The gene sequences of the second largest subunits of RNA polymerases I and II of Euplotes octocarinatus, RPA2 and RPB2, were determined and compared to the respective known sequences of Saccharomyces cerevisiae. The similarity of the derived polypeptide sequences permitted their assignment to the respective polymerases and allowed the comparison of the zinc binding regions. In frame TGA codons were detected, which are likely to encode conserved cysteinyl residues in the putative zinc-finger region of the RPA2 gene. They were also found in other positions in both the RPA2 and RPB2 genes. The RPB2 gene contains a 30 bp intron close to the 5'-end of its coding region. The 5'-ends of the coding regions of all three genes encoding the largest subunits of the three different polymerases were also analyzed. The zinc finger structures again show the use of TGA codons for conserved cysteinyl residues in two of the genes. An N-terminal intron is located in the RPB1 gene at a conserved position as compared to the respective genes of several other eucarya.  相似文献

Summary Splenic tissue of human fetuses from the 14th to the 24th week of gestation (menstrual age) were investigated by light- and electron microscopy to describe the development of the red and white pulp in close relationship to the differentiation of the vascular tree. Special interest is focussed on the differentiation of the T-cell- and the B-cell regions and their specific stationary cells.The preliminary stage, here called the primary vascular reticulum, lasts up to the 14th gestational week (gw). Numerous erythrocytes, normoblasts and macrophages are seen among a network of mesenchymal cells and argyrophilic fibers. Hematopoiesis, especially erythropoiesis, can be recognized.The characteristic organ structure becomes established during the subsequent transformation stage of the fetal spleen, beginning with the 15th gw. Splenic lobules begin to form during the 15th to 17th gw. They consist of a central artery, surrounded by a sheath of lightly stained stationary cells which resemble myofibroblasts. At the periphery of these lobules the red pulp forms. Initially mobile cells are distributed throughout the reticulum. Soon they begin to accumulate in the venous sinuses, which develop from lacunae among the reticular network and come into contact with the venous system. The endothelial wall of these sinuses remains discontinuous, confirming the theory of the open vascularization of the spleen. The development of the larger veins is correlated with the differentiation of the splenic trabeculae.The development of the white pulp is correlated with the stage of lymphoid colonization within the spleen, beginning around the 18th gw. An accumulation of lymphocytes around the central arteries can be recognized during the 19th and 20th gw. These lymphoid cells show morphological and immunohistochemical characteristics of T-precursor cells. Within the now assembling periarterial lymphoid sheath (PALS) a few precursors of interdigitating cells (IDC) are recognizable, giving evidence for the differentiation of the T-cell region.Around the 23rd gw the assemblage of primary follicles is discernible at the periphery of the PALS. Precursors of the follicular dendritic reticulum cell (FDRC), the specific stationary cell of the B-cell region, have been recognized. Zusammenfassung Die Aufnahme von i. v. injiziertem 14C- und 3H- markiertem Noradrenalin (10–400 g/kg Körpergewicht) wurde an Ratten a) nach Verabreichung von Desmethylimipramin und b) von Normetanephrin autoradiographisch untersucht. a) Desmethylimipramin-HCl (2–40 mg/kg Ratte) bewirkte eine starke Hemmung der intraneuralen Aufnahme und Stapelung von Noradrenalin. Trotzdem wurden das Myokard und andere extraneurale Organgewebe radioaktiv markiert. b) Normetanephrin (33 mg/kg Körpergewicht) verminderte lediglich die Aufnahme von radioaktiv markiertem Noradrenalin in die Myokardfasern, während die intraneurale Aufnahme unbeeinflußt blieb. Beide Substanzen verbesserten die periphere Durchblutung nach toxischen Dosen von Noradrenalin.
Inhibition of the uptake of norepinephrine by desmethylimipramine and normetanephrine, an autoradiographic investigation
Summary The uptake of 14C- or 3H-labelled norepinephrine (dose 10 to 400 g/kg body weight) injected intravenously into rats was studied after the application a) of desmethylimipramine and b) of normetanephrine by autoradiography. a) Desmethylimipramine-HCl (2–40 mg/kg body weight) caused a strong inhibition of the intraneuronal uptake and storage of norepinephrine. b) Normetanephrine (dose 33 mg/kg body weight) on the other side diminished only the uptake into the myocardial fibers without an effect of the intraneuronal uptake. Both substances, desmethylimipramine and normetanephrine, improved the peripheral circulation after toxic doses of norepinephrine.

