Sunflower seed oil residue, a by-product of sunflower seed oil refining, was utilized as a feedstock for preparation of activated carbon (SSHAC) via microwave induced K(2)CO(3) chemical activation. SSHAC was characterized by Fourier transform infrared spectroscopy, nitrogen adsorption-desorption and elemental analysis. Surface acidity/basicity was examined with acid-base titration, while the adsorptive properties of SSHAC were quantified using methylene blue (MB) and acid blue 15 (AB). The monolayer adsorption capacities of MB and AB were 473.44 and 430.37 mg/g, while the Brunauer-Emmett-Teller surface area, Langmuir surface area and total pore volume were 1411.55 m(2)/g, 2137.72 m(2)/g and 0.836 cm(3)/g, respectively. The findings revealed the potential to prepare high surface area activated carbon from sunflower seed oil residue by microwave irradiation. 相似文献

PHF2 belongs to a class of α-ketoglutarate-Fe2+-dependent dioxygenases. PHF2 harbors a plant homeodomain (PHD) and a Jumonji domain. PHF2, via its PHD, binds Lys4-trimethylated histone 3 in submicromolar affinity and has been reported to have the demethylase activity of monomethylated lysine 9 of histone 3 in vivo. However, we did not detect demethylase activity for PHF2 Jumonji domain (with and without its linked PHD) in the context of histone peptides. We determined the crystal structures of PHF2 Jumonji domain in the absence and presence of additional exogenous metal ions. When Fe2+ or Ni2+ was added at a high concentration (50 mM) and allowed to soak in the preformed crystals, Fe2+ or Ni2+ was bound by six ligands in an octahedral coordination. The side chains of H249 and D251 and the two oxygen atoms of N-oxalylglycine (an analog of α-ketoglutarate) provide four coordinations in the equatorial plane, while the hydroxyl oxygen atom of Y321 and one water molecule provide the two axial coordinations as the fifth and sixth ligands, respectively. The metal binding site in PHF2 closely resembles the Fe2+ sites in other Jumonji domains examined, with one important difference—a tyrosine (Y321 of PHF2) replaces histidine as the fifth ligand. However, neither Y321H mutation nor high metal concentration renders PHF2 an active demethylase on histone peptides. Wild type and Y321H mutant bind Ni2+ with an approximately equal affinity of 50 μM. We propose that there must be other regulatory factors required for the enzymatic activity of PHF2 in vivo or that perhaps PHF2 acts on non-histone substrates. Furthermore, PHF2 shares significant sequence homology throughout the entire region, including the above-mentioned tyrosine at the corresponding iron-binding position, with that of Schizosaccharomyces pombe Epe1, which plays an essential role in heterochromatin function but has no known enzymatic activity. 相似文献

In recent years, the Lower Cretaceous (Aptian) Xiagou Formation has yielded approximately 100 avian partial skeletons, many with soft‐tissue traces, from sites in the Changma Basin of Gansu Province, north‐western China. The most abundant taxon amongst these is the ornithuromorph Gansus yumenensis, but enantiornithines have also been identified in the sample. Here we describe two incomplete, semi‐articulated appendicular skeletons, the first consisting of a partial left pelvic girdle and complete pelvic limb, and the second comprised of a nearly complete right pelvic limb. Both specimens bear characteristics diagnostic of Enantiornithes, and are referred to a new taxon, Qiliania graffini gen. et sp. nov. The exceptional, three‐dimensional preservation of these specimens (compared to the crushed, nearly two‐dimensional condition of most other Early Cretaceous avian fossils) reveals new information regarding enantiornithine anatomy, evolution, and diversity. © 2011 The Linnean Society of London, Zoological Journal of the Linnean Society, 2011, 162 , 201–219. 相似文献

