Update of AMmtDB: a database of multi-aligned Metazoa mitochondrial DNA sequences

The AMmtDB database (http://bighost.area.ba.cnr.it/mitochondriome) has been updated by collecting the multi-aligned sequences of Chordata and Invertebrata mitochondrial genes coding for proteins and tRNAs. Links to the multi-aligned mtDNA intraspecies variants, collected in VarMmtDB at the Mitochondriome web site, have been introduced. The genes coding for proteins are multi-aligned based on the translated sequences and both the nucleotide and amino acid multi-alignments are provided. AMmtDB data selected through SRS can be viewed and managed using GeneDoc or other programs for the management of multi-aligned data depending on the user’s operative system. The multiple alignments have been produced with CLUSTALW and PILEUP programs and then carefully optimized manually.     

Systematic review of Atractus schach (Serpentes: Dipsadidae) species complex from the Guiana Shield with description of three new species

The Guiana Shield harbours one of the best preserved and largest extents of tropical forest on Earth and an immense biodiversity. The herpetofauna of this region remains poorly known. The species-rich snake genus Atractus contains ～140 species, many with complicated taxonomic histories, including A. schach. Examination of specimens in museums and newly collected material from French Guiana has allowed the illustration of hemipenial morphology for the first time and an expanded diagnosis. Concatenated molecular phylogenetic (mitochondrial and nuclear genes) and phenotypic (morphometrics, external and hemipenial morphology) analyses confirm non-monophyly of the A. flammigerus group and indicate that A. schach is a species complex with three new species described here. The geographic distribution of A. schach sensu stricto is restricted to Guiana, Surinam, and French Guiana north of Tumucumaque massif. Populations tentatively assigned to A. schach from the east from French Guiana in the Roura lowlands to Almeirim, and from central Amazonia between the Negro and Trombetas rivers in Brazil are also recognized as new species. Our results suggest that populations from south of the Amazon River are not conspecific with those from the Guiana Shield.

人胰岛素原C肽的合成及专一性抗体制备

利用固相多肽合成中的Boc途径合成了人胰岛素原C肽 ,经TSK柱层析一步纯化 ,产物达HPLC及毛细管电泳均一 ,蛋白质序列分析和质谱分析符合理论值 ,总回收率高达 41 %。为了提高寡肽抗体的专一性 ,我们将丙烯酰化的C肽经催化聚合形成了以聚丙酰为骨架 ,C肽为侧链的聚合体 ,分子量约为 2 5kD。免疫新西兰大白兔 1月 ,获得专一性抗体。用酶联免疫吸附法测得抗血清滴度达 2 .5× 1 0 4 ,与BSA无交叉反应。聚丙酰C肽抗原是制备C肽抗体的一种新的尝试 ,而且也可能成为新的多肽工程疫苗之一 ,为其他传染性疾病多肽疫苗的研究提供新的途径     

脱脂蛋白模型多肽与脂质体相互作用的高效液相色谱研究

应用凝胶过滤高效液相色谱,测定了两个两亲性多肽Ａｍｐ１和Ａｍｐ２进入磷脂酰甘油／磷脂酰胆碱脂双层的表观分配常数,并利用三硝基苯磺酸分析研究了与脂质体结合的多肽的氨基暴露状况。由结果推测,处于膜结合状态的多肽的氨基端是暴露于水相的;Ａｍｐ１与脂双层相互作用强于Ａｍｐ２,一方面表现为Ａｍｐ１比Ａｍｐ２埋膜较深,另一方面表现为Ａｍｐ１与脂双层的结合能力比Ａｍｐ２强,而主要表现在于后者。此外也发现两个多肽在缓冲液中处于几乎不存在暴露的氨基的聚合状态。     