Summary Paired vessels, consisting of one artery and one vein, have so far only been described in the brains of some lower vertebrates and in the brains of marsupials (opossum). However, as shown in this paper, similar vessels also occur in the medulla oblongata, midbrain, hypothalamus, septum pellucidum, hippocampus and in the olfactory lobe of the cat's brain. In the neocortex and in the cerebellum no such vessels could be traced.—Whereas some paired vessels are first surrounded by connective tissue (Virchow-Robin's space) and then by a layer of glial fibres, most of the vessels are only ensheathed by a coat of glial fibres. These and other histological details are described and some functional implications are discussed under the assumption that the paired vessels allow diffusion of blood gases between venous and arterial blood and vice versa.

Zusammenfassung Die Körpergröße von Aphelinus semillavus How. variiert in Abhängigkeit von Große und Qualität des Wirtes. Tiere aus dem gesamten Variationsbereich wurden auf größenabhängige Proportionsverschiebungen untersucht.Relative Länge und Breite des Kopfes (bezogen auf die Körperlange) nehmen mit zunehmender Körperlange ab. Die relative Lange des Abdomens wächst etwa um den gleichen Betrag:Es besteht eine negative Korrelation zwischen der relativen Länge des letzten Fühlergliedes, des Trägers wichtiger Sinnesorgane, und der Körperlange. Seine Oberfläche steht in einem etwa konstanten Verhältnis zur Körperlänge. Die Anzahl der Porenplatten nimmt mit der Körperlänge nur wenig, ihre Länge deutlicher zu.Im Komplexauge wächst die Anzahl der Ommatidien etwa proportional zur Korperlange, während seine gesamte Fläche zwar linear, aber ïiberproportional ansteigt, was durch zunehmende Große der einzelnen Facetten erreicht wird.Die Länge der Vorderfliigel ist bei Männchen und Weibchen von mehr als 1 mm Länge etwa der Körperlange proportional. Unterhalb 1 mm nimmt die relative Flügellänge mit der Körperlange ab, bis die Flügel nur noch funktionslose Rudimente bilden. Die Relation Flügel-fläche:Körpergewicht ist bei mittelgroßen Tieren am günstigsten.Jedes Ovar hat stets drei Ovariolen, unabhängig von der Körperlänge. Große Weibchen legen nur etwas größere, aber bedeutend mehr Eier als kleine. Die Anzahl der (am 7.–10. Lebenstag) täglich abgelegten Eier steigt etwa linear mit dem Logarithmus des Körpergewichts an.Die Eiablage wird während des ganzen Lebens (im Labor etwa 40 Tage) fortgesetzt, ist aber zum. Ende hin etwas geringer. Relativ zum Körper-gewicht produzieren mittelgroße Weibchen die größte Eimasse. Die Anzahl der zum Nahrungsgewinn ausgesaugten Läuse steigt mit dem Körpergewicht.Zwischen der relativen Länge des Legestachels und der Körperlange besteht eine fast lineare negative Korrelation.Die gröBenabhangigen Proportionsverschiebungen werden als Auswirkungen unterschiedlicher Wachstumstendenzen der einzelnen Organe gedeutet, die ihrerseits genetisch fixiert sind und den biologischen Sinn haben, die Lebensf ähigkeit modifikatorisch kleiner Exemplare zu sichern und damit die Ausnutzung entsprechend kleiner Wirte zu ermöglichen.Die Bedeutung der Proportionsverschiebungen für die Leistung der ganzen Population wird diskutiert. Sic ist abhängig von der Häufigkeits-verteilung der verschiedenen Körpergrößen und these wiederum von der Art-Zusammensetzung und Ernährung der Wirtsfauna.
Summary The body size of Aphelinus semiflavus How. varies with the size and the quality of the hosts. Specimens from the whole range of variation were investigated for differences between organ proportions, which depended on size.The relative length and width of the head (based on the body length) are reduced with increasing body length. The relative length of the abdomen increases approximately by the same amount.There is a negative correlation between the relative length of the last joint of the antennae and the body length. Its surface shows a nearly constant proportion to the body length. The length of the pore plates increases with the body length more distinctly than their numbers.Within the compound eye, the number of ommatidia increases almost proportionally to the body length; the whole area of the eye increases more than proportionally because the size of the single facets also becomes greater.When the body length is greater than 1 mm, the length of the forewings is nearly proportional to the body length. At less than 1 mm, the relative length of the fore-wings is reduced with declining body length until, finally, only rudiments without any function remain. The ratio between wing area and body weight is most favourable at medium body size.Three ovarioles, independent of the body size, are present in each ovary. Big females produce considerably more eggs than small ones. The size of their eggs is only slightly enlarged. The daily egg production (at the 7th–10th day of life) increases nearly linearly with the logarithm of body weight. Oviposition is continued throughout the life-time of the insect (ca. 40 days in the laboratory) but it is somewhat reduced at the end. Relative to their body weight, females of medium size produce the biggest egg masses. The number of aphids utilized by the parasite for nourishment increases with the body weight of the female.A negative correlation exists between the relative length of the extended ovipositor and the body length.The proportion differences of the organs, dependent on size, are interpreted to be consequences of different growth tendencies which are genetically fixed. Apparently, the biological role of such different growth tendencies is to ensure the viability of phenotypically small specimens and in this way to make the utilisation of small hosts possible.The importance of the differences of body proportions for the effect of the whole population is discussed. It depends on the frequency distribution of the different body lengths which itself is the consequence of the composition of host species and the nutriation of the hosts.

