Serving capacity studies were conducted on Santa Gertrudis, 5/8 Brahman, Brahman and Belmont Red bulls at 10 sites. Modifications, such as providing prior sexual experience, using females in oestrus and comparing restrained with unrestrained females, were made to the standard Bos taurus serving capacity test in a series of experiments with Santa Gertrudis bulls. Providing sexual experience to virgin bulls prior to testing improved the number of serves and libido scores in a serving capacity test. In serving capacity tests that used restrained females, expressions of sexual behaviour of bulls were not improved by having females in oestrus compared with non-oestrus females. There were more sexual behaviours, particularly mounts, in serving capacity tests involving restrained rather than unrestrained females. Summary statistics were collated from over 1100 serving capacity tests, in the 2 months prior to mating, across the 10 sites. In serving capacity tests using restrained females, in bulls of the same age, there were more expressions of sexual behaviours (except interest) in Belmont Red than in Brahman or 5/8 Brahman bulls with Santa Gertrudis bulls intermediate. The percentage of bulls displaying serves tended to increase with age; 82, 83 and 86% of 2-, 3- and > or =4-year-old Belmont Red bulls, respectively, completed 1 or more serves. This compared with 50, 50 and 66% of 2-, 3- and > or =4-year-old Santa Gertrudis bulls and 31% of 2-year-old Brahman bulls. Between 2 and 9% of bulls did not display any sexual behaviour in serving capacity tests with restrained females. Across genotypes, there were few and inconsistent relationships between either mounts or serves and liveweight, condition score, scrotal circumference, testicular tone and sheath measurements. The exceptions were in 2-year-old Brahmans where mounts were negatively correlated (r=-0.61; P<0.05) with umbilicus thickness and in 3-year-old Santa Gertrudis bulls, where serves (r=-0.53; P<0.01) and mounts+serves (r=-0.49; P<0.01) were negatively correlated with navel score. Measures of sexual behaviour were unable to consistently predict calf output of bulls in multiple-sire mating groups in extensive pastoral conditions. The main value of the serving capacity test in these genotypes is to identify whether a bull is capable of natural service.  相似文献

The alcohol dehydrogenase (Adh) region from five planitibia subgroup species of Hawaiian picture-wing Drosophila has been cloned. A total of 15 kb of DNA in and around the Adh gene has been compared among the five species. Genetic distances were calculated to determine evolutionary relationships. These distances agree with previous distances determined by protein polymorphism and DNA hybridization techniques and can be interpreted in terms of specific island colonization and speciation (founder) events over the past 5 Myr. Examination of the restriction maps of the cloned Adh region from the five species shows many instances of small deletions, insertion of a transposable element in D. heteroneura, and the existence of a highly variable region on the 3' side of the Adh gene. Clustering relationships and rates of DNA change are calculated and compared with the relationship found for other species of Drosophila.   相似文献

The goal in this study was to determine how increased impeller power affects enzyme expression in large-scale (80 m(3)), fed-batch Aspergillus oryzae fermentations. An approximate 50% increase in average impeller power was achieved by increasing impeller diameter approximately 10%, while operating at slightly reduced speed. Measured decreases in terminal (95%) mixing time show increased power improved bulk mixing. However, batches operated at increased power had lower recombinant enzyme productivity. Biomass assays and image analysis tests showed no significant difference between "high power" and control batches, suggesting that slower growth, altered morphology, or increased hyphal fragmentation were not the cause of reduced productivity. Off-line tests on the shear-thinning, highly viscous broth show oxygen limitation occurred after transport through the air-liquid interface and imply the limitation may involve bulk mixing. Specifically, oxygen transfer may be limited to a small zone surrounding each impeller. When this is the case, oxygen mass transfer will be determined by both impeller shear and fluid circulation, which have been characterized with the energy dissipation/circulation function (EDCF). EDCF values during control fermentations were approximately constant at 25 kW m (-3) s(-1), while EDCF values during "high power" batches fell linearly from 40 to 15 kW m (-3) s(-1). The point at which "high power" EDCF values drop below those in control fermentations corresponds almost exactly with the point at which product titer stops increasing. Thus, our findings suggest oxygen mass transfer was less efficient during the latter half of "high power" fermentations because of reductions in impeller speed and subsequent decreases in EDCF values. This observation has clear implications during the scale-up of viscous fungal fermentations, implying that not only is the level of impeller power important, but also relevant is how this power is applied.  相似文献

The phi-screen, a method of phylogenetic screening, can be employed to detect repetitive sequence families that differentially hybridize between closely related species. Such differences may involve sequence divergence or variations in copy number, including total presence versus absence of a family of repeated DNA. We present the results of a phi-screen comparing the human genome to that of the prosimian, Galago crassicaudatus. Three human repetitive families that are divergent or not present in galago have been detected. One of these families is described in detail; it is similar among the anthropoids but is present in a lower copy number and/or divergent form in prosimians. The family is clearly related to the transposon-like human element (THE) described by Paulson et al. (1985). THEs have long terminal repeats reminiscent of retroviruses but are unique in that they have no sequence similarity to known mammalian retroviruses. The sequence of a solo long terminal repeat, found unassociated with THE internal sequence, is presented. This family member, THE p2, is bordered by a 5-bp target-site repeat and is interrupted by the insertion of an Alu element. A solo THE element sequenced by Wiginton et al. (1986) contains an insertion of Alu at precisely the same position as does THE p2.   相似文献

There is a long tradition in ecology of evaluating the relative contribution of the regional species pool and local interactions on the structure of local communities. Similarly, a growing number of studies assess the phylogenetic structure of communities, relative to that in the regional species pool, to examine the interplay between broad-scale evolutionary and fine-scale ecological processes. Finally, a renewed interest in the influence of species source pools on communities has shown that the definition of the source pool influences interpretations of patterns of community structure. We use a continent-wide dataset of local ant communities and implement ecologically explicit source pool definitions to examine the relative importance of regional species pools and local interactions for shaping community structure. Then we assess which factors underlie systematic variation in the structure of communities along climatic gradients. We find that the average phylogenetic relatedness of species in ant communities decreases from tropical to temperate regions, but the strength of this relationship depends on the level of ecological realism in the definition of source pools. We conclude that the evolution of climatic niches influences the phylogenetic structure of regional source pools and that the influence of regional source pools on local community structure is strong.  相似文献
