The renal morphology of 24 species of mormoopid and phyllostomid bats feeding on six different diets was examined to test evolutionary changes in several structural traits presumably led by dietary shifts from ancestral insectivorous diets. The kidneys of a fish-eating vespertilionid and an insect-eating emballonurid were also examined but not included in the phylogenetic comparison. The length, width, and breadth of the kidneys were used to calculate relative medullary thickness (RMT). Tissues were processed for stereological analysis, and the volumes of the kidney, nephron components, and vasculature were determined. RMT did not correlate with body mass in either animal-eating or plant-eating phyllostomid and mormoopid bats. The shift from insectivory to frugivory and nectarivory was accompanied by a reduction in RMT, a reduction in the percent of renal medulla, and an increase in the percent of renal cortex. No changes in these traits were observed in bats that shifted to carnivorous, omnivorous or sanguinivorous habits. No changes were observed in renal vasculature, in the percentage of cortical and medullary nephron components or of capillaries surrounding the nephrons in any feeding group. Vespertilionid and emballonurid species had similar values in all traits examined as compared to insectivorous phyllostomids and mormoopids. Our data suggest that diet does not influence a single area of the nephron, but rather the entire nephron such that the relative amounts of renal cortex and medulla are affected. 相似文献

We report the cloning of the pykA and pykF genes from Escherichia coli, which code for the two pyruvate kinase isoenzymes (ATP:pyruvate 2-O-phosphotransferases; EC 2.7.1.40) in this microorganism. These genes were insertionally inactivated with antibiotic resistance markers and utilized to interrupt one or both pyk genes in the E. coli chromosome. With these constructions, we were able to study the role of these isoenzymes in pyruvate biosynthesis. 相似文献

Volcanic ocean crust contains a global chemosynthetic microbial ecosystem that impacts ocean productivity, seawater chemistry and geochemical cycling. We examined the mineralogical effect on community structure in the aquifer ecosystem by using a four-year in situ colonization experiment with igneous minerals and glasses in Integrated Ocean Drilling Program Hole 1301A on the Juan de Fuca Ridge. Microbial community analysis and scanning electron microscopy revealed that olivine phases and iron-bearing minerals bore communities that were distinct from iron-poor phases. Communities were dominated by Archaeoglobaceae, Clostridia, Thermosipho, Desulforudis and OP1 lineages. Our results suggest that mineralogy determines microbial composition in the subseafloor aquifer ecosystem. 相似文献

The heritable disorder ataxia telangiectasia (AT) is caused by mutations in the AT-mutated (ATM) gene with manifestations that include predisposition to lymphoproliferative cancers and hypersensitivity to ionizing radiation (IR). We investigated gene expression changes in response to IR in human lymphoblasts and fibroblasts from seven normal and seven AT-affected individuals. Both cell types displayed ATM-dependent gene expression changes after IR, with some responses shared and some responses varying with cell type and dose. Interestingly, after 5 Gy IR, lymphoblasts displayed ATM-independent responses not seen in the fibroblasts at this dose, which likely reflect signaling through ATM-related kinases, e.g., ATR, in the absence of ATM function. 相似文献

