The grey mould fungus Botrytis cinerea produces two major phytotoxins, the sesquiterpene botrydial, for which the biosynthesis gene cluster has been characterized previously, and the polyketide botcinic acid. We have identified two polyketide synthase (PKS) encoding genes, BcPKS6 and BcPKS9, that are up-regulated during tomato leaf infection. Gene inactivation and analysis of the secondary metabolite spectra of several independent mutants demonstrated that both BcPKS6 and BcPKS9 are key enzymes for botcinic acid biosynthesis. We showed that BcPKS6 and BcPKS9 genes, renamed BcBOA6 and BcBO9 (for B. cinerea botcinic acid biosynthesis), are located at different genomic loci, each being adjacent to other putative botcinic acid biosynthetic genes, named BcBOA1 to BcBOA17. Putative orthologues of BcBOA genes are present in the closely related fungus Sclerotinia sclerotiorum, but the cluster organization is not conserved between the two species. As for the botrydial biosynthesis genes, the expression of BcBOA genes is co-regulated by the Gα subunit BCG1 during both in vitro and in planta growth. The loss of botcinic acid production does not affect virulence on bean and tomato leaves. However, double mutants that do not produce botcinic acid or botrydial (bcpks6Δbcbot2Δ) exhibit markedly reduced virulence. Hence, a redundant role of botrydial and botcinic acid in the virulence of B. cinerea has been demonstrated. 相似文献

This study was carried out in central Tanzania on a group of 45 Zebu and 37 crossbred cows which were 4 to 10 years old. At calving time, the animals were allocated to one of the 4 treatment groups. In addition to free access to grazing for all cows in the study, in group H: AR (n = 18), cows were fed a high level of concentrate supplementation (4kg/day) and calves were artificially reared; in group H:RS (n = 24), cows were fed a high level of concentrate supplementation (4kg/day) and calves were only allowed restricted suckling up until the weaning age of 6 months. In group L:AR (n = 23) cows were fed a low level of concentrate supplementation (2kg/day) and calves were artificially reared; and in group L:RS (n = 17) cows were fed a low level of concentrate supplementation (2kg/day) and calves were only allowed restricted suckling up until the weaning age of 6 months. Milk progesterone was used as a means of determining the postpartum resumption interval (PRI) and the interval from parturition to conception (PCI). The overall PRI was 47.4 ± 0.4 days and was significantly affected by breed but not by calving season, with crossbred cows exhibiting a shorter PRI than Zebu cows. The effect of the treatments was significant, with cows in the group H:AR displaying a significantly shorter PRI than those in the other groups, while cows in group L:RS showed a significantly longer PRI than those in the other groups. The overall PCI was 149.5 ±3.7 days, and was not significantly affected by breed or calving season. The effect of the treatments was significant, with cows in the group H:AR having a significantly shorter PCI than cows in the other groups, while cows in group L:RS showed a significantly longer PCI than those in the other groups. Crossbred cows had higher live weights at calving (299.4 kg) than Zebu cows (272.6 kg), while all cows gained weight during the first 3 months after calving. The treatments had a significant effect on weight gain, with cows in the group H:AR gaining significantly more weight than those in the other groups. Cows which had high live weights at calving exhibited significantly shorter PRI and PCI than the lighter cows. Animals which had gained more than 5 kg during the first month after calving, or which had gained more than 8 kg during the first 3 months after calving, showed significantly shorter PRI and PCI than cows which had gained less weight. The results show that the calf rearing system and the level of feed supplementation interact with each other and can influence the postpartum anoestrous period in Zebu and Zebu crossbred cattle. Increasing the level of nutrition in restricted suckling cows tended to improve the postpartum anoestrous period, but the positive effects of supplementation could not completely compensate for the negative effects of suckling. 相似文献

