrates and affected the Fusarium population with different intensity depending on the substrate used. Substrates were found to influence disease development, Fusarium population in the substrate, root colonization by the endomycorrhizal fungus and growth of the host plant. In addition to providing good experimental conditions, the use of calcined montmorillonite clay also facilitated washing, recuperation, necrosis evaluation and staining of roots. Rhizobium bacteroids in nodule cells are surrounded by the peribacteroid membrane (pbm), which is derived from the host plasma membrane during infection. The pbm was purified from R. japonicum 61A76-induced soybean nodules and analyzed by comparing it with the host cell plasma membrane for the presence of nodulins, nodule-specific plant proteins. Nodulins were found in pbm by reacting Western blots with a nodule-specific antiserum raised against the pbm. Peribacteroid fluid (the fluid enclosed in the pbm) was also found to contain several nodulins. The pbm nodulins were confirmed to be of plant origin by in vitro translation of poly(A)+ nodule mRNA followed by immunoprecipitation by the nodule-specific antiserum. Antibodies raised against a synthetic peptide corresponding to a repeated domain in nodulin-24, a pbm nodulin, and the nodule-specific pbm antiserum reacted exclusively with the pbm. The absence of pbm-nodulins in the plasma membrane suggests that the infected cells direct the intracellular transport of the pbm nodulins exclusively to this de novo synthesized subcellular compartment essential for symbiotic nitrogen fixation.  相似文献

Acetosyringone, a phenolic inducer of the virulence (vir) genes of Agrobacterium tumefaciens, inhibited the growth of the nopaline-type strains T37 and C58 incubated under acidic conditions. In the course of a 6-day incubation with acetosyringone, avirulent clones were produced in different proportions by strains T37 and C58 and also by a spontaneous variant of strain C58, denominated C58F. The proportion of avirulent clones in acetosyringone-treated cultures often exceeded 50% for strains T37 and C58F and was of the order of 1% for strain C58. Control cultures not exposed to acetosyringone did not yield avirulent clones. Two other vir inducers, sinapinic acid and syringaldehyde, also inhibited growth and promoted accumulation of avirulent clones in cultures of strains C58F and T37. On the other hand, various acetosyringone analogs reported not to induce the vir genes did not act as growth inhibitors. All of the T37 and most of the C58F avirulent clones examined still carried a Ti plasmid. In all instances examined, avirulent clones still carrying a Ti plasmid were mutated in this plasmid. Mutants of strain C58F lacked the capacity to induce a virB::lacZ fusion in the presence of acetosyringone.  相似文献

Vesicular arbuscular mycorrhizal fungi can reduce plant disease symptoms and populations of pathogens through mechanisms that are not well understood. Glomus intraradices was grown on Daucus carota transformed roots in a two-compartment in vitro system. One compartment contained mycorrhizal roots on a complete growth medium, while the other contained a medium lacking sugar on which only mycelial growth was allowed. The direct interaction between G. intraradices and Fusarium oxysporum f. sp. chrysanthemi was studied in the compartment lacking sugar during a 5-day period. G. intraradices hyphal density and spore number were estimated along with F. o. chrysanthemi conidial germination, mycelial growth and sporulation. Five hours after inoculation, germination of F. o. chrysanthemi conidia doubled in the presence of G. intraradices. Radial growth of F. o. chrysanthemi colonies was always slightly but significantly enhanced in the presence of G. intraradices. No correlation was obtained between G. intraradices hyphae or spore densities and F. o. chrysanthemi hyphal growth. Overall sporulation of the 5-day-old F. o. chrysanthemi colonies was not influenced by the presence of G. intraradices. However, significant negative correlations were found between F. o. chrysanthemi conidia production and G. intraradices hyphae or spore concentrations. G. intraradices increased F. o. chrysanthemi conidial germination and slightly stimulated its hyphal growth in dual culture without any root influences. No antibiosis was observed between the fungi. The significance of the results and their potential implication for rhizosphere biology are discussed.  相似文献

Summary Mill tailings resulting from mining and metallurgical processes are usually disposed of into open-air impoundments, where they become subjected to chemical or microbial leaching. At the surface of the tailings, where oxic conditions prevail, acidophilic bacteria, such as thiobacilli, can oxidize sulfidic minerals (e.g. pyrite and pyrrhotite) and generate acidic metal-rich leachates as by-products of their metabolism. This, combined with chemical oxidation, leads to acid mine drainage (AMD). Biomineralization, whereby a proportion of the metal leachate is precipitated, can also occur in the oxidized tailings, often as a result of a close metal-bacteria interaction. Iron-rich precipitates are usually found on bacterial cell walls, and are thought to serve as nucleation sites for further mineralization within the tailings impoundments. As depth increases in mine tailings, oxygen depletion and the presence of water-saturated pores usually lead to anoxic conditions. Under such redox and chemical conditions, populations of sulfate-reducing bacteria (SRBs) can colonize the tailings. As a result of their metabolic activity, sulfate is reduced to hydrogen sulfide, which in turn can react with dissolved metals to form metal sulfide precipitates. Microbial sulfate reduction also generates alkalinity, although chemical dissolution of carbonate and oxide minerals probably also play an important role in the generation of alkaline conditions in mine tailings.  相似文献