Botulinum C2 toxin is known to ADP-ribosylate actin. The toxin effect was studied on [3H]noradrenaline secretion of PC12 cells. [3H]Noradrenaline release was stimulated five- to 15-fold by carbachol (100 microM) or K+ (50 mM) and 10-30-fold by the ionophore A23187 (5 microM). Pretreatment of PC12 cells with botulinum C2 toxin for 4-8 h at 20 degrees C, increased carbachol-, K+-, and A23187-induced, but not basal, [3H]noradrenaline release maximally 1.5-to three-fold, whereas approximately 75% of the cellular actin pool was ADP-ribosylated. Treatment of PC12 cells with botulinum C2 toxin for up to 1 h at 37 degrees C also increased stimulated [3H]noradrenaline secretion, whereas toxin treatment for greater than 1 h decreased the enhanced [3H]noradrenaline release stimulated by carbachol and K+ but not by A23187. Concomitantly with toxin-induced stimulation of secretion, 20-50% of the cellular actin was ADP-ribosylated, whereas greater than 60% of actin was modified when exocytosis was attenuated. The data indicate that ADP-ribosylation of actin by botulinum C2 toxin largely modulates stimulation of [3H]noradrenaline release. Moreover, the biphasic toxin effects suggest that distinct mechanisms are involved in the role of actin in secretion.  相似文献

Summary Vitamin E levels in serum, liver and blubber (subcutaneous adipose tissue) were determined for 66 male and female grey seals of varying age in the pupping colony on Sable Island in the Northwest Atlantic by high-performance liquid chromatography (HPLC). Fatty acid concentrations were determined for all blubber specimens. Adult males and pups had significantly higher levels of vitamin E and cholesterol in serum than females and juveniles. A close relationship between vitamin E and cholesterol in serum could be observed. Suckling pups had significantly higher levels of vitamin E in liver (191 mg·kg–1) than juveniles and adults (21–41 mg·kg–1). Levels of vitamin E in blubber showed an age-dependent increase, with the highest levels being found in adult males; overall, these levels were much lower than in man. Vitamin E levels in blubber and liver of lactating females were only half that of adult males. This might be due to an intensive transfer of vitamin E from mother to pup during lactation, a process which may also explain the much higher levels of vitamin E in serum and liver of nursing pups. The low levels of vitamin E in blubber of seals might be a result of its high percentage of unsaturated fatty acids (79%). Highest percentage was represented by 18:1, 16:1, 22:6 and 16:0. Pups had lower values of monounsaturated, and a higher percentage of saturated fatty acids compared to mothers.  相似文献