We collected gravid king ratsnakes (Elaphe carinata) from three geographically separated populations in Chenzhou (CZ), Lishui (LS) and Dinghai (DH) of China to study the geographical variation in female reproductive traits and trade‐offs between the size and number of eggs. Not all reproductive traits varied among the three populations. Of the traits examined, five (egg‐laying date, post‐oviposition body mass, clutch size, egg mass and egg width) differed among the three populations. The egg‐laying date, ranging from late June to early August, varied among populations in a geographically continuous trend, with females at the most northern latitude (DH) laying eggs latest, and females at the most southern latitude (CZ) laying eggs earliest. Such a trend was less evident or even absent in the other traits that differed among the three populations. CZ and DH females, although separated by a distance of approximately 1100 km as the crow flies, were similar to each other in most traits examined. LS females were distinguished from CZ and DH females by the fact that they laid a greater number of eggs, but these were smaller. ? Gibberellin (GA) deficiency resulting from the na mutation in pea (Pisum sativum) causes a reduction in nodulation. Nodules that do form are aberrant, having poorly developed meristems and a lack of enlarged cells. Studies using additional GA-biosynthesis double mutants indicate that this results from severe GA deficiency of the roots rather than simply dwarf shoot stature. ? Double mutants isolated from crosses between na and three supernodulating pea mutants exhibit a supernodulation phenotype, but the nodule structures are aberrant. This suggests that severely reduced GA concentrations are not entirely inhibitory to nodule initiation, but that higher GA concentrations are required for proper nodule development. ? na mutants evolve more than double the amount of ethylene produced by wild-type plants, indicating that low GA concentrations can promote ethylene production. The excess ethylene may contribute to the reduced nodulation of na plants, as application of an ethylene biosynthesis inhibitor increased na nodule numbers. However, these nodules were still aberrant in structure. ? Constitutive GA signalling mutants also form significantly fewer nodules than wild-type plants. This suggests that there is an optimum degree of GA signalling required for nodule formation and that the GA signal, and not the concentration of bioactive GA per se, is important for nodulation. 相似文献

建立长期增加或减少年轻小鼠切牙的咀嚼刺激模型.通过形态学的观察,研究小鼠下颌骨和咬肌的变化以及大脑皮层和海马的厚度改变;通过行为学实验(旷场实验、高架十字迷宫、新异物体识别和Morris水迷宫实验)观察小鼠认知能力和焦虑情绪的变化;通过高效液相色谱(HPLC)检测小鼠皮层、海马中4种单胺类神经递质(去甲肾上腺素(NE)、肾上腺素(E)、多巴胺(DA)和5-羟色胺(5-HT))的变化,以探讨小鼠焦虑情绪改变的机理;通过荧光实时定量PCR检测小鼠皮层和海马与认知紧密相关的4种基因(BDNF、Synapsin I、NR2B和CREB)的mRNA表达,以研究小鼠认知能力改变的机理.形态学结果显示:各组小鼠下颌骨和咬肌纤维的形态没有影响,各组小鼠皮层和海马的厚度也没有差异.行为学实验结果显示:咀嚼增加组小鼠焦虑倾向低于对照组和咀嚼减少组,各组小鼠短期记忆能力没有统计学差异,咀嚼增加组小鼠空间认知能力优于咀嚼减少组.高效液相色谱结果显示,咀嚼减少组小鼠与咀嚼增加组和对照组相比,去甲肾上腺素在皮层中明显升高(P<0.05).mRNA检测结果显示,咀嚼减少组小鼠4种与认知相关基因的表达量与咀嚼增加组相比都明显下调(P<0.05).上述结果提示,切牙咀嚼刺激的长期增加可以提高小鼠的自主运动能力,降低焦虑程度,提高空间认知能力,上调海马中认知相关基因的表达.切牙咀嚼刺激的长期减少,会降低小鼠的自主运动能力,下调认知相关的基因表达,增加大脑皮层中去甲肾上腺素的含量. 相似文献

The thermal environment during development influences many aspects of the phenotype of hatchling reptiles. We hypothesized that temperature should differentially affect early incubation stages, in which differentiation dominates over growth, and late incubation stages, characterized by high growth rates. To test this idea, we incubated eggs of wall lizard ( Podarcis muralis ) under three regimes with the same mean temperature (29 °C), one constant and two variable with opposite sequences: first cold (25 °C) and then hot (32 °C), and vice versa. Hatchlings incubated at high temperature during the initial period had shorter hindlimbs and tails than those incubated under the other two temperature regimes and shorter heads than those incubated initially at low temperature. Thus, temperature experienced by embryos during the early external incubation period produced similar phenotypic responses compared to those reported in previous studies for the same constant temperature applied over the whole incubation period. Because female wall lizards select lower body temperatures during pregnancy, an increase of intrauterine retention would extend the time of exposure of developing embryos to suitable temperatures. Diminution of body temperature during pregnancy is contrary to the expected pattern under the hypothesis that egg retention has evolved to accelerate development, as proposed by the cold-climate model for evolution of viviparity in squamates, and the results of the present study support the alternative hypothesis of developmental optimization as a special case of the broader maternal manipulation view. © 2007 The Linnean Society of London, Biological Journal of the Linnean Society , 2007, 92 , 441–447. 相似文献