To establish an infection, Yersinia pseudotuberculosis utilizes a plasmid-encoded type III translocon to microinject several anti-host Yop effectors into the cytosol of target eukaryotic cells. YopD has been implicated in several key steps during Yop effector translocation, including maintenance of yop regulatory control and pore formation in the target cell membrane through which effectors traverse. These functions are mediated, in part, by an interaction with the cognate chaperone, LcrH. To gain insight into the complex molecular mechanisms of YopD function, we performed a systematic mutagenesis study to search for discrete functional domains. We highlighted amino acids beyond the first three N-terminal residues that are dispensable for YopD secretion and confirmed that an interaction between YopD and LcrH is essential for maintenance of yop regulatory control. In addition, discrete domains within YopD that are essential for both pore formation and translocation of Yop effectors were identified. Significantly, other domains were found to be important for effector microinjection but not for pore formation. Therefore, YopD is clearly essential for several discrete steps during efficient Yop effector translocation. Recognition of this modular YopD domain structure provides important insights into the function of YopD. 相似文献

When examined using SEM, Chinese samples of Tuber indicum and T. sinense displayed the same ascospore ornamentation as that of T. pseudohimalayense, T. indicum, collected in India by Duthie in 1899, and samples renamed T. himalayense in 1988. The different authors who named the four taxa (T. indicum, T. himalayense, T. sinense, T. pseudohimalyense) described differences in the surface of the peridium which could be considered as usual variations within a single species. We consider T. indicum, T. himalayense, T. sinense and T. pseudohimalayense as one species, T. indicum. Within this T. indicum complex, according to ITS and β-tubulin sequences, there are two groups in China, which could be considered as geographical ecotypes. A DNA fragment coding for a cellodextrinase of Bacteroides succinogenes S85 was isolated by screening of a pBR322 gene library in Escherichia coli HB101. Of 100,000 colonies screened on a complex medium with methylumbelliferyl-beta-D-cellobioside as the indicator substrate, two cellodextrinase-positive clones (CB1 and CB2) were isolated. The DNA inserts from the two recombinant plasmids were 7.7 kilobase pairs in size and had similar restriction maps. After subcloning from pCB2, a 2.5-kilobase-pair insert which coded for cellodextrinase activity was isolated. The enzyme was located in the cytoplasm of the E. coli host. It exhibited no activity on carboxymethyl cellulose, Avicel microcrystalline cellulose, acid-swollen cellulose, or cellobiose but hydrolyzed p-nitrophenyl-beta-D-cellobioside and p-nitrophenyl-beta-D-lactoside. The Km (0.1 mM) for the hydrolysis of p-nitrophenyl-cellobioside by the enzyme expressed in E. coli was similar to that reported for the purified enzyme from B. succinogenes. Expression of the cellodextrinase gene was subjected to catabolite repression by glucose and was not induced by cellobiose. The origin of the DNA insert from B. succinogenes was confirmed by Southern blot analysis. Western blotting (immunoblotting) using antibodies raised against the purified B. succinogenes cellodextrinase revealed a protein with a molecular weight of approximately 50,000 in E. coli clones which comigrated with the native enzyme isolated from B. succinogenes. These data indicate that the cellodextrinase gene expressed in E. coli is fully functional and codes for an enzyme with properties similar to those of the native enzyme. 相似文献

目的:研究慢性肾脏病(CKD)患者心脏结构及功能的变化.方法:选择我院肾内科175例慢性肾脏病未透析患者,按照2003年美国国家肾脏基金会-肾脏病转归质量(NKF-K/DOQI)指南的标准进行分期,观察所有患者心脏结构及功能在超声中的变化.结果:慢性肾脏病患者随着肾功能的恶化,各组之间比较,室间隔厚度(IVST)、左心室后壁厚度(LVPW)、左心室心肌重量指数(LVMI)、左心室舒张末期内径(LVDd)、左心房内径(LAD)具有升高的趋势(P<0.05,P<0.01);但E/A比值未出现伴随着肾功能恶化而逐渐减低的趋势(P>0.05);射血分数(EF)、短轴缩短卒(FS)在各期之间无明显变化(P>0.05);而TVI技术测定的Em、Em/Am具有显著减低的趋势(P<0.05,P<0.01);瓣膜返流以二尖瓣返流为主.结论:慢性肾脏病患者心脏结构与功能随肾功能减退而加重,超声心动图检查结合组织速度显像(TVI)技术能更好地检测心脏结构和功能变化,尤其是检测左心室舒张功能障碍. 相似文献
