When constraints on antipredator coloration shift over the course of development, it can be advantageous for animals to adopt different color strategies for each life stage. Many caterpillars in the genus Papilio exhibit unique ontogenetic color sequences: for example, early instars that masquerade as bird feces, with later instars possessing eyespots. I hypothesize that larvae abandon feces masquerade in favor of eyespots due to ontogenetic changes in signaler size. This ontogenetic pattern also occurs within broader seasonal shifts in background color and predator activity. I conducted predation experiments with artificial prey to determine how potential signaling constraints (specifically size and season) shape predation risk, and consequently the expression of ontogenetic color change in Papilio larvae. Seasonally, both predation and background greenness declined significantly from July to September, though there was little evidence that these patterns impacted the effectiveness of either color strategy. Caterpillar size and color strongly affected the attack rate of avian predators: attacks increased with prey size regardless of color, and eyespotted prey were attacked more than masquerading prey overall. These results may reflect a key size-mediated tradeoff between conspicuousness and intimidation in eyespotted prey, and raise questions about how interwoven aspects of behavior and signal environment might maintain the prevalence of large, eyespotted larvae in nature.  相似文献

Wnt/β-catenin signaling plays a major role in embryonic development and adult stem cell maintenance. Reduced activation of the Wnt/β-catenin pathway underlies neurodegenerative disorders and aberrations in bone formation. Screening of a small molecule compound library with a β-galactosidase fragment complementation assay measuring β-catenin nuclear entry revealed bona fide activators of β-catenin signaling. The compounds stabilized cytoplasmic β-catenin and activated β-catenin-dependent reporter gene activity. Although the mechanism through which the compounds activate β-catenin signaling has yet to be determined, several key regulators of Wnt/β-catenin signaling, including glycogen synthase kinase 3 and Frizzled receptors, were excluded as the molecular target. The compounds displayed remarkable selectivity, as they only induced β-catenin signaling in a human osteosarcoma U2OS cell line and not in a variety of other cell lines examined. Our data indicate that differences in cellular Wnt/β-catenin signaling machinery can be exploited to identify cell type-specific activators of Wnt/β-catenin signaling.  相似文献

Dry phospholipid bilayers are known to undergo transient changes in permeability during rehydration. In this review, we present evidence from which we suggest that this permeability change is due to a gel to liquid-crystaline phase transition accompanying rehydration. If the transition is avoided, as in lipids that remain in gel phase whether dry or rehydrated, the problem of leakage during rehydration is obviated, at least in part. Further, the evidence that the transition temperature for dry bilayers can be depressed by certain sugars is discussed. Finally, we show that these principles can be extended to intact cells. Using pollen grains as a model, we have measured the transition temperature for membrane phospholipids and show that the transition is correlated with physiological measurements including permeability changes and subsequent germination. From theT m values taken from pollen grains at different water contents, we have constructed a phase diagram for the intact pollen that has high predictive value for physiological properties.  相似文献

In situ Fourier transform infrared microspectroscopy was used to study the heat stability of proteins and hydrogen bonding interactions in dry maturation-defective mutant seeds of Arabidopsis thaliana . α-Helical, turn and β-sheet conformations were the major protein secondary structures in all of these seeds. On heating, intermolecular extended β-sheet structures, typical of protein denaturation, were formed in abscisic acid-insensitive ( abi3 ) and leafy cotyledon ( lec ) mutant seeds. Proteins in dry wild-type seeds did not denature up to 150°C, but those in dry desiccation-sensitive, lec1–1 , lec1–3 and abi3–5 seeds did at 68, 89 and 87°C, respectively. In the desiccation-tolerant abi3–7 and abi3–1 seeds, denaturation commenced above 120 and 135°C, respectively. Seeds of the aba1–1 abi3–1 double mutant showed signs of denaturation already upon drying. The molecular packing in the seeds was studied by observing the shift in the position of the OH-stretching vibration band with temperature. The maximal rate of change of this band with temperature was much higher in the desiccation-sensitive abi3–5 , aba1–1 abi3–1 , lec1–1 , and lec1–3 mutant seeds than in the desiccation-tolerant wild-type, abi3–1 , abi3–7 , and lec2–1 seeds. We interpret this to mean that the molecular packing density is higher in dry desiccation-tolerant than in dry desiccation-sensitive seeds, which is associated with a higher or lower protein denaturation temperature, respectively. The results are discussed in relation to the physiological and biochemical characteristics of these mutant seeds.  相似文献

