Glycolytic enzyme levels in synaptosomes   总被引:2,自引:0,他引:2  
The specific activities of glucosephosphate isomerase, aldolase, triosephosphate isomerase, glyceraldehydephosphate dehydrogenase, phosphoglycerate kinase, phosphoglycerate mutase, pyruvate kinase and lactate dehydrogenase were all higher in the synaptoplasmic fraction from rat brain than in 100,000 g supernatant fraction of rat brain homogenates when the supernatants were prepared in high ionic strength solutions. Four enzymes in synaptosomes and two enzymes in homogenates were associated with particulate fractions as indicated by the large increase in specific activity of the enzymes when samples were treated with 0.3 M KCl before centrifugation. Glucosephosphate isomerase, aldolase, pyruvate kinase and lactate dehydrogenase were the enzymes that showed a large increase in specific activity following salt treatment of isolated, synaptosomal membrane while aldolase and pyruvate kinase were the two enzymes which showed a large increase in specific activity in the high speed supernatant fractions. Because the specific activities of many enzymes are found to be elevated not only in synaptosomes but in synaptosomal membrane fractions it is suggested that these enzymes may provide the potential for significantly enhanced glycolysis at these locations.  相似文献

Hot-air temperature cycling of 1- to 10-microliters samples in glass capillary tubes can amplify DNA by the polymerase chain reaction in 15 min or less. A rapid temperature cycler of low thermal mass was constructed to change sample temperatures among denaturation, annealing, and elongation segments in a few seconds. After 30 cycles of 30 s each, a 536-bp beta-globin fragment of human genomic DNA was easily visualized with ethidium bromide on agarose gels. With rapid cycling, amplification yield depended on polymerase concentration. The time required for DNA amplification can be markedly reduced from prevailing protocols if appropriate equipment and sample containers are used for rapid heat transfer to the sample.  相似文献

The growth of two strains of Myxococcus virescens exhibiting dispersed growth was followed in casamino acids (N III-C) media and casitone media. The changes in optical density, pH, pigmentation as well as the secretion of bacteriolytic and proteolytic enzymes, DNA and polysaccharides during growth were recorded. In both media the bacteria grew exponentially with a generation time of 4 (casitone) and 20 hours (N III-C) respectively. The maximal cell mass was about 4 times higher in casitone than in casamino acids media. The amounts of bacteriolytic enzymes produced by the two strains in N III-C medium were different but in casitone medium they were about equal and considerably higher. The maximal values of proteolytic enzymes were about the same in both media and always occurred later than the bacteriolytic maxima. Both activity peaks appeared before the phase of decline. The polysaccharide production reached a maximum during the stationary growth phase in both media. A higher value was reached during growth in casitone medium than in N III-C medium. During the phase of decline a second increase of polysaccharide in the medium appeared. No DNA could be detected in the cell-free solutions until the beginning of the phase of decline.  相似文献

Seventy-eight heterocyclic compounds, which included derivatives of furan, isoxazole, pyridine, pyrrole, and thiophene, were tested in an in vitro system for their antimicrobial activity and their strain differentiating properties against three strains of Trypanosoma cruzi (Brazil, Corpus Christi, Tulahuen) and for their effect, in tissue culture, on African Green Monkey Kidney Cells—strain Vero. Forty-eight compounds were 75–100% effective in immobilizing trypomastigote forms of the three strains tested, and 29 of these were rated nondestructive to African Green Monkey Kidney (AGMK) cells. Of these 29 compounds, 12 gave variable results ranging from complete immobilization of one strain to no apparent effect on the motility of one or two of the other strains. Nine of the compounds rated harmless to AGMK fibroblasts are proposed as a basis for constructing molecules which might be active chemotherapeutic agents against various strains of T. cruzi.  相似文献

Undichna, the swimming trace of fish, was recovered from the middle member of the Mauch Chunk Formation in eastern Pennsylvania in strata of Late Mississippian (Visean) age. These traces represent the only evidence of fish known from the Mauch Chunk Formation and the Carboniferous of Eastern Pennsylvania. The Mauch Chunk Formation in the study area is characterized by an ephemeral fluvial depositional environment that yields a diverse invertebrate ichnoassemblage of the Scoyenia ichnofacies and a tetrapod footprint assemblage dominated by the tracks of temnospondyl amphibians. The Undichna specimens are preserved on fine-grained, mudstone-draped, rippled sandstone. Two specimens, assigned to U. Britannica, consist of pairs of well-defined, narrowly incised, sinusoidal wave traces that are out-of-phase. Two specimens, assigned to U. quina, consist of two pairs of in-phase sinusoidal waves intersected by an additional single wave with a greater amplitude. Two specimens, each composed of a single wave, are assigned to U. unisulca.

The slabs of rock with the Undichna traces also preserve tetrapod undertracks assigned to Batrachichnus, Matthewichnus, and Hylopus. This trace fossil association indicates the presence of small fish, some with caudal and/or anal fins and some with pectoral, pelvic, and anal and/or caudal fins, that touched the sediment during periods of intermittent flooding of subaerial surfaces walked upon by early tetrapods. Examples of Undichna from Mississippian or older deposits, especially from a fluvial paleoenvironment, are rare; few have been reported from the Devonian and the Lower Carboniferous.  相似文献
