Separation of autoinhibitor from cytokinin activity in spores of Dictyostelium discoideum 总被引:1,自引:1,他引:0
Spores of Dictyostelium discoideum NC-4H contain a spore germination autoinhibitor (SGI) that, on purification using Sephadex LH-20 eluted with 35% ethanol, can be separated from the bulk of cytokinin activity. Inhibitor studies and chromatographic analysis indicate that SGI is not a known cytokinin such as zeatin or IPA (N6 -(δ2 -isopentenyl)-adenine), N2 -dimethylguanosine or discadenine. Considering these findings and conflicting views in the literature concerning the structure of the autoinhibitor, the identity of SGI is discussed. 相似文献

TRAIL, tumor necrosis factor-related apoptosis-inducing ligand, is a member of the TNF family of proteins.Tumour cells were initially found to have increased sensitivity to TRAIL compared with normal cells, raising hopes that TRAIL would prove useful as an anti-tumor agent. The production of reliable monoclonal antibodies against TRAIL and its receptors that can stain fixed specimens will allow a thorough analysis of their expression on normal and malignant tissues. Here we report the generation of monoclonal antibodies against TRAIL and its four membrane-bound receptors(TR1-4), which have been used to stain a range of normal and malignant cells, as routinely fixed specimens. Low levels of TRAIL expression were found to be limited mostly to smooth muscle in lung and spleen as well as glial cells in the cerebellum and follicular cells in the thyroid. Expression of the TRAIL decoy receptors (TR3 and 4) was not as widespread as indicated by Northern blotting, suggesting that they may be less important for the control of TRAIL cytotoxicity than previously thought. TR1 and TR2 expression increases significantly in a number of malignant tissues,but in some common malignancies their expression was low, or patchy, which may limit the therapeutic role of TRAIL.Taken together, we have a panel of monoclonal antibodies that will allow a better assessment of the normal role of TRAIL and allow assessment of biopsy material, possibly allowing the identification of tumors that may be amenable to TRAIL therapy. 相似文献

The study of the regulation and cellular dynamics of receptor kinase signaling in plants is a rapidly evolving field that promises to give enormous insights into the molecular control of signal perception. In this study, we have analyzed the behavior of the L1-specific receptor kinase ARABIDOPSIS CRINKLY4 (ACR4) from Arabidopsis thaliana in planta and have shown it to be present in two distinct compartments within cells. These represent protein export bodies and a population of internalized vesicles. In parallel, deletion analysis has shown that a predicted beta-propeller-forming extracellular domain is necessary for ACR4 function. Nonfunctional ACR4 variants with deletions or point mutations in this domain behave differently to wild-type fusion protein in that they are not internalized to the same extent. In addition, in contrast with functional ACR4, which appears to be rapidly turned over, they are stabilized. Thus, for ACR4, internalization and turnover are linked and depend on functionality, suggesting that ACR4 signaling may be subject to damping down via internalization and degradation. The observed rapid turnover of ACR4 sets it apart from other recently studied plant receptor kinases. Finally, ACR4 kinase activity is not required for protein function, leading us to propose, by analogy to animal systems, that ACR4 may hetero-oligomerize with a kinase-active partner during signaling. Plant and animal receptor kinases have distinct evolutionary origins. However, with other recent work, our study suggests that there has been considerable convergent evolution between mechanisms used to regulate their activity. 相似文献

We investigated the effect of "psyching-up" on force production during the bench press. Twelve men (mean age +/- SD: 27.4 +/- 11.2 years) and 8 women (20.9 +/- 2.5 years) with strength-training experience performed 5 bench press repetitions on a modified Biodex isokinetic dynamometer during 3 interventions. The interventions were counterbalanced and included a free-choice psych-up, a cognitive distraction, and an attention-placebo. Peak force recorded after psyching-up (mean +/- SD: 764 +/- 269 N.m) was significantly different from both distraction (703 +/- 282 N.m, p = 0.003) and attention-placebo (708 +/- 248 N.m, p = 0.01). The mean percentage increase in peak force from distraction to psyching-up was 11.8% (6 to 18%, 95% confidence interval [CI]) and 8.1% from placebo to psyching-up (3 to 13%, 95% CI). The results of the present study indicate that psyching-up may increase force production during the bench press exercise in participants with at least 1 year strength-training experience. 相似文献

The X-linked inhibitor of apoptosis protein (XIAP) uses its second baculovirus IAP repeat domain (BIR2) to inhibit the apoptotic executioner caspase-3 and -7. Structural studies have demonstrated that it is not the BIR2 domain itself but a segment N-terminal to it that directly targets the activity of these caspases. These studies failed to demonstrate a role of the BIR2 domain in inhibition. We used site-directed mutagenesis of BIR2 and its linker to determine the mechanism of executioner caspase inhibition by XIAP. We show that the BIR2 domain contributes substantially to inhibition of executioner caspases. A surface groove on BIR2, which also binds to Smac/DIABLO, interacts with a neoepitope generated at the N-terminus of the caspase small subunit following activation. Therefore, BIR2 uses a two-site interaction mechanism to achieve high specificity and potency for inhibition. Moreover, for caspase-7, the precise location of the activating cleavage is critical for subsequent inhibition. Since apical caspases utilize this cleavage site differently, we predict that the origin of the death stimulus should dictate the efficiency of inhibition by XIAP. 相似文献

Oil spills often spell disaster for marine birds caught in slicks. However, the impact of oil pollution on seabird population parameters is poorly known because oil spills usually occur in wintering areas remote from breeding colonies where birds may be distributed over a wide area, and because it is difficult to separate the effects of oil pollution from the effect of natural environmental variation on seabird populations. Using a long-term data set we show that over-winter survival of adult common guillemots (Uria aalge) is negatively affected by both the incidence of four major oil-spills in their wintering grounds and high values of the North Atlantic Oscillation (NAO) index. After controlling for the effect of the NAO index, we show that winter mortality of adult guillemots is doubled by major oil pollution incidents. Our results demonstrate that oil pollution can have wide-scale impacts on marine ecosystems that can be quantified using populations of marked individuals to estimate survival. 相似文献

The availability of short stature apple scions that required minimal applications of chemical growth retardants and could be used with a range of rootstocks would be of considerable benefit to fruit growers. We have suppressed the expression of a gene encoding the gibberellin (GA) biosynthetic enzyme GA 20-oxidase to reduce the levels of bioactive GAs in a scion variety, resulting in significant reductions in stem height. Application of GA3 reversed the effect. The scion remained dwarfed after grafting on to normally invigorating rootstocks, whilst control plants of the same cultivar displayed the expected vigour when grafted on to these rootstocks. This approach could be applicable to any perennial crop variety, allowing dwarf trees to be obtained on any available rootstock or on their own roots without the need for chemical growth retardant application. In effect, seedlings that are well suited to local conditions (drought, salinity) could be employed as tree rootstocks, as could existing rootstocks valued for characters other than vigour control, such as pest and disease resistance. 相似文献

Recent studies, including the Cryptosporidium parvum Genome Project, have provided evidence for a mitochondrial-derived compartment in this parasite. This organelle appears to lack a genome, and thus must be entirely dependent on nuclear-encoded proteins. Here, we review the evidence for such an organelle in C. parvum and its probable function. There is no adequate treatment for infection by this parasite and so the elucidation of the role of this organelle and the effective targeting of its functions by antimicrobial agents might provide new treatments for infection by C. parvum. 相似文献