Proteins secreted from adipose tissue are increasingly recognized to play an important role in the regulation of glucose metabolism. However, much less is known about their effect on lipid metabolism. The fasting-induced adipose factor (FIAF/angiopoietin-like protein 4/peroxisome proliferator-activated receptor gamma angiopoietin-related protein) was previously identified as a target of hypolipidemic fibrate drugs and insulin-sensitizing thiazolidinediones. Using transgenic mice that mildly overexpress FIAF in peripheral tissues we show that FIAF is an extremely powerful regulator of lipid metabolism and adiposity. FIAF overexpression caused a 50% reduction in adipose tissue weight, partly by stimulating fatty acid oxidation and uncoupling in fat. In addition, FIAF overexpression increased plasma levels of triglycerides, free fatty acids, glycerol, total cholesterol, and high density lipoprotein (HDL)-cholesterol. Functional tests indicated that FIAF overexpression severely impaired plasma triglyceride clearance but had no effect on very low density lipoprotein production. The effects of FIAF overexpression were amplified by a high fat diet, resulting in markedly elevated plasma and liver triglycerides, plasma free fatty acids, and plasma glycerol levels, and impaired glucose tolerance in FIAF transgenic mice fed a high fat diet. Remarkably, in mice the full-length form of FIAF was physically associated with HDL, whereas truncated FIAF was associated with low density lipoprotein. In human both full-length and truncated FIAF were associated with HDL. The composite data suggest that via physical association with plasma lipoproteins, FIAF acts as a powerful signal from fat and other tissues to prevent fat storage and stimulate fat mobilization. A high sodium (HS) intake is associated to increased cardiovascular and renal risk, especially in overweight subjects. We hypothesized that abnormal sodium and fluid handling is involved, independent of hypertension or insulin resistance. Therefore, we studied the relation between BMI and sodium‐induced changes in extracellular fluid volume (ECFV; distribution volume of 125I‐iothalamate) in 78 healthy men, not selected for BMI. A total of 78 subjects with a median BMI of 22.5 (range: 19.2–33.9 kg/m2) were studied after 1 week on a low sodium (LS) diet (50 mmol Na+/d) and after 1 week on HS (200 mmol Na+/d). The change from LS to HS resulted in an increase in ECFV of 1.2 ± 1.8 l. Individual changes in ECFV were correlated to BMI (r = 0.361, P < 0.01). Furthermore, in response to HS, a higher BMI was associated to a higher rise in filtered load of sodium (FLNa+ = [Na+] × GFR, r = 0.281, P < 0.05). Thus, a shift to HS leads to a larger rise in ECFV in healthy subjects with higher BMI, associated with an elevated FLNa+ during HS. Although no hypertension occurred in these healthy subjects, our data provide a potential explanation for the interaction of sodium intake and BMI on cardiovascular and renal risk. Exaggerated fluid retention may be an early pathogenic factor in the cardiorenal complications of overweight.  相似文献

Migration counts can offer a cost-effective method for monitoring the state of migrant raptor populations. However, differential migration strategies between inexperienced juveniles and experienced non-juveniles are rarely accounted for when inferring population trends from raptor migration counts. Since 2011, the Batumi Raptor Count (BRC) monitors the autumn migration of more than 1 million raptors along the eastern Black Sea coast in the Republic of Georgia. We also systematically sampled age information to assess differential migration timing between age groups and estimate age-specific linear trends in abundance between 2011 and 2018 for eight focal species. In so doing we aimed (1) to reassess the global relevance of BRC counts for each species and the potential for monitoring abundance of juveniles and non-juveniles, and (2) to identify demographic changes underlying recent trends in overall abundance. We found that the mean annual passage of non-juveniles at Batumi represents at least 1% of the estimated global breeding population of five study species. As expected, counts of juveniles were more variable than counts of non-juveniles. Yet despite our short monitoring period our models had sufficient statistical power to detect changes in abundance of 10%/year or less for at least one age group in all species except Pallid Harrier Circus macrourus. Our results indicate stable abundance and demography for half of the study species. We also found strong and significant increases in the abundance of Black Kites Milvis migrans and Short-toed Eagles Circaetus gallicus that were primarily due to increasing numbers of non-juveniles. By contrast, juvenile Montagu's Harriers Circus pygargus and Booted Eagles Hieraaetus pennatus significantly decreased in abundance. The first decade of BRC surveys offers an important benchmark for monitoring raptor populations using the eastern African–Palearctic flyway in the 21st century. We discuss possible causes of the observed trends and hope our work will stimulate demographic monitoring at migration count sites.  相似文献

BackgroundThere is limited evidence on the use of high-sensitivity C-reactive protein (hsCRP) as a biomarker for selecting patients for advanced cardiovascular (CV) therapies in the modern era. The prognostic value of mildly elevated hsCRP beyond troponin in a large real-world cohort of unselected patients presenting with suspected acute coronary syndrome (ACS) is unknown. We evaluated whether a mildly elevated hsCRP (up to 15 mg/L) was associated with mortality risk, beyond troponin level, in patients with suspected ACS.Methods and findingsWe conducted a retrospective cohort study based on the National Institute for Health Research Health Informatics Collaborative data of 257,948 patients with suspected ACS who had a troponin measured at 5 cardiac centres in the United Kingdom between 2010 and 2017. Patients were divided into 4 hsCRP groups (<2, 2 to 4.9, 5 to 9.9, and 10 to 15 mg/L). The main outcome measure was mortality within 3 years of index presentation. The association between hsCRP levels and all-cause mortality was assessed using multivariable Cox regression analysis adjusted for age, sex, haemoglobin, white cell count (WCC), platelet count, creatinine, and troponin.Following the exclusion criteria, there were 102,337 patients included in the analysis (hsCRP <2 mg/L (n = 38,390), 2 to 4.9 mg/L (n = 27,397), 5 to 9.9 mg/L (n = 26,957), and 10 to 15 mg/L (n = 9,593)). On multivariable Cox regression analysis, there was a positive and graded relationship between hsCRP level and mortality at baseline, which remained at 3 years (hazard ratio (HR) (95% CI) of 1.32 (1.18 to 1.48) for those with hsCRP 2.0 to 4.9 mg/L and 1.40 (1.26 to 1.57) and 2.00 (1.75 to 2.28) for those with hsCRP 5 to 9.9 mg/L and 10 to 15 mg/L, respectively. This relationship was independent of troponin in all suspected ACS patients and was further verified in those who were confirmed to have an ACS diagnosis by clinical coding. The main limitation of our study is that we did not have data on underlying cause of death; however, the exclusion of those with abnormal WCC or hsCRP levels >15 mg/L makes it unlikely that sepsis was a major contributor.ConclusionsThese multicentre, real-world data from a large cohort of patients with suspected ACS suggest that mildly elevated hsCRP (up to 15 mg/L) may be a clinically meaningful prognostic marker beyond troponin and point to its potential utility in selecting patients for novel treatments targeting inflammation.Trial registrationClinicalTrials.gov - NCT03507309
