Phorid flies (Diptera: Phoridae, Metopinini) are natural enemies of leafcutter ants (Hymenoptera: Formicidae, Attini), which are among the most important pests in the Neotropical region. These parasitoids lay their eggs inside worker ants, causing the death of the parasitized ants, and the oviposition attacks on foraging workers interfere with the collection of vegetal material used in fungal gardens, thereby affecting the whole colony. However, because of the large number of ants per colony, more than one type of biological control agent is needed to have a significant impact. We collected parasitized leafcutter ants Acromyrmex lundii (Guérin‐Méneville), Acromyrmex heyeri (Forel), and Acromyrmex ambiguus (Emery) from various places in Argentina and reared their parasitoids. We recorded developmental times, host and phorid sizes, and other aspects of the biology of the phorids Apocephalus neivai Borgmeier, Apocephalus noetingerorum Brown & Disney, Myrmosicarius gracilipes Borgmeier, Myrmosicarius catharinensis Borgmeier, Myrmosicarius crudelis Borgmeier, and Myrmosicarius cristobalensis Disney et al. This is the first record of A. ambiguus, an important pest of pine plantations, being a host of phorids. We found the first cases of gregarious parasitoidism of a parasitoid with no‐free pupae: M. catharinensis on A. lundii ants. We also evaluated the effect on pupal survival of two cosmopolitan fungi, Beauveria bassiana (Balsamo) Vuill. and Trichoderma lentiforme (Rehm) Chaverri et al., which are also considered as potential biological control agents of leafcutter ants. Suspensions of conidia were tested on Myrmosicarius and Apocephalus parasitoid pupae of different ages. No negative effect on pupal survival was found under either fungal treatment. Therefore, the use of both natural enemies separately but simultaneously, or the introduction of fungi‐inoculated pupae in the pest habitats, seems to be a promising strategy for the multiple biological control of leafcutter ants.  相似文献

It is well established that p16INK4A protein acts as a cell cycle inhibitor in the nucleus. Therefore, cytoplasmic localization of p16 INK4A usually is disregarded by investigators as nonspecific. Three recent studies reported findings that differ from the current view concerning p16INK4A immunohistochemical localization. All three demonstrated that breast and colon cancers expressing cytoplasmic p16INK4 represent distinct biological subsets. We previously detected in a percentage of non-small cell lung carcinomas simultaneous nuclear and cytoplasmic p16INK4A staining. In view of the reports concerning breast and colon carcinomas, we conducted an ultrastructural re-evaluation of our cases to clarify the specificity of p16INK4A cytoplasmic expression. We observed p16 INK4A immunolocalization in both the nucleus and the cytoplasm of a proportion of tumor cells. Diffuse dense nuclear staining was detected in the nucleoplasm, whereas weaker granular immunoreactivity was observed in the cytoplasm near the rough endoplasmic reticulum. Negative tumor cells also were visible. In the tumor-associated stromal, cells p16INK4A immunoreactivity was detected only in the nuclei. We have demonstrated that p16INK4A cytoplasmic staining is specific and suggest that it represents a mechanism of p16INK4A inactivation similar to that observed in other tumor suppressor genes.  相似文献

We investigated the genomic diversity of a local population of the symbiotic bacterium Sinorhizobium medicae, isolated from the roots of wild Medicago lupulina plants, in order to assess genomic diversity, to identify genomic regions influenced by duplication, deletion or strong selection, and to explore the composition of the pan-genome. Partial genome sequences of 12 isolates were obtained by Roche 454 shotgun sequencing (average 5.3 Mb per isolate) and compared with the published sequence of S. medicae WSM 419. Homologous recombination appears to have less impact on the polymorphism patterns of the chromosome than on the chromid pSMED01 and megaplasmid pSMED02. Moreover, pSMED02 is a hot spot of insertions and deletions. The whole chromosome is characterized by low sequence polymorphism, consistent with the high density of housekeeping genes. Similarly, the level of polymorphism of symbiosis genes (low) and of genes involved in polysaccharide synthesis (high) may reflect different selection. Finally, some isolates carry genes that may confer adaptations that S. medicae WSM 419 lacks, including homologues of genes encoding rhizobitoxine synthesis, iron uptake, response to autoinducer-2, and synthesis of distinct polysaccharides. The presence or absence of these genes was confirmed by PCR in each of these 12 isolates and a further 27 isolates from the same population. All isolates had rhizobitoxine genes, while the other genes were co-distributed, suggesting that they may be on the same mobile element. These results are discussed in relation to the ecology of Medicago symbionts and in the perspective of population genomics studies.  相似文献