Polyamine biosynthesis in senescing leaves of Avena sativa L. was measured by determining the activities of arginine decarboxylase (EC 4.1.1.19), ornithine decarboxylase (EC 4.1.1.17) and S-adenosyl-l-methionine decarboxylase (EC 4.1.1.50). Polyamine content was also estimated by thin layer chromatography and high performance liquid chromatography. Arginine decarboxylase activity decreases progressively in aging attached first leaves and in senescing excised leaves in the dark. Conversely, it increases during light exposure of excised leaves, which retards senescence. Ornithine decarboxylase activity is high and constant in the attached leaf, irrespective of age; it decreases in excised leaves kept in the dark and in the light, irrespective of senescence. S-Adenosyl-l-methionine decarboxylase shows no correlation with age or senescence. Levels of putrescine, diaminopropane, agmatine, and spermidine are high in young leaves and decline with age. The best single indicator of senescence is usually spermidine, which decreases in excised leaves incubated in the dark, but increases in such leaves with time of light exposure. Spermidine generally has a reciprocal relationship with putrescine, indicating that spermidine synthase, which converts putrescine to spermidine, may exert important physiological control. We have cloned and characterized the gene PYC1, encoding the unique pyruvate carboxylase in the dimorphic yeast Yarrowia lipolytica. The protein putatively encoded by the cDNA has a length of 1,192 amino acids and shows around 70% identity with pyruvate carboxylases from other organisms. The corresponding genomic DNA possesses an intron of 269 bp located 133 bp downstream of the starting ATG. In the branch motif of the intron, the sequence CCCTAAC, not previously found at this place in spliceosomal introns of Y. lipolytica, was uncovered. Disruption of the PYC1 gene from Y. lipolytica did not abolish growth in glucose-ammonium medium, as is the case in other eukaryotic microorganisms. This unusual growth phenotype was due to an incomplete glucose repression of the function of the glyoxylate cycle, as shown by the lack of growth in that medium of double pyc1 icl1 mutants lacking both pyruvate carboxylase and isocitrate lyase activity. These mutants grew when glutamate, aspartate, or Casamino Acids were added to the glucose-ammonium medium. The cDNA from the Y. lipolytica PYC1 gene complemented the growth defect of a Saccharomyces cerevisiae pyc1 pyc2 mutant, but introduction of either the S. cerevisiae PYC1 or PYC2 gene into Y. lipolytica did not result in detectable pyruvate carboxylase activity or in growth on glucose-ammonium of a Y. lipolytica pyc1 icl1 double mutant. 相似文献

Land-use change alters catchment hydrology by influencing the quality and quantity of partitioned rainfall. We compared rainfall partitioning (throughfall, stemflow and interception) and nutrient concentrations in rainfall, throughfall and stemflow in three land-use types [primary forest (PF), secondary forest (SF) and agriculture (A)] in Panama. Measurements of throughfall were highly variable which may have masked seasonal and land use differences but it was clear that throughfall at agricultural sites made up a larger proportion of gross precipitation than at forest sites. Of incident precipitation, 94% became throughfall in agriculture sites while 83 and 81% of gross precipitation became throughfall in PF and SF, respectively. The size of the precipitation event was the main driver of variation in throughfall and stemflow. Consistent patterns in nutrient cycling were also difficult to identify. Vegetation has a vital role in delivering nutrients as throughfall deposition of K was often larger than precipitation deposition. A canopy budget model indicated that canopy exchange was often more dominant than dry deposition. Throughfall was generally enriched with nutrients, especially K and Mg, with enrichment factors of up to 17 and 5 for K and Mg, respectively, in PF. In contrast, Ca was sometimes taken up by the canopy. Values of nutrient deposition were high (with up to 15, 3, 30 and 15 kg ha?1 month?1 in stand deposition of Ca, Mg, K and Na, respectively in PF), possibly due to the slash-and-burn agricultural practices in the area or marine inputs. The need to replace conventional polymers due to environmental pollution caused by them has led to increased production of biodegradable polymers such as starch. Thus, the application possibilities of starch have increased. In this study, we produced and characterized biodegradable films derived from native and oxidized potato starch. The film-forming solution was prepared with different concentrations of extracted starch (native or oxidized) and a plasticizer (glycerol or sorbitol). Then, the mechanical, barrier, morphological, and structural properties of the films were characterized. The moisture content of the films varied from 15.35?±?1.31 to 21.78?±?0.49%. The elastic modulus of the films ranged from 219?±?14.97 to 2299?±?62.91 MPa. The film of oxidized starch plasticized with sorbitol in the lowest content was the most resistant and flexible; moreover, this film also presented lower water vapor permeability and low solubility in water. Fourier-transform infrared spectroscopic analysis of the biodegradable films indicated the presence of same functional groups as those of starch with bands in the same regions. The film thickness was lower for the films plasticized with glycerol whereas the color variation (Δ?) was lower for the ones plasticized with sorbitol. In case of both plasticizers, the increase in their content decreased the Δ? value. All the biodegradable films presented stability against water absorption owing to their low solubility in water. Morphological evaluation revealed the presence of partially gelatinized starch granules in the films. The roughness parameter (Rq) of the films varied from 3.39 to 10.9 nm, indicating that their surfaces are smooth. X-ray diffraction studies showed a B-type pattern for the starches, which is representative of tubers. Further, the films present higher relative crystallinity (RC) compared to the starches. The biodegradable starch films are uniform, transparent and with low solubility in water. The oxidation of starch and use of sorbitol as a plasticizer resulted in improved properties of the starch films, which is suitable for application. 相似文献
