A method for estimating the parameters of the single autosomal two-allele locus model with incomplete penetrance (the gene frequency and penetrances of the three genotypes) is presented. The method requires the presence of (1) the population frequency of the trait; (2) any set of incidences in relatives of probands that would allow to obtain the estimations of additive and dominant genetic variances and (3) segregation frequencies determined by the generalized Weinberg's method. The method proposed yields the uniquel estimations of the model parameters for traits which have additive genetic variance differed from zero. During continuous cultivation of Yarrowia lipolytica N 1, oxygen requirements for growth and citric acid synthesis were found to depend on the iron concentration in the medium. A coupled effect of oxygen and iron concentrations on the functioning of the mitochondrial electron transport chain in Y. lipolytica N 1 was established. Based on the results obtained in continuous culture, conditions for citric acid production in a batch culture of Y. lipolytica N 1 were proposed. At relatively low pO(2) value and a high iron concentration, citric acid accumulation was as high as 120 g l(-1); the specific rate of citric acid synthesis reached 120 mg citric acid (g cells h)(-1). The mass yield coefficient was 0.87 and the energy yield coefficient was 0.31.  相似文献

MOTIVATION: Gene annotation is the final goal of gene prediction algorithms. However, these algorithms frequently make mistakes and therefore the use of gene predictions for sequence annotation is hardly possible. As a result, biologists are forced to conduct time-consuming gene identification experiments by designing appropriate PCR primers to test cDNA libraries or applying RT-PCR, exon trapping/amplification, or other techniques. This process frequently amounts to 'guessing' PCR primers on top of unreliable gene predictions and frequently leads to wasting of experimental efforts. RESULTS: The present paper proposes a simple and reliable algorithm for experimental gene identification which bypasses the unreliable gene prediction step. Studies of the performance of the algorithm on a sample of human genes indicate that an experimental protocol based on the algorithm's predictions achieves an accurate gene identification with relatively few PCR primers. Predictions of PCR primers may be used for exon amplification in preliminary mutation analysis during an attempt to identify a gene responsible for a disease. We propose a simple approach to find a short region from a genomic sequence that with high probability overlaps with some exon of the gene. The algorithm is enhanced to find one or more segments that are probably contained in the translated region of the gene and can be used as PCR primers to select appropriate clones in cDNA libraries by selective amplification. The algorithm is further extended to locate a set of PCR primers that uniformly cover all translated regions and can be used for RT-PCR and further sequencing of (unknown) mRNA.   相似文献

Oligotripeptides Z-(Gly-Pro-Pro)n-OMe (n = 1,2,…,8), Z-Gly-Pro_Pro-Gly-Pro-Gly-OMe, Z-Gly-Pro-Pro-Gly-Pro-Gly-Gly-Pro-Pro-OMe, Z-Gly-Pro-Pro-(Gly-Pro-Gly)2-Gly-Pro-Pro-OMe, and Z-(Gly-Pro-Ala)n-OMe (n = 1,2,…,4) were synthesized step-by-step and then studied by means of x-ray diffraction, ir spectroscopy, the kinetics of hydrogen-deuterium exchange of peptide groups, and circular dichroism,. Different stages in the formation of a triple helix in Z-(Gly-Pro-Pro)n-OMe were revealed during the chain elongation. In the solid state, at the first stage a conformation of the polyproline II-type is formed in the tripeptide and in the second stage a triple helical complex appears in the hexapeptide. Interpeptide hydrogen bonds in this complex are still of low order. At further stages an ordered set of interpeptide hydrogen bonds is gradually formed. It is shown that the degree of order of interpeptide H bonds depends on the length of the molecular chain, the amino acid composition, and residue sequence in the triplets.  相似文献

The effect of succinate on the growth and respiration of the yeast Dipodascus magnusii VKM Y-1072, which is auxotrophic for thiamine and biotin, was studied. The addition of succinate to a culture grown on glucose was found to activate the respiration of cells on various substrates by enhancing the processes related to transamination reactions. In this case, aerobic fermentation (ethanol production) decreased, whereas pyruvate production increased. When succinate was added to the medium as the sole carbon source, it supported yeast growth in the absence of one of the two vitamins, thiamine or biotin, but not both. The yeast metabolism was completely respiratory, without any signs of aerobic fermentation. A drastic rise in pyruvate production in the yeast grown on glucose in the presence of succinate and the absence of biotin are also indicative of metabolic changes.  相似文献

The NAD+-dependent isocitrate dehydrogenase of the organic acid–producing yeast Yarrowia lipolytica was isolated, purified, and partially characterized. The purification procedure included four steps: ammonium sulfate precipitation, acid precipitation, hydrophobic chromatography, and gel-filtration chromatography. The enzyme was purified 129-fold with a yield of 31% and had a specific activity of 22 U/mg protein. The molecular mass of the enzyme was found to be 412 kDa. The enzyme consists of eight identical subunits with a molecular mass of about 52 kDa. The K m for NAD+ is 136 M, and that for isocitrate is 581 M. The effect of some intermediates of the citric acid cycle and nucleotides on the enzyme activity was studied. The role of isocitrate dehydrogenase (NAD+) in the overproduction of citric and keto acids is discussed.  相似文献

The comparative studies performed in this work showed that overproduction of alpha-ketoglutaric acid (KGA) from ethanol by the mutant Yarrowia lipolytica strain I requires both a deficiency of thiamine and a relatively high concentration of ammonium ions in the medium, whereas CA overproduction requires an almost zero concentration of ammonium ions. The threshold value of the dissolved oxygen concentration in the medium, pO2, for CA overproduction is considerably higher than for KGA overproduction. The respiration rate of CA-overproducing cells was 2-3.5 times higher than that of KGA-overproducing cells. The main terminal electron carrier functioning in the KGA-overproducing cells was cytochrome oxidase. In the CA-overproducing cells, the main terminal oxidase was presumably o-type cytochrome.  相似文献

The glyoxylate cycle is thought to be present in bacteria, protists, plants, fungi, and nematodes, but not in other Metazoa. However, activity of the glyoxylate cycle enzymes, malate synthase (MS) and isocitrate lyase (ICL), in animal tissues has been reported. In order to clarify the status of the MS and ICL genes in animals and get an insight into their evolution, we undertook a comparative-genomic study.  相似文献
