The polypeptides of relative molecular masses (Mr) 22,000, 29,000, and 36,000 represent three major constituents of the chloroplast envelope of spinach (Spinacia oleracea L.) leaves. The Mr 22,000 polypeptide has been localized in the outer membrane, whereas the two other peptides have been attributed to the inner envelope membrane (Joyard et al., 1983). The Mr 29,000 polypeptide has been identified as the "phosphate translocator" (Flügge and Heldt, 1979). In this investigation, we studied the three envelope polypeptides by means of immunocytochemistry. Using indirect immunofluorescence, all three polypeptides were visualized in cryostat sections of formaldehyde-fixed leaf tissue. They were found in both palisade and spongy parenchyma cells and in guard cells, as indicated by a strong fluorescence in the chloroplast periphery. In contrast, fluorescein isothiocyanate or protein A-gold labeling of isolated fixed chloroplasts resulted only in visualization of the Mr 22,000 polypeptide, a constituent of the outer membrane. We further studied the morphological distribution and frequency of this peptide by electron microscopic evaluation of platinum-carbon replicas after freeze-etching or label-fracture and of ultra-thin sections. By use of these three methods, the polypeptide was found to be randomly distributed in the outer envelope membrane and easily accessible to the immunomarker. Average marker density, as obtained by freeze-etching and label-fracture, was approximately 130 gold particles per square micron.  相似文献

Summary A prospective study of structural rearrangements occurring in normal lymphocytes was carried out. For each of two newborns and four young and two old adults, about 1000 metaphases from 72-h and 120 from 48-h cultures were studied. The frequency of rearrangements between bands 7p14, 7q35, 14q11.2 or 14q12 and 14qter, which is on the average about 0.003, is higher in newborns (0.0043) than in adults (0.0024). Conversely, the rearrangements involving other bands, which have a frequency of 0.025 on the average, are more frequent in old adults (f=0.038) than in young adults (f=0.025) and newborns (f=0.013). The first type of rearrangement, which occurs in utero, may correspond to immunoglobulin and related gene rearrangements. The other rearrangements seem to accumulate progressively and may reflect exposure to mutagens. It is import to discriminate these two types of rearrangements when studying the effect of low doses of mutagens.  相似文献

The two human proteins Ki-1/57 and CGI-55 have highly similar amino acid sequences but their functions are unknown. We analyzed them by yeast two-hybrid screens and found that they interact with the C-terminal region of the human chromatin-remodeling factor CHD-3 (chromo-helicase-DNA-binding domain protein-3). The interaction of CGI-55 and CHD-3 could be confirmed in vitro and in vivo by co-immunoprecipitations from Sf9 insect cells. Mapping showed that CGI-55 interacts with CHD-3 via two regions at its N- and C-terminals. The CGI-55 and Ki-1/57 mRNAs show highest expression in muscle, colon and kidney. A CGI55-GFP fusion protein was localized in the cytoplasm, nucleus and perinuclear regions of HeLa cells. These data suggest the possibility that CGI-55 and Ki-1/57 might be involved in nuclear functions like the remodeling of chromatin.  相似文献

Individual variation in postjuvenile molt in male Black Redstart is pronounced with about 90% of young males retaining female‐like coloration (cairei plumage type) and about 10% acquiring adult male‐like feathers (paradoxus plumage type). We examined whether autumn migration timing and body condition differed between individuals of the two plumage types. We used the data of 10,977 Black Redstarts captured during autumn at a ringing site in northern Switzerland where a protocol to record plumage types of captures has been applied since 1980. As cairei individuals cannot be distinguished from young females while sexing is comparatively easy for paradoxus individuals, the proportion of missing data on sex was likely to be higher for cairei individuals than for paradoxus individuals. We formally accounted for captures with unidentified sex using a Bayesian approach and conducted a simulation study to show that our approach was able to provide unbiased results even if the proportion of unsexed captures was high. Applying the method to the Black Redstart data, we found that the proportion of individuals with paradoxus plumage type increased from 7.6% in 1980 to 18.1% in 2013. Individuals with the paradoxus plumage type were on average 0.25 g heavier and had 0.62 mm longer third primaries than individuals with the cairei plumage type. However, we found no support for our expectation of later migration of paradoxus males compared to cairei individuals based on the assumption that paradoxus individuals should occupy autumn territories like adult males. Our results shed new light on the understudied timing of autumn migration in birds and are in line with available studies on Black Redstarts, suggesting a molt‐constraint that allows only young males in good body condition to molt into adult‐like plumages.  相似文献