The lactose permease is being used as a model system for the rational redesign of a membrane protein with the goal of increasing the likelihood of crystallization. Various modifications to the protein have been added for the purposes of purification, stability, and potential for crystallization. The addition of six consecutive histidines at the C-terminus of the protein allows for the rapid purification by nickel-chelate chromatography, and the insertion of an entire protein domain into one of the inner cytoplasmic loops of the permease gives the resulting protein a larger hydrophilic surface area. The increase in polar surface area makes the fusion protein easier to handle and more likely to crystallize. In particular, the introduction of cytochromeb562 ofE. coli into the central hydrophilic domain of the lac permease results in a fusion protein with the transport activity of the permease and the visible absorbance spectrum of the cytochrome. The red permease is very easy to monitor through the steps of expression, purification, concentration, and crystallization.  相似文献

Summary Despite extensive discussions during the last 20 years stromatolites are still used by many geologists as unequivocal indicators of very shallow-water conditions. We investigated four stratigraphic units from the Lower and Middle Jurassic of southern Germany (Posidonien-Schiefer, Amaltheen-Ton) and of the Northern Calcareous Alps (Adneter Kalk, Klauskalk), which were formerly interpreted as shallow marine sediments by some authors due to the occurrence of stromatolites. Our interpretations of the macro-, micro- and ultrafacies of these sediments are not compatible with shallow-water settings. We therefore propose a deep-marine, aphotic origin of these stromatolites. Former interpretations of the Posidonien-Schiefer as a shallow-water deposit are mainly based on the occurrence of stromatolites. We favour the model of a temporarily stagnant, deep, aphotic basin for these planktonrich sediments. Particles resembling ooids, but lying within mudstones cannot be taken as evidence for shallow agitated water. They either formed within the mud or are allochthonous. The deep-water setting of the red limestone of the Alpine Early and Middle Jurassic is indicated by a lack of platform-typical components like coated grains and phototrophic benthos and by shells of plankton and nekton forming a major part of the sediment. Stromatolites occur on the steep slope of a drowned Rhaetian reef with an estimated relief of 50–100 m and immediately below and within radiolarian limestones, deposited below the aragonite compensation depth (ACD). The aphotic stromatolites show some morphological differences to their shallow water counterparts. In all of our sections they occurred during intervals of reduced sedimentation. They form only thin horizons and probably grew very slowly. Mineralizations by Fe−Mn oxides and phosphate are very common. The presence of a microbial film is evident from binding of sedimentary particles, but the nature of the microbes is not known. Growth habits within the very distinct environments of red limestone and black shales show some common features, but also clear differences. The microproblematicumFrutexites Maslov is a very common component in deep-water stromatolites, but may also itself form small crusts or dendrolites. It occurs in two different forms. Opaque, slender forms with indistinct outlines probably grew within the weakly lithified sediment. Thicker, transparent forms with well defined outlines are found in cavities and probably also grew on the seafloor. Well preserved specimens display an internal fabric of radially arranged fibres of Fe−Mn oxides and calcite. It is suggested that calcite or aragonite were one original mineralogy ofFrutexites, which was later replaced by Fe−Mn oxides or phosphate. It is not certain whetherFrutexites is an organic, biomineralized structure or an inorganic mineralization, but the variable mineralogy and growth forms in different environments point to an organic origin. But even if organic, the occurrence in cryptic habitats and negative phototactic growth-directions make it clear thatFrutexites was not phototrophic.  相似文献