多发性骨髓瘤细胞中一个表达上调基因的克隆与分析

根据GenBank收录的多发性骨髓瘤细胞株 (ARH 77)表达上调ESTAF4 2 5 30 0设计引物 ,运用RT PCR检测了 5例多发性骨髓瘤患者及 4例正常人骨髓细胞中该EST的表达水平 .Northern印迹杂交分析该EST在多种组织中的表达 .进一步利用该EST作探针 ,筛选ARH 77cDNA文库 ,获得全长cDNA克隆 ,对该序列进行了分析 .结果显示 ,该EST在多发性骨髓瘤患者骨髓细胞中亦有较高的表达 ,而在正常人骨髓细胞中低表达 .经测序证实 ,该cDNA全长为 4 5 2bp(GenBank收录号 :AF4 87338) .预测其编码一个 5 7个氨基酸的小分子量蛋白质 ,属于与DNA复制有关的解旋酶 引物酶基因家族的新成员 .该基因在多发性骨髓瘤细胞中表达上调 ,其表达水平的改变可能与多发性骨髓瘤的发生与发展有关     

铁线蕨中间纤维的研究及某些植物类角蛋白的比较分析

应用整装电镜制样技术,结合选择性抽提方法在铁线蕨（ＡｄｉａｎｔｕｍｐｈｉｌｉｐｐｅｎｓｅＬ．）叶细胞中观察到直径１０ｎｍ的纤维网络结构。免疫印迹结果显示：经选择性抽提得到的纤维蛋白与动物角蛋白抗体有免疫交叉反应,间接免疫荧光标记也得到类似结果,而且此类蛋白能在体外自组装为１０ｎｍ或更粗的纤维。说明蕨类植物细胞中存在类角蛋白中间纤维网络。免疫印迹结果表明,螺旋藻（ＳｐｉｒｕｌｉｎａｓｕｂｔｉｌｌｉｓｉｎａＫｕｔｚ．）细胞,地钱（ＭａｒｃｈａｎｔｉａｐｏｌｙｍｏｒｐｈａＬ．）叶状体,铁线蕨（Ａ．ｐｈｉｌｉｐｐｅｎｓｅＬ．）、银杏（ＧｉｎｋｇｏｂｉｌｏｂａＬ．）、白菜（ＢｒａｓｉｃａｐｅｋｉｎｅｎｓｉｓＲｕｐｒ．）的叶组织经选择性抽提后得到的蛋白均与动物角蛋白抗体有免疫交叉反应。其中,螺旋藻仅含有两种类酸性角蛋白,而其余４种植物材料均含有３种类酸性角蛋白及３种类碱性角蛋白。结合以往实验结果,我们认为类角蛋白在植物细胞中是普遍存在的。     

纯化膜蛋白质复合体Cytb6f的新方法

建立了纯化光合膜蛋白质复合体Ｃｙｔｂ６ｆ的新方法。与现有方法相比,新方法简单明了,只包括透析离心和用硫酸铵分级沉淀两个步骤,而且适于大批量制备。按此方法从菠菜叶绿体分离纯化的制剂含９．８ｎｍｏｌＣｙｔｆ·ｍｇ－１;ＳＤＳＰＡＧＥ显示４条主要区带及１条低分子量区带,背景干净;Ｃｙｔｂ６（ｂ型血红素）∶Ｃｙｔｆ＝２∶１（ｍｏｌ∶ｍｏｌ）,Ｃｈｌａ∶Ｃｙｔｆ＝１∶１（ｍｏｌ∶ｍｏｌ）;酶活性（ＰＱ２Ｈ２→Ｃｙｔｃ）８０μｍｏｌＣｙｔｃ·ｎｍｏｌＣｙｔｆ－１·ｈ－１左右;产率可达３８％。

近年来抗菌肽由于自身的优点成为抗生素的替代品 ,并已成为研究与开发的热点 ,其中乳铁蛋白肽因其强大的生物学功能而备受关注。就其结构、功能进行了综述 ,并根据其结构和生物学活性的关系探讨该肽的作用机制 ,同时也提出了乳铁蛋白肽研究中存在的一些问题及相应对策。