Ecosystem services are natural functions of an ecosystem that can be, secondarily, used for the benefit of humans. A recent estimate showed that ecosystem services equal, on average, 33 trillion dollars a year, with pollination being responsible for 112 billions dollars. The alteration of natural systems and the loss of pollinating species have caused a decrease in many crops' productivity. The objective of this work is to evaluate the pollination as an ecological service in agriculture, testing the hypothesis that the presence of forest remnants increases coffee agricultural productivity through an increase in pollination. This argument is based on the assumption that areas of preservation of native forest required by Brazilian law provide pollinators to local agroecosystems. Fruit production was compared among three different planting regimes: agrosilviculture, and conventional monoculture with and without preserved forest remnants nearby. The average flower production by branch was different among the farms and was not related to the planting methods. The first flowering was larger than the second, representing 81–98% of the flowers' total production. The farms near forest fragments had an increase of 14.6% in production that can be related to the pollinating services.  相似文献

Posttranslational modifications (PTMs) of proteins determine their structure-function relationships, interaction partners, as well as their fate in the cell and are crucial for many cellular key processes. For instance chromatin structure and hence gene expression is epigenetically regulated by acetylation or methylation of lysine residues in histones, a phenomenon known as the ‘histone code’. Recently it was shown that these lysine residues can furthermore be malonylated, succinylated, butyrylated, propionylated and crotonylated, resulting in significant alteration of gene expression patterns. However the functional implications of these PTMs, which only differ marginally in their chemical structure, is not yet understood. Therefore generation of proteins containing these modified amino acids site specifically is an important tool. In the last decade methods for the translational incorporation of non-natural amino acids using orthogonal aminoacyl-tRNA synthetase (aaRS):tRNAaaCUA pairs were developed. A number of studies show that aaRS can be evolved to use non-natural amino acids and expand the genetic code. Nevertheless the wild type pyrrolysyl-tRNA synthetase (PylRS) from Methanosarcina mazei readily accepts a number of lysine derivatives as substrates. This enzyme can further be engineered by mutagenesis to utilize a range of non-natural amino acids. Here we present structural data on the wild type enzyme in complex with adenylated ε-N-alkynyl-, ε-N-butyryl-, ε-N-crotonyl- and ε-N-propionyl-lysine providing insights into the plasticity of the PylRS active site. This shows that given certain key features in the non-natural amino acid to be incorporated, directed evolution of this enzyme is not necessary for substrate tolerance.  相似文献

Sepsis is one of the highest causes of mortality in hospitalized people and a common complication in both surgical and clinical patients admitted to hospital for non-infectious reasons. Sepsis is especially common in older people and its incidence is likely to increase substantially as a population ages. Despite its increased prevalence and mortality in older people, immune responses in the elderly during septic shock appear similar to that in younger patients. The purpose of this study was to conduct a genome-wide gene expression analysis of circulating neutrophils from old and young septic patients to better understand how aged individuals respond to severe infectious insult. We detected several genes whose expression could be used to differentiate immune responses of the elderly from those of young people, including genes related to oxidative phosphorylation, mitochondrial dysfunction and TGF-β signaling, among others. Our results identify major molecular pathways that are particularly affected in the elderly during sepsis, which might have a pivotal role in worsening clinical outcomes compared with young people with sepsis.  相似文献

This study evaluated the feasibility of using the Ribulose Bisphosphate Carboxylase Large subunit gene (rbcL) and the Internal Transcribed Spacers 1 and 2 of the nuclear rDNA (nuITS1 and nuITS2) markers for identifying a very diverse, albeit poorly known group, of green microalgae from neotropical inland waters. Fifty-one freshwater green microalgae strains isolated from Brazil, the largest biodiversity reservoir in the neotropics, were submitted to DNA barcoding. Currently available universal primers for ITS1-5.8S-ITS2 region amplification were sufficient to successfully amplify and sequence 47 (92%) of the samples. On the other hand, new sets of primers had to be designed for rbcL, which allowed 96% of the samples to be sequenced. Thirty-five percent of the strains could be unambiguously identified to the species level based either on nuITS1 or nuITS2 sequences’ using barcode gap calculations. nuITS2 Compensatory Base Change (CBC) and ITS1-5.8S-ITS2 region phylogenetic analysis, together with morphological inspection, confirmed the identification accuracy. In contrast, only 6% of the strains could be assigned to the correct species based solely on rbcL sequences. In conclusion, the data presented here indicates that either nuITS1 or nuITS2 are useful markers for DNA barcoding of freshwater green microalgae, with advantage for nuITS2 due to the larger availability of analytical tools and reference barcodes deposited at databases for this marker.  相似文献