The healthy synovial lining layer consists of a single cell layer that regulates the transport between the joint cavity and the surrounding tissue. It has been suggested that abnormalities such as somatic mutations in the p53 tumor-suppressor gene contribute to synovial hyperplasia and invasion in rheumatoid arthritis (RA). In this study, expression of epithelial markers on healthy and diseased synovial lining tissue was examined. In addition, we investigated whether a regulated process, resembling epithelial to mesenchymal transition (EMT)/fibrosis, could be responsible for the altered phenotype of the synovial lining layer in RA. Synovial tissue from healthy subjects and RA patients was obtained during arthroscopy. To detect signs of EMT, expression of E-cadherin (epithelial marker), collagen type IV (indicator of the presence of a basement membrane) and alpha-smooth muscle actin (alpha-sma; a myofibroblast marker) was investigated on frozen tissue sections using immunohistochemistry. Fibroblast-like synoviocytes (FLSs) from healthy subjects were isolated and subjected to stimulation with synovial fluid (SF) from two RA patients and to transforming growth factor (TGF)-beta. To detect whether EMT/fibrotic markers were increased, expression of collagen type I, alpha-sma and telopeptide lysylhydroxylase (TLH) was measured by real time PCR. Expression of E-cadherin and collagen type IV was found in healthy and arthritic synovial tissue. Expression of alpha-sma was only found in the synovial lining layer of RA patients. Stimulation of healthy FLSs with SF resulted in an upregulation of alpha-sma and TLH mRNA. Collagen type I and TLH mRNA were upregulated after stimulation with TGF-beta. Addition of bone morphogenetic protein (BMP)-7 to healthy FLS stimulated with SF inhibited the expression of alpha-sma mRNA. The finding that E-cadherin and collagen type IV are expressed in the lining layer of healthy and arthritic synovium indicates that these lining cells display an epithelial-like phenotype. In addition, the presence of alpha-sma in the synovial lining layer of RA patients and induction of fibrotic markers in healthy FLSs by SF from RA patients indicate that a regulated process comparable to EMT might cause the alteration in phenotype of RA FLSs. Therefore, BMP-7 may represent a promising agent to counteract the transition imposed on synoviocytes in the RA joint.  相似文献

We studied the developmental performance of the large morph of Pseudacteon nocens Borgmeier (Diptera: Phoridae), a prospective biological control agent of imported fire ants (Hymenoptera: Formicidae). We measured selected life history traits of this parasitoid as a function of 1) host species (Solenopsis invicta Buren versus Solenopsis richteri Forel), 2) temperature (22 versus 28 degrees C), 3) source population of the fly (Corrientes and Santiago del Estero, Argentina), and 4) varied size distributions of offered host ants. Developmental periods were influenced by host species, although the populations responded in opposing manners. Developmental times, however, were most strongly influenced by temperature with total developmental periods lengthened by 17-32% at 22 degrees C. Pupal mortality was also significantly lower at this temperature. Although numbers of progeny per female were significantly higher for the Corrientes population, we found no significant differences in progeny per female according to host species. Interestingly, we found that females were larger than males, and flies from Corrientes were larger than those from Santiago del Estero, even after statistical adjustments for host size. The modal frequency of host size elected in all treatment combinations tested was identical (0.6 mm), a size that represented the apparent threshold for producing female progeny. These laboratory and additional field observations demonstrate considerable interpopulational variation in P. nocens and lend further support to the applied approach focusing at the population, as opposed to the species level, with respect to both source and target areas for classical biological control introductions of Pseudacteon flies.  相似文献

ProADD, a database for protein aggregation diseases, is developed to organize the data under a single platform to facilitate easy access for researchers. Diseases caused due to protein aggregation and the proteins involved in each of these diseases are integrated. The database helps in classification of proteins involved in the protein aggregation diseases based on sequence and structural analysis. Analysis of proteins can be done to mine patterns prevailing among the aggregating proteins.

The objective of the study was to examine salivary biomarker response to fluid consumption in exercising athletes. Exercise induces stress on the body and salivary alpha amylase (sAA) and salivary cortisol are useful biomarkers for activity in the sympathoadrenal medullary system and the hypothalamic pituitary adrenal axis which are involved in the stress response. Fifteen college students were given 150 ml and 500 ml of water on different days and blinded to fluid condition. The exercise protocol was identical for both fluid conditions using absolute exercise intensities ranging from moderate to high. Saliva was collected prior to exercise, post moderate and post high intensities and analyzed by Salimetrics assays. Exercise was significant for sAA with values different between pre-exercise (85 ± 10 U · ml−1) and high intensity (284 ± 30 U · ml−1) as well as between moderate intensity (204 ± 32 U · ml−1) and high intensity. There was no difference in sAA values between fluid conditions at either intensity. Exercise intensity and fluid condition were each significant for cortisol. Cortisol values were different between pre-exercise (0.30 ± 0.03 ug · dL−1) and high intensity (0.45 ± 0.05 ug · dL−1) as well as between moderate intensity (0.33 ± 0.04 ug · dL−1) and high intensity. Moderate exercise intensity cortisol was lower in the 500 ml condition (0.33 ± 0.03 ug · dL−1) compared with the 150 ml condition (0.38 ± 0.03 ug · dL−1). This altered physiological response due to fluid consumption could influence sport performance and should be considered. In addition, future sport and exercise studies should control for fluid consumption.  相似文献
