We observed a wide distribution of the carbohydrate epitopes galactosyl alpha(1-3)galactose (gal alpha1-3 gal), alpha-glucoside and alpha-mannoside in mono- and heteroxenic trypanosomatids by using fluorescein-labelled lectins of Euonymus europaeus (EE) and Concanavalin A (Con A) as well as sera from acute chagasic patients who have very high levels of anti-gal alpha(1-3)gal antibodies. The direct fluorescence test for gal alpha1-3 gal with EE was positive at minimum concentrations of 6 micrograms/ml for heteroxenic trypanosomatids and 0.7 micrograms/ml for monoxenic ones and for the plant parasite, Phytomonas. On the other hand, heteroxenic trypanosomatids that infect vertebrates bound ten-fold more Con A than monoxenic flagellates and Phytomonas. These data were confirmed in ELISA and Western Blot assays carried out with peroxidase-labelled EE and Con A. Euonymus europaeus recognized several glycoproteins in all trypanosomatids that we tested. Con A, however, recognized a glycoprotein cluster in heteroxenic protozoa, which ranging from 60-120 kDa, seemed to lack monoxenic parasites and Phytomonas. These findings suggest that alpha-D-mannose and alpha-D-glucose might play an important role in the interaction between trypanosomatids and vertebrate hosts.  相似文献

Routine oxygen consumption and blood glucose were determined from freshwater fishes, Prochilodus scrofa and Cyprinus carpio, exposed at high temperatures for 1 hr. Prochilodus scrofa had a significantly higher rate of oxygen consumption at 30 degrees C than at 25 degrees C, and carp higher at 25 degrees C than at 30 degrees C. Blood glucose was significantly higher for Cyprinus carpio than for Prochilodus scrofa at 25 and 30 degrees C; however, after exposure to these temperatures for 1 hr blood glucose did not change significantly for both species. The results suggest that these interspecific variations may be linked to the differences between native and foreign fishes and their way of life.  相似文献

  1. Detailed understanding of the mechanisms enabling or limiting the impacts of invasive generalist predators is needed. Harmonia axyridis is an invasive generalist coccinellid predator that may have destabilised coccinellid communities worldwide and is an excellent candidate for investigating invasion dynamics.
  2. We evaluated four hypotheses: (1) Nine years after being detected in Central Brazil, Ha. axyridis has dominated the coccinellid community. This is facilitated by (2) its broader aphid prey range than the other aphidophagous coccinellids, (3) its superiority as an asymmetrical intraguild predator, and (4) the invasibility of the present coccinellid community.
  3. We sampled the invertebrate communities associated with six organic vegetable farms in Brazil during 2017 and 2018, conducted a feeding trial to measure prey niche breadth and overlap, and estimated intra- and interspecific interaction strengths corresponding to first-order Lotka-Volterra parameters to evaluate the strength of intraguild predation and create a community matrix.
  4. We found (1) Ha. axyridis comprised ≤8.3% of the coccinellid community. (2) Hi. convergens had the broadest prey niche and dominated the prey niche of Ha. axyridis. (3) We could not determine from the population data if Ha. axyridis was the dominant intraguild predator because its density was too low and constant. (4) The community matrix indicated that the present-day coccinellid community was stable.
  5. We conclude Ha. axyridis has not become invasive in Central Brazil, in part because it does not have the broadest prey niche breadth and the present coccinellid community is stable to the present perturbation of Ha. axyridis.

The spread of antibiotic-resistant bacteria is a threatening risk for human health at a global scale. Improved knowledge on the cycle of antibiotic resistance spread between human and the environment is a major requirement for the management of the current crisis. Compared to the well-studied cycle in clinical settings much less is known about the factor allowing their persistence in the environment. In fact, lakes and rivers exposed to high anthropogenic impact seem to become long-term reservoirs for resistance genes. The presence of antibiotic resistance genes (ARGs) within the resident microbiome of large subalpine lakes (i.e. Lake Geneva, Lake Maggiore) has recently been investigated in both the water column and the sediment. These studies suggest a correlation between the abundance of some ARGs and the anthropogenic impact. Within the system, however, abiotic factors and the food-web structure determine the survival of specific bacterial genotypes and thus the resistance genes they harbour. Thus, a growing body of work suggests an important role of ecological interactions in the persistence or elimination of such genes from the environment. This article reviews the current literature regarding the presence of ARGs in subalpine lakes, the impact of anthropogenic pollution on their appearance and the potential role of various ecological interactions on their persistence in the system.  相似文献

The NSP-interacting kinase (NIK) receptor-mediated defense pathway has been identified recently as a virulence target of the geminivirus nuclear shuttle protein (NSP). However, the NIK1–NSP interaction does not fit into the elicitor–receptor model of resistance, and hence the molecular mechanism that links this antiviral response to receptor activation remains obscure. Here, we identified a ribosomal protein, rpL10A, as a specific partner and substrate of NIK1 that functions as an immediate downstream effector of NIK1-mediated response. Phosphorylation of cytosolic rpL10A by NIK1 redirects the protein to the nucleus where it may act to modulate viral infection. While ectopic expression of normal NIK1 or a hyperactive NIK1 mutant promotes the accumulation of phosphorylated rpL10A within the nuclei, an inactive NIK1 mutant fails to redirect the protein to the nuclei of co-transfected cells. Likewise, a mutant rpL10A defective for NIK1 phosphorylation is not redirected to the nucleus. Furthermore, loss of rpL10A function enhances susceptibility to geminivirus infection, resembling the phenotype of nik1 null alleles. We also provide evidence that geminivirus infection directly interferes with NIK1-mediated nuclear relocalization of rpL10A as a counterdefensive measure. However, the NIK1-mediated defense signaling neither activates RNA silencing nor promotes a hypersensitive response but inhibits plant growth and development. Although the virulence function of the particular geminivirus NSP studied here overcomes this layer of defense in Arabidopsis, the NIK1-mediated signaling response may be involved in restricting the host range of other viruses.  相似文献
